AIM:To investigate the efficacy of aflibercept combined with sub-tenon injection of triamcinolone acetonide(TA)in treating diabetic macular edema(DME)and to examine changes in growth factors and inflammatory mediator levels in aqueous humor after ***:Totally 67 DME patients(67 eyes)and 30 cataract patients(32 eyes)were enrolled as the DME group and the control group,*** DME group was divided into the aflibercept group(34 cases)and the aflibercept combined with TA group(combined group,33 cases).The aqueous humor of both groups was collected during the study *** aqueous levels of vascular endothelial growth factor(VEGF),monocyte chemoattractant protein-1(MCP-1),interleukin-6(IL-6),interleukin-8(IL-8),and interleukin-1β(IL-1β)were detected using a microsphere suspension array technology(Luminex 200TM).Aqueous cytokines,best-corrected visual acuity(BCVA),central macular thickness(CMT),and complications before and after treatment were compared between the aflibercept group and combined ***:The concentrations of VEGF,MCP-1,IL-6,and IL-8 in the aqueous humor were significantly higher in the DME group than those of the control group(all Pgroup than those of the aflibercept group(all Pgroups were statistically different after 1 and 2mo of treatment(P0.05).CONCLUSION:The cytokines VEGF,MCP-1,IL-6,and IL-8 in the aqueous humor of DME patients are significantly *** and aflibercept combined with TA have good efficacy in DME patients,can effectively reduce CMT,improve the patient’s vision,and have high *** combined with TA can quickly downregulate the aqueous humor cytokines and help to relieve macular edema ***,the long-term eff

In vertebrates, most somatosensory pathways begin with the activation of dorsal root ganglion(DRG) neurons. The development of an appropriate DRG culture method is a prerequisite for establishing in vitro peripheral nerve disease models and for screening therapeutic drugs. In this study, we compared the changes in morphology, molecular biology, and transcriptomics of chicken embryo DRG cultured on tissue culture plates(T-DRG) versus three-dimensional collagen hydrogels(C-DRG). Our results showed that after 7 days of culture, the transcriptomics of T-DRG and C-DRG were quite different. The upregulated genes in C-DRG were mainly related to neurogenesis, axon guidance, and synaptic plasticity, whereas the downregulated genes in C-DRG were mainly related to cell proliferation and cell division. In addition, the genes related to cycles/pathways such as the synaptic vesicle cycle, cyclic adenosine monophosphate signaling pathway, and calcium signaling pathway were activated, while those related to cell-cycle pathways were downregulated. Furthermore, neurogenesis-and myelination-related genes were highly expressed in C-DRG, while epithelial–mesenchymal transition-, apoptosis-, and cell division-related genes were suppressed. Morphological results indicated that the numbers of branches, junctions, and end-point voxels per C-DRG were significantly greater than those per T-DRG. Furthermore, cells were scattered in T-DRG and more concentrated in C-DRG, with a higher ratio of 5-ethynyl-2′-deoxyuridine(EdU)-positive cells in T-DRG compared with C-DRG. C-DRG also had higher S100 calcium-binding protein B(S100 B) and lower α-smooth muscle actin(α-SMA) expression than T-DRG, and contained fewer terminal deoxynucleotidyl transferase dUTP nick end labeling(TUNEL)-positive cells after 48 hours of serum starvation. After cryopreservation, C-DRG maintained more intact morphological characteristics, and had higher viability and less TUNEL-positive cells than T-DRG. Furthermore, newly for

AIM:To evaluate the effect of symmetrical arc incision correcting corneal astigmatism in femtosecond laserassisted phacoemulsification(FLACS).METHODS:This study enrolled patients with cataract combined with regular corneal astigmatism of>0.75 D,who underwent *** arc incision was set at 8 mm diameter and 85%*** follow-up time was 3-24mo(4.92±3.49mo).Pentacam recorded the corneal astigmatism and higher-order aberration at pre-operation and *** changes in corneal astigmatism were analyzed by Alpins *** correlation of astigmatism type,age,corneal horizontal diameter,corneal thickness,arc incision length,and correction index(CI)was analyzed,and the residual corneal astigmatism was compared with the residual whole eye ***:Totally 79 patients(102 eyes)were enrolled,10 patients had corneal epithelial injury,1 patient occurred corneal epithelial *** corneal astigmatism was 1.23±0.38 D pre-operation,and decreased to 0.76±0.39 D post-operation(t=10.146,P=0.000).Corneal high-order aberration was 0.17±0.08μm pre-operation and 0.24±0.11μm post-operation(t=-5.186,P=0.000).The residual corneal astigmatism and residual whole eye astigmatism were no significant difference(t=-0.347,P=0.729).Using Alpin’s method,the following were determined:target-induced astigmatism(TIA)=1.23±0.38 D,surgeryinduced astigmatism(SIA)=0.77±0.45 D,difference vector(DV)=0.77±0.39 D,and CI=0.54±***,astigmatism size,corneal horizontal diameter,corneal thickness,and arc incision length were not correlated with *** CI for against the rule astigmatism(ATR)was better than that for with the rule astigmatism(WTR;P=0.001).CONCLUSION:Femtosecond laser-assisted astigmatic keratotomy has better CI of ATR,but increase higher-order corneal *** is not ideal,it’s not a perfect choice if we pursue ideal correction effect.

