Charcot-Marie-Tooth-1A and sciatic nerve crush rat models:insights from proteomics

The sensorimotor and histological aspects of peripheral neuropathies were already studied by our team in two rat models:the sciatic nerve crush and the Charcot-Marie-Tooth-1A disease.In this study,we sought to highlight and compare the protein signature of these two pathological situations.Indeed,the identification of protein profiles in diseases can play an important role in the development of pharmacological targets.In fact,Charcot-Marie-Tooth-1A rats develop motor impairments that are more severe in the hind limbs.Therefore,for the first time,protein expression in sciatic nerve of Charcot-Marie-Tooth-1A rats was examined.First,distal sciatic nerves were collected from Charcot-Marie-Tooth-1A and uninjured wild-type rats aged 3 months.After protein extraction,sequential window acquisition of all theoretical fragment ion spectra liquid chromatography and mass spectrometry was employed.445 proteins mapped to Swiss-Prot or trEMBL Uniprot databases were identified and quantified.Of these,153 proteins showed statistically significant differences between Charcot-Marie-Tooth-1A and wild-type groups.The majority of these proteins were overexpressed in Charcot-Marie-Tooth-1A.Hierarchical clustering and functional enrichment using Gene Ontology were used to group these proteins based on their biological effects concerning Charcot-Marie-Tooth-1A pathophysiology.Second,proteomic characterization of wild-type rats subjected to sciatic nerve crush was performed sequential window acquisition of all theoretical fragment ion spectra liquid chromatography and mass spectrometry.One month after injury,distal sciatic nerves were collected and analyzed as described above.Out of 459 identified proteins,92 showed significant differences between sciatic nerve crush and the uninjured wild-type rats used in the first study.The results suggest that young adult Charcot-Marie-Tooth-1A rats(3 months old)develop compensatory mechanisms at the level of redox balance,protein folding,myelination,and axonogenesis.These mechanisms seem insufficient

Using emotions for the development of human-agent societies

Human-agent societies refer to applications where virtual agents and humans coexist and interact transparently into a fully integrated environment. One of the most important aspects in this kind of applications is including emotional states of the agents(humans or not) in the decision-making process. In this sense, this paper presents the applicability of the JaCalIVE(Jason Cartago implemented intelligent virtual environment) framework for developing this kind of society. Specifically, the paper presents an ambient intelligence application where humans are immersed into a system that extracts and analyzes the emotional state of a human group. A social emotional model is employed to try to maximize the welfare of those humans by playing the most appropriate music in every moment.

Modeling of shear walls using finite shear connector elements based on continuum plasticity

Light-frame timber buildings are often stabilized against lateral loads by using diaphragm action of roofs, floors and walls. The mechanical behavior of the sheathing-to-framing joints has a significant impact on the structural performance of shear walls. Most sheathing-to-framing joints show nonlinear load-displacement characteristics with plastic behavior. This paper is focused on the finite element modeling of shear walls. The purpose is to present a new shear connector element based on the theory of continuum plasticity. The incremental load-displacement relationship is derived based on the elastic-plastic stiffness tensor including the elastic stiffness tensor, the plastic modulus, a function representing the yield criterion and a hardening rule, and function representing the plastic potential. The plastic properties are determined from experimental results obtained from testing actual connections. Load-displacement curves for shear walls are calculated using the shear connector model and they are compared with experimental and other computational results. Also, the ultimate horizontal load-carrying capacity is compared to results obtained by an analytical plastic design method. Good agreements are found.

Quantifying entanglement of two-qubit Werner states

In this article,we introduce a framework for entanglement quantification of photon pairs represented by two-qubit Werner states.The measurement scheme is based on the symmetric informationally complete POVM.To make the framework realistic,we impose the Poisson noise on the measured two-photon coincidences.For various settings,numerical simulations were performed to evaluate the efficiency of the framework.

Research on bivalves is fast-growing,including genome-wide analyses and genome sequencing.Several characteristics qualify oysters as a valuable model to explore repetitive DNA sequences and their genome organization.Here we char-acterize the satellitomes of five species in the family Ostreidae(Crassostrea angulata,C.virginica,C.hongkongensis,C.ariakensis,Ostrea edulis),revealing a substantial number of satellite DNAs(satDNAs)per genome(ranging between 33 and 61)and peculiarities in the composition of their satellitomes.Numerous satDNAs were either associated to or derived from transposable elements,displaying a scarcity of transposable element-unrelated satDNAs in these genomes.Due to the non-conventional satellitome constitution and dominance of Helitron-associated satDNAs,comparative satellitomics demanded more in-depth analyses than standardly employed.Comparative analyses(including C.gigas,the first bivalve species with a defined satellitome)revealed that 13 satDNAs occur in all six oyster genomes,with Cg170/HindIII satDNA being the most abundant in all of them.Evaluating the“satDNA library model”highlighted the necessity to adjust this term when studying tandem repeat evolution in organisms with such satellitomes.When repetitive sequences with potential variation in the organizational form and repeat-type affiliation are examined across related species,the introduction of the terms“TE library”and“repetitive DNA library”becomes essential.

