The role of plant tyrosyl-DNA phosphodiesteritdase 1a in genome stability is studied using a Medicago Etruncatula Mt Tdp1a-depleted line. Lack of Mt Tdp1 a results e in a 39% reduction of methylated cytosines as compared thto control. RNA-Seq analyses revealed that 11 DNA transto posons and 22 retrotransposons were differentially exrpressed in the Tdp1a-2a line. Among them all, DNA etttransposons（Mu DR, h AT, DNA3-11_Mad） and seven retroetransposons（LTR（Long Terminal Repeat）/Gipsy, LTR/Copia,LLTR and Non LTR/L1） were down-regulated, while the15 retrotransposons were up regulated. Results suggest that the occurrence of stress-responsive cis-elements as well as changes in the methylation pattern at the LTR promoters might be responsible for the enhanced retrotransposon transcription.

Alzheimer＇s disease is a multifactorial pathology, for which no cure is currently available. Nowadays, researchers are moving towards a new hypothesis of the onset of the illness, linking it to a metabolic impairment, q-his innovative approach will lead to the identification of new targets for the preparation of new effective drugs. Peroxisome proliferator-activated receptors and their ligands are the ideal candidates to reach the necessary breakthrough to defeat this complicate disease.

Nowadays, Alzheimer＇s disease（AD） is widely recognized as a real social problem. In fact, only five drugs are FDA approved for the therapy of this widespread neurodegenerative disease, but with low results so far. Three of them（rivastigmine, donepezil and galantamine） are acetylcholinesterase inhibitors, memantine is a N-methyl-D-aspartate receptor antagonist, whereas the fifth formulation is a combination of donepezil with memantine. The prevention and treatment of AD is the new challenge for pharmaceutical industry, as well as for public institutions, physicians, patients, and their families. The discovery of a new and safe way to cure this neurodegenerative disease is urgent and should not be delayed further. Because of the multiple origin of this pathology, a multi-target strategy is currently strongly pursued by researchers. In this review, we have discussed new structures designed to better the activity on the classical AD targets. We have also examined old and new potential drugs that could prove useful future for the therapy of the pathology by acting on innovative, not usual, and not yet fully explored targets like peroxisome proliferator-activated receptor（PPARs）.

Oxygen(O_(2))-sensing matrices are promising tools for the live monitoring of extracellular O_(2) consumption levels in long-term cell *** this study,ratiometric O_(2)-sensing membranes were prepared by electrospinning,an easy,low-cost,scalable,and robust method for fabricating ***(ε-caprolactone)and poly(dimethyl)siloxane polymers were blended with tris(4,7-diphenyl-1,10-phenanthroline)ruthenium(II)dichloride,which was used as the O_(2)-sensing probe,and rhodamine B isothiocyanate,which was used as the reference *** functionalized scaffolds were morphologically characterized by scanning electron microscopy,and their physicochemical profiles were obtained by Fourier transform infrared spectroscopy,thermogravimetric analysis,and water contact angle *** sensing capabilities were investigated by confocal laser scanning microscopy,performing photobleaching,reversibility,and calibration curve studies toward different dissolved O_(2)(DO)*** sensing nanofibers showed a high response to changes in DO concentrations in the physiological-pathological range from 0.5%to 20%and good stability under ratiometric *** addition,the sensing systems were highly biocompatible for cell growth promoting adhesiveness and growth of three cancer cell lines,namely metastatic melanoma cell line SK-MEL2,breast cancer cell line MCF-7,and pancreatic ductal adenocarcinoma cell line Panc-1,thus recreating a suitable biological environment in *** O_(2)-sensing biomaterials can potentially measure alterations in cell metabolism caused by changes in ambient O_(2)content during drug testing/validation and tissue regeneration processes.

Bulk-surface partial differential equations(BS-PDEs)are prevalent in manyapplications such as cellular,developmental and plant biology as well as in engineeringand material *** numerical methods for BS-PDEs in three space dimensions(3D)are *** this work,we present a bulk-surface virtual elementmethod(BS-VEM)for bulk-surface reaction-diffusion systems,a form of semilinearparabolic BS-PDEs in *** previous studies in two space dimensions(2D),the3D bulk is approximated with general polyhedra,whose outer faces constitute a flatpolygonal approximation of the *** this reason,the method is restricted tothe lowest order case where the geometric error is not *** BS-VEM guaranteesall the advantages of polyhedral methods such as easy mesh generation andfast matrix assembly on general *** advantages are much more relevantthan in *** allowing for general polyhedra,general nonlinear reaction kineticsand general surface curvature,the method only relies on nodal values without needingadditional evaluations usually associated with the quadrature of general *** latter is particularly costly in *** BS-VEM as implemented in thisstudy retains optimal convergence of second order in space.
