Salsolinol as an RNA m~6A methylation inducer mediates dopaminergic neuronal death by regulating YAP1 and autophagy

Salsolinol(1-methyl-6,7-dihydroxy-1,2,3,4-tetrahydroisoquinoline,Sal)is a catechol isoquinoline that causes neurotoxicity and shares structural similarity with 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine,an environmental toxin that causes Parkinson's ***,the mechanism by which Sal mediates dopaminergic neuronal death remains *** this study,we found that Sal significantly enhanced the global level of N~6-methyladenosine(m~6A)RNA methylation in PC12 cells,mainly by inducing the downregulation of the expression of m~6A demethylases fat mass and obesity-associated protein(FTO)and alk B homolog 5(ALKBH5).RNA sequencing analysis showed that Sal downregulated the Hippo signaling *** m~6A reader YTH domain-containing family protein 2(YTHDF2)promoted the degradation of m~6A-containing Yes-associated protein 1(YAP1)mRNA,which is a downstream key effector in the Hippo signaling ***,downregulation of YAP1 promoted autophagy,indicating that the mutual regulation between YAP1 and autophagy can lead to *** findings reveal the role of Sal on m~6A RNA methylation and suggest that Sal may act as an RNA methylation inducer mediating dopaminergic neuronal death through YAP1 and *** results provide greater insights into the neurotoxic effects of catechol isoquinolines compared with other studies and may be a reference for assessing the involvement of RNA methylation in the pathogenesis of Parkinson's disease.

An FFT-Based High SNR MWC Back-End Architecture with Analog Low-Pass Filter Compensation

We study the impacts of non-ideal filters in Modulated wideband converter(MWC). An analog filter compensating method is presented. The limitations of the conventional back-end of the advanced MWC is analyzed and an improved back-end based on Fast Fourier transform(FFT) is proposed. By setting proper length of the digital signal processed each time, the proposed system increases the recovery Signal-to-noise ratio(SNR)and significantly reduces the computation loads. The compensating operation is accurate to each frequency point and the expanding operation is simplified to a splitting of the frequency points. Numerical simulations demonstrate our proposed system against the ideal MWC with different filters. The results indicate that the recovery SNR of over 70 decibel is achieved with almost only one-tenth the computation complexity of the conventional MWC backend.

The ICM method with objective function transformed by variable discrete condition for continuum structure

ICM （Independent Continuous Mapping） method can solve topological optimization problems with the minimized weight as the objective and subjected to displacement constraints. To get a clearer topological configuration, by introducing the discrete condition of topological variables and integrating with the original objective, an optimal model with multi-objectives is formulated to make the topological variables approach 0 or 1 as near as possible, and the model reduces the effect of deleting rate on the result. The image-filtering method is employed to eliminate the checkerboard patterns and mesh dependence that occurred in the topology optimization of a continuum structure. The computational efficiency is enhanced through selecting quasi-active displacement constraints and a design region. Numerical examples indicate that this algorithm is robust and practicable, though the number of iterations is slightly increased with respect to the original algorithm.

Uniform Convergence of Wavelet Solution to the Sideways Heat Equation

We consider the problem uxx（x, t） = ut（x, t）, 0 ≤ x 〈 1, t ≥ 0, where the Cauchy data g（t） is given at x = 1. This is an ill-posed problem in the sense that a small disturbance on the boundary g（t） can produce a big alteration on its solution （if it exists）. We shall define a wavelet solution to obtain the well-posed approximating problem in the scaling space Vj. In the previous papers, the theoretical results concerning the error estimate are L2-norm and the solutions aren＇t stable at x = 0. However, in practice, the solution is usually required to be stable at the boundary. In this paper we shall give uniform convergence on interval x ∈ [0, 1].

Study on terahertz spectra of SO_2 and H_2S

The spectral characteristics of air pollution gas sulfur dioxide and hydrogen sulfide has been studied experimentally and theo-retically in the range of 0.2-2.6 THz. The gases absorption spectra of sulfur dioxide and hydrogen sulfide, as measured by terahertz Time-Domain Spectroscopy (THz-TDS) technique, show equi-spaced absorption peaks. The peak intervals are varied for different gas and may relate with the molecule structures and rotation modes. We have calculated the intervals of rotational transition frequency according to the gases molecule structure and the rotational modes. The results are consistent with experimental results which confirm the suggestion that the absorption is coming from the molecular rotational transition. The study suggests a technique to detect air pollutants by THz-TDS and the rotational modes of gas molecules.

Channel Mismatches Correction in Time-Interleaved ADCs Based on Hybrid Filter Bank Reconstruction

Time-interleaved analog-to-digital converter(TIADC) is an efficient way to achieve higher sampling rate for medium-to-high resolution applications. The performance of a TIADC suffers from the mismatch errors among the sub-channels. This paper presents a method to estimate the channel mismatches using the sub-channels output data. The proposed method introduces an equivalent transfer function for each channel to model and estimate the mismatch errors. A Hybrid filter bank(HFB)structure is used to both model the TIADC and reconstruct the desired uniformly sampled sequence based on the perfect reconstruction conditions of the HFB system.A four-channel 12-bit 400 MHz TIADC has been implemented in hardware to verify the proposed calibration method. The measured results show that the Spuriousfree dynamic range(SFDR) can be improved up to 74 d B after being corrected with 64-tap Finite-impulse response(FIR) filters.

Construction of a plasmid for human brain-derived neurotrophic factor and its effect on retinal pigment epithelial cell viability

Several studies have investigated the protective functions of brain-derived neurotrophic factor（BDNF） in retinitis pigmentosa. However, a BDNF-based therapy for retinitis pigmentosa is not yet available. To develop an efficient treatment for fundus disease, an eukaryotic expression plasmid was generated and used to transfect human 293 T cells to assess the expression and bioactivity of BDNF on acute retinal pigment epithelial-19（ARPE-19） cells, a human retinal epithelial cell line. After 96 hours of co-culture in a Transwell chamber, ARPE-19 cells exposed to BDNF secreted by 293 T cells were more viable than ARPE-19 cells not exposed to secreted BDNF. Western blot assay showed that Bax levels were downregulated and that Bcl-2 levels were upregulated in human ARPE-19 cells exposed to BDNF. Furthermore, 293 T cells transfected with the BDNF gene steadily secreted the protein. The powerful anti-apoptotic function of this BDNF may be useful for the treatment of retinitis pigmentosa and other retinal degenerative diseases.

Semantic composition of distributed representations for query subtopic mining

Inferring query intent is significant in information retrieval tasks. Query subtopic mining aims to find possible subtopics for a given query to represent potential intents. Subtopic mining is challenging due to the nature of short queries. Learning distributed representations or sequences of words has been developed recently and quickly, making great impacts on many fields. It is still not clear whether distributed representations are effective in alleviating the challenges of query subtopic mining. In this paper, we exploit and compare the main semantic composition of distributed representations for query subtopic mining. Specifically, we focus on two types of distributed representations: paragraph vector which represents word sequences with an arbitrary length directly, and word vector composition. We thoroughly investigate the impacts of semantic composition strategies and the types of data for learning distributed representations. Experiments were conducted on a public dataset offered by the National Institute of Informatics Testbeds and Community for Information Access Research. The empirical results show that distributed semantic representations can achieve outstanding performance for query subtopic mining, compared with traditional semantic representations. More insights are reported as well.