Objectives To evaluate degrees and percentages of cyclorotation in Chinese patients during femtosecond laser assisted cataract surgery performed by using LenSx femtosecond laser platform and to further sub-analyze bilateral cases to assess the congruency between both eyes. Methods This was a single-centre retrospective study(Beijing aier-Intec eye hospital, Beijing, China) in which the records of 330 eyes of 274 patients who underwent femtosecond laser-assisted cataract surgery in 1 or two eyes(with placement of posterior chamber IOL) between May 2017 and July 2019 by using femtosecond laser platform(LenSx) were *** Femtosecond Laser(Alcon Laboratories, Inc, Fort Worth, Texas, USA) was used to carry out corneal incisions, anterior capsulotomies and lens fragmentation in this procedure. All the patients were marked on corneal limbus(at 3 and 9 o’clock positions) by using sterile skin marker(***) while sitting upright preoperatively. Percentages of cyclorotation were calculated. Absolutes values were taken for all calculations. The cyclorotation data was tested to determine whether it followed a normal distribution. Descriptive statistics were applied to calculate percentile values of cyclorotation which is presented as(M [P25;P75]). Results This retrospective study evaluated 330 eyes of 274 Chinese patients who had femtosecond laser-assisted cataract surgery using the femtosecond laser platform. Of the 330 eyes, there were 90 eyes(27.27%) that did not show cyclorotation. Out of 330 eyes that were analyzed, 72.72%(240 eyes) exhibited cyclorotation. The median value of cyclorotation was 3.0[0.0;5.0](ranged between 0 to 19 degrees). Of the 330 eyes, there were 90 eyes(27.27%) that did not show cyclorotation. Overall anticlockwise rotation(172 eyes, 52.12%) was more common than clockwise rotation(68 eyes, 20.60%).The total numbers of right and left eyes were 169 and 161 respectively. Photoreceptor cell degeneration leads to blindness, for which there is currently no effective treatment. Our previous studies have shown that Lycium barbarum(L. barbarum) polysaccharide(LBP) protects degenerated photoreceptors in rd1, a transgenic mouse model of retinitis pigmentosa. L. barbarum glycopeptide(Lb GP) is an immunoreactive glycoprotein extracted from LBP. In this study, we investigated the potential protective effect of Lb GP on a chemically induced photoreceptor-degenerative mouse model. Wild-type mice received the following: oral administration of Lb GP as a protective pre-treatment on days 1–7;intraperitoneal administration of 40 mg/kg N-methylN-nitrosourea to induce photoreceptor injury on day 7;and continuation of orally administered Lb GP on days 8–14. Treatment with Lb GP increased photoreceptor survival and improved the structure of photoreceptors, retinal photoresponse, and visual behaviors of mice with photoreceptor degeneration. Lb GP was also found to partially inhibit the activation of microglia in N-methyl-N-nitrosourea-injured retinas and significantly decreased the expression of two pro-inflammatory cytokines. In conclusion, Lb GP effectively slowed the rate of photoreceptor degeneration in N-methyl-N-nitrosourea-injured mice, possibly through an anti-inflammatory mechanism, and has potential as a candidate drug for the clinical treatment of photoreceptor degeneration.

目的:评估飞秒激光FS200在不同模式下切割猫角膜基质板层界面的光滑度。方法:新鲜的猫眼球20只,随机平均分为4组(A、B、C、D组),分别在不同的模式下进行飞秒激光角膜基质板层切割:A组为常规切割组:负压吸引环吸引、角膜完全压平,压平直径约为13mm;B组不使用负压吸引环、角膜完全压平,压平直径约13mm;C组负压吸引环吸引、角膜压平直径约为8mm;D组不使用负压吸引环、角膜压平直径约为8mm。采用5分制的评分标准,对切割后的猫角膜基质界面的电子显微镜扫描照片进行评分,以评估切割后的角膜基质界面的光滑度。结果:裂隙灯显微镜图片显示出四组角膜基质界面光滑度的结果,D组最光滑,A组最粗糙。30倍扫描电镜照片的评分结果显示D组光滑度明显优于A组(P=0.007),D组光滑度明显优于B组(P=0.007),D组光滑度明显优于C组(P=0.016)。100倍电镜照片评分结果显示D组光滑度明显优于A组(P=0.01),D组光滑度明显优于B组(P=0.016)。其余各组间差异无统计学意义。结论:在无负压环吸引且压平面积小的模式-对角膜组织的压缩变形较小的切割状态下,飞秒激光辅助的猫角膜基质后板层切割界面的光滑度可得以部分改善。
