Severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)was reported at the end of 2019 as a worldwide health concern causing a pandemic of unusual viral pneumonia and many other organ damages,which was defined by the World Health Organization as coronavirus disease 2019(COVID-19).The pandemic is considered a significant threat to global public health till *** this review,we have summarized the lessons learnt during the emergence and spread of SARS-CoV-2,including its prototype and *** overall clinical features of variants of concern(VOC),heterogeneity in the clinical manifestations,radiology and pathology of COVID-19 patients are also discussed,along with advances in therapeutic agents.

SEMA7A belongs to the Semaphorin family and is involved in the oncogenesis and tumor *** glycosylation has been intricately linked with immune escape and tumor ***7A is a highly glycosylated protein with five glycosylated *** underlying mechanisms of SEMA7A glycosylation and its contribution to immunosuppression and tumorigenesis are ***,we identify overexpression and aberrant N-glycosylation of SEMA7A in head and neck squamous cell carcinoma,and elucidate fucosyltransferase FUT8 catalyzes aberrant core fucosylation in SEMA7A at N-linked oligosaccharides(Asn 105,157,258,330,and 602)via a direct protein‒protein interaction.A glycosylated statue of SEMA7A is necessary for its intra-cellular trafficking from the cytoplasm to the *** EGF triggers SEMA7A N-glycosylation through increasing the binding affinity of SEMA7A toward FUT8,whereas TGF-β1 promotes abnormal glycosylation of SEMA7A via induction of epithelial–mesenchymal *** N-glycosylation of SEMA7A leads to the differentiation of CD8^(+)T cells along a trajectory toward an exhausted state,thus shaping an immunosuppressive microenvironment and being resistant immunogenic cell *** of SEMA7A significantly improves the clinical outcome of EGFR-targeted and anti-PD-L1-based ***,we also define RBM4,a splice regulator,as a downstream effector of glycosylated SEMA7A and a pivotal mediator of PD-L1 alternative *** findings suggest that targeting FUT8-SEMA7A axis might be a promising strategy for improving antitumor responses in head and neck squamous cell carcinoma patients.

Objective:To assess the feasibility of single-port transperitoneal robotic-assisted laparoscopic radical prostatectomy(spRALP)and discuss its surgical ***:A 60-year-old male was admitted with an elevated prostate-specific antigen(PSA)level of 13.89 ng/mL and confirmed with prostate cancer on biopsy showing three of 22 positive cores with a Gleason score of 3þ4=*** magnetic resonance(MR)and bone scintigraphy showed organ-confined *** was performed using da Vinci Si HD surgical system,with access of a quadri-channel laparoscopic port placed *** drainage tubes were placed before wound *** surgical procedure was largely in consistence with a conventional robotic-assisted laparoscopic radical ***:The surgery was successfully carried out with a duration of 152 min and an estimated blood loss of 100 *** patient was discharged on postoperative Day 4 after removal of both pelvic drainage *** catheter was removed on postoperative Day *** major complications were *** pathology showed a Gleason score of 3þ4=7 with no extraprostatic extension and negative surgical ***:Single-port robotic prostatectomy is feasible using the currently available robotic instruments in most Chinese robotic urological *** preoperative planning and careful patient selection are *** studies concerning perioperative complications and pentafecta outcome compared with the conventional multi-port robotic prostatectomy is required.

Osteoarthritis(OA)is a common chronic inflammatory *** remodeling of inflammatory microenvironment in the joint is a promising strategy to prevent ***,current drugs remain unsatisfactory due to a lack of targeted and effective ways for relieving inflammatory conditions in OA ***(BTZ),a proteasome inhibitor,could effectively inhibit proinflammatory cytokines but with poor accumulation in the inflammatory *** overcome the shortcomings of BTZ delivery and to improve the efficacy of OA therapy,herein,we designed a novel nanomedicine(denoted as BTZ@PTK)by the co-assembly of BTZ and an amphiphilic copolymer(denoted as PTK)with ROS-cleaved thioketal(TK)*** TK units in BTZ@PTK are first cleaved by the excessive ROS at OA sites,and then triggered the controlled release of BTZ,resulting in the accurate delivery and the inflammatory microenvironment ***,BTZ@PTK suppressed ROS generation and proinflammatory cytokines while promoting M1 macrophage apoptosis in lipopolysaccharide(LPS)-activated RAW264.7 macrophages or LPS/IFN-γ-treated primary macrophages,which leads to a better effect than *** OA mice,BTZ@PTK passively accumulates into inflamed joints to attenuate pain sensitivity and gait ***,BTZ@PTK treatment successfully ameliorates synovitis with the reduction of synovial hyperplasia and synovitis scores by suppressing M1 macrophage polarization and promoting M1 macrophage apoptosis in the synovium,thereby delaying cartilage ***,BTZ@PTK can effectively modulate inflammatory microenvironment for OA recession by activating M1 macrophage apoptosis and inhibiting M1macrophage-mediated inflammatory response.

G-protein-coupled receptors(GPCRs)are the largest family of transmembrane receptors and regulate various physiological and pathological *** extensive studies,the roles of GPCRs in mouse embryonic stem cells(mESCs)remain poorly ***,we show that GPR160,a class A member of GPCRs,is dramatically downregulated concurrent with mESC differentiation into embryoid bodies in *** of Gpr160 leads to downregulation of the expression of pluripotency-associated transcription factors and upregulation of the expression of lineage markers,accompanying with the ar-rest of the mESC cell-cycle in the G0/G1 ***-seq analysis shows that GPR160 participates in the JAK/STAT signaling pathway crucial formaintaining ESC stemness,and the knockdown of Gpr160 results in the downregulation of STAT3 phosphorylation level,which in turn is partially rescued by colivelin,a STAT3 *** with these observations,GPR160 physically interacts with JAK1,and co-operates with leukemia inhibitory factor receptor(LIFR)and gp130 to activate the STAT3 *** summary,our results suggest that GPR160 regulates mESC self-renewal and pluripotency by interacting with the JAK1-LIFR-gp130 complex to mediate the JAK1/STAT3 signaling pathway.

Stem cell senescence is characterized by progressive functional dysfunction and secretory phenotypic changes including decreased proliferation,dysfunction of osteogenic and angiogenic differentiation,increased secretion of the senescence-associated secretory phenotype(SASP),which bring difficulties for bone *** or delaying senescence of aged bone marrow mesenchymal stem cells(O-BMSCs)was considered as effective strategy for bone regeneration in aging ***(Mg)ion released from bioceramics was reported to facilitate bone regeneration via enhancing osteogenesis and alleviating *** this study,Akermanite biocreamics(Akt)containing Mg ion as a model was demonstrated to promote osteogenesis and angiogenesis effects of O-BMSCs by activating the MAPK signaling pathway in ***,the enhanced osteogenesis effects might be attributed to enhanced Mg-containing Akt-mediated exosomal miR-196a-5p cargo targeting Hoxa7 and activation of MAPK signaling ***,the in vivo study confirmed that 3D-printed porous Mg-containing Akt scaffolds effectively increased bone regeneration in cranial defects of aged *** current results indicated that the exosomal-miR-196a-5p/Hoxa7/MAPK signaling axis might be the potential mechanism underlying Akt-mediated *** exosome-meditaed therapy stimulated by the released Mg ion contained in Akt biocreamics or other biomaterials might serve as a candidate strategy for bone repair in aged individuals.
