TMPRSS2 Polymorphism and Its Correlation with SARS-CoV-2 Susceptibility - A Secondary Publication

Background:Severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)virus affected more people than SARS-CoV-1 and related *** genetic factors,besides respiratory droplets and direct exposure to the virus,are found highly responsible for transmitting SARS-CoV-2 ***:The objective of this study was to determine the plasma levels of the TMPRSS2 gene and its role in SARS-CoV-2 ***:A total of 100 patients,i.e.50 SARS-CoV-2 positive patients as the case group and 50 SARS-CoV-2 negative samples as the control group,were selected randomly and included in this case-control study to determine the association between TMPRSS2 gene and susceptibility to SARS-CoV-2 infection and severity of coronavirus disease 2019(COVID-19).The TMPRSS2 levels of case and control samples were measured through an enzyme-linked immunosorbent assay(ELISA).Following the genomic DNA extraction,a set of reverse and forward primers of human TMPRSS2 gene primers were used for the amplification of the TMPRSS2 ***:In the control group,the ratio of men to women was more or less the same while in the case group 62%of the population were *** TMPRSS2 level was found to be 4.70±7.7 ng/ml in case samples while it was 4.73±5.7 ng/ml in control ***:The levels of TMPRSS2 in plasma samples of both controls and cases were found to be the same indicating that the entry of SARS-CoV-2 is not dependent on the plasma levels of this protein.

BACKGROUND:Focal nodular hyperplasia,a benign hepatic tumor,is usually ***,rarely the entity can cause symptoms,mandating intervention. METHOD:We present a case of focal nodular hyperplasia of the liver,which caused a considerable diagnostic dilemma due to its atypical presentation. RESULTS:A 29-year-old woman presented with a 15-year history of a progressively increasing mass in the right upper quadrant which was associated with pain and emesis. Examination showed a firm,mobile mass palpable below the right subcostal margin.A computed tomography scan of the abdomen showed an exophytic mass arising from hepatic segments III and *** biopsy of the hepatic mass was *** showed a vascular tumor that was supplied by a tortuous branch of the proper hepatic artery. Surgical intervention for removal of the mass was undertaken. Intra-operatively,two large discrete tumors were found and completely *** examination showed features consistent with focal nodular hyperplasia. CONCLUSION:This description of an unusual case of focal nodular hyperplasia of the liver highlights the point that the diagnosis of otherwise benign hepatic tumors may be difficult despite extensive work-up in some cases.

Association of post-traumatic stress disorder and work performance: A survey from an emergency medical service, karachi, Pakistan

BACKGROUND: The purpose of the study was to explore the association between posttraumatic stress disorder(PTSD) and work performance of emergency medical services personnel in karachi, ***: Emergency medical service personnel were screened for potential PTSD using Impact of Event Scale-Revised(IES-R). Work performance was assessed on the basis of fi ve variables: number of late arrivals to work, number of days absent, number of days sick, adherence to protocol, and patient satisfaction over a period of 3 months. In order to model outcomes like the number of late arrivals to work, days absent and days late, negative binomial regression was applied, whereas logistic regression was applied for adherence to protocol and linear for patient satisfaction ***: Mean scores of PTSD were 24.0±12.2. No association was found between PTSD and work performance measures: number of late arrivals to work(RRadj 0.99; 0.98–1.00), days absent(RRadj 0.98; 0.96–0.99), days sick(RRadj 0.99; 0.98–1.00), adherence to protocol(ORadj 1.01; 0.99–1.04) and patient satisfaction(β 0.001%–0.03%) after adjusting for years of formal schooling, living status, coping mechanism, social support, working hours, years of experience and anxiety or ***: No statistically significant association was found between PTSD and work performance amongst EMS personnel in karachi, Pakistan.

A feeding trial was conducted for 75 d to evaluate the nutritive value of a mixture of animal by-products （MAB） as a possible protein source in diets for juvenile mangrove red snapper, Lutjanus argentimaculatus （mean initial body weight, 30 g）. Fish were fed one of five isonitrogenous diets （40% crude protein） replacing 0, 25% （MAB25）, 50% （MAB50）, 75% （MAB75） and 100% （MAB100） of fish meal protein with similar percentages of MAB. The MAB consisted of 25% cow liver meal, 20% leather meal, 20% meat and bone meal, 15% blood meal, 10% APC （poultry feather meal）, 8% poultry manure dried, 1.5% choline and 0.5% chromic oxide. After 75 d of feeding, fish fed with diets MAB50, MAB75 and MABI00 exhibited significantly lower growth performance than that of fish fed with control and MAB25 diets. The optimum level of MAB was estimated to be 23%. Replacement of fish meal by MAB23% showed the following performance： maximum weight gain, 510%; SGR, 2.39% and FCE, 2.83%. The MAB substitution up to 75% of fish meal protein in diets did not show differences in apparent protein digestibility （83.6% for MAB25, 79.2% for MAB50, 78.7% for MAB75） compared with control （83.4%）, whereas in MABI00 group digestibility （65.3%） was significantly lower than in other groups. The apparent phosphorus absorption of test diet groups was significantly higher （37.1% for MAB25, 28.5% for MABS0, 55.6% for MAB75 and 54.5% for MABI00） than that of control （1 1.2%）. The levels of protein and ash in the whole body, carcass and viscera increased as MAB substitution in diets increased, whereas lipids and moisture remained consistent among all treatment groups. These results showed that approximately 23% of fish meal protein could be replaced by a mixture of animal by-products for juvenile snapper growing from 30 g to 167 g in 75 d without compromising growth performance and feed efficiency.

Urease enzyme(UE)has been reported to be a potent virulence factor for Helicobacter pylori(HP)bacteria indicated to be responsible for various gastrointestinal ***,the spread of HP,currently regarded by the World Health Organization as a class 1 carcinogen,could be better controlled by targeting *** is in this line that we have synthesized three new derivatives(2–4)of the naturally occurring olean-12-en-3-one(1),which was previously isolated from the figs of Ficus vallis-choudae Delile(Moraceae).Among the synthesized compounds,3 and 4 contain an indole *** structures were unambiguously assigned by spectroscopic and spectrometric techniques(1D-NMR,2D-NMR and MS).The starting material and the synthesized compounds were screened for UE inhibition activity,and showed significant activities with IC50 values ranging from 14.5 to 24.6 lM,with compound(1)being the most potent as compared to the positive control thiourea(IC50=21.6 lM).Amongst the synthetic derivatives,compound 4 was the most potent(IC50-=17.9 lM),while the others showed activities close to that of the *** addition,molecular docking study of target compounds 2–4 was performed in an attempt to explore their binding mode for the design of more potent UE inhibitors.