Peri-implant gas accumulation in response to magnesium-based musculoskeletal biomaterials:Reframing current evidence for preclinical research and clinical evaluation

Historically,the rapid degradation and massive gas release from magnesium(Mg)implants resulted in severe emphysema and mechanical failure.With the advent of new alloys and surface treatment methods,optimized Mg implants have re-entered clinics since last decade with reliable performance.However,the optimization aims at slowing down the degradation process,rather than exemption of the gas release.This study involved a systematic evaluation of current preclinical and clinical evidence,regarding the physical signs,symptoms,radiological features,pathological findings and complications potentially associated with peri±implant gas accumulation(PIGA)after musculoskeletal Mg implantation.The literature search identified 196 potentially relevant publications,and 51 papers were enrolled for further analysis,including 22 preclinical tests and 29 clinical studies published from 2005 to 2023.Various Mg-based materials have been evaluated in animal research,and the application of pure Mg and Mg alloys have been reported in clinical follow-ups involving multiple anatomical sites and musculoskeletal disorders.Soft tissue and intraosseous PIGA are common in both animal tests and clinical follow-ups,and potentially associated with certain adverse events.Radiological examinations especially micro-CT and clinical CT scans provide valuable information for quantitative and longitudinal analysis.While according to simulation tests involving Mg implantation and chemical processing,tissue fixation could lead to an increase in the volume of gas cavity,thus the results obtained from ex vivo imaging or histopathological evaluations should be interpreted with caution.There still lacks standardized procedures or consensus for both preclinical and clinical evaluation of PIGA.However,by providing focused insights into the topic,this evidence-based study will facilitate future animal tests and clinical evaluations,and support developing biocompatible Mg implants for the treatment of musculoskeletal disorders.

Cultivating Pleurotus ostreatus and Pleurotus eryngii mushroom strains onagro-industrial residues in solid-state fermentation. Part II: Effect onproductivity and quality of carposomes

The potential of selected Pleurotus ostreatus and P. eryngii wild-type and commercial strains to colonize andproduce carposomes after solid-state fermentation of five substrates constituted of agro-residues namely wheatstraw (WS), beech wood shavings (BWS), coffee residue (CR), barley and oats straw (BOS), rice bark (RB),supplemented with wheat bran was investigated. The effect of substrate composition on bioprocess feasibilitywas assessed for the different strains by quantitative (i.e., mushroom yield and Biological Efficiency-BE %) andcarposome qualitative parameters (i.e., weight, size, colour, firmness). P. ostreatus strains produced carposomesearlier than P. eryngii ones. Early fruiting formation was promoted in WS for P. ostreatus strains, whereas forP. eryngii the lowest values of earliness were observed on BOS substrate. As for crop productivity, P. ostreatusstrain AMRL 150 provided the highest BE (>70%) in all substrates except for RB, while P. ostreatus strain AMRL144 achieved the highest yield and BE in BOS and BWS (75.30 and 64.26%, respectively). P. eryngii stainsproduced less number but heavier carposomes. The highest values of BE% for these strains were recorded on WSand BOS substrates. The BE was further correlated to growth parameters of fungal vegetative phase. As formushroom colour, P. ostreatus were lighter than P. eryngii and BOS substrate promoted lightness in both strains.Firmer P. ostreatus mushrooms were produced at CR. No differences in the whiteness and firmness were detectedamong P. eryngii mushrooms and at any substrate used. The data included in this paper showed that Pleurotusmushrooms can be cultivated on low- or zero-value agro-industrial residues of great financial and environmentalimportance towards the production of value-added food products.

Numerical analysis of anisotropic stiffness and strength for geomaterials

In numerical modelling,selection of the constitutive model is a critical factor in predicting the actual response of a geomaterial.The use of oversimplified or inadequate models may not be sufficient to reproduce the actual geomaterial behaviour.That selection is especially relevant in the case of aniso-tropic rocks,and particularly for shales and slates,whose behaviour may be affected,e.***.well stability in geothermal or oil and gas production operations.In this paper,an alternative anisotropic constitutive model has been implemented in the finite element method software CODE_BRIGHT,which is able to account for the anisotropy of shales and slates in terms of both deformability and strength.For this purpose,a transversely isotropic version of the generalised Hooke’s law is adopted to represent the stiffness anisotropy,while a nonuniform scaling of the stress tensor is introduced in the plastic model to represent the strength anisotropy.Furthermore,a detailed approach has been proposed to determine the model parameters based on the stressestrain results of laboratory tests.Moreover,numerical analyses are performed to model uniaxial and triaxial tests on Vaca Muerta shale,Bossier shale and slate from the northwest of Spain(NW Spain slate).The experimental data have been recovered from the literature in the case of the shale and,in the case of the slate,performed by the authors in terms of stress-strain curves and strengths.A good agreement can be generally observed between numerical and experi-mental results,hence showing the potential applicability of the approach to actual case studies.Therefore,the presented constitutive model may be a promising approach for analysing the anisotropic behaviour of rocks and its impact on well stability or other relevant geomechanical problems in aniso-tropic rocks.

Slow light bimodal interferometry in onedimensional photonic crystal waveguides

Strongly influenced by the advances in the semiconductor industry,the miniaturization and integration of optical circuits into smaller devices has stimulated considerable research efforts in recent decades.Among other structures,integrated interferometers play a prominent role in the development of photonic devices for on-chip applications ranging from optical communication networks to point-of-care analysis instruments.However,it has been a longstanding challenge to design extremely short interferometer schemes,as long interaction lengths are typically required for a complete modulation transition.Several approaches,including novel materials or sophisticated configurations,have been proposed to overcome some of these size limitations but at the expense of increasing fabrication complexity and cost.Here,we demonstrate for the first time slow light bimodal interferometric behaviour in an integrated single-channel one-dimensional photonic crystal.The proposed structure supports two electromagnetic modes of the same polarization that exhibit a large group velocity difference.Specifically,an over 20-fold reduction in the higher-order-mode group velocity is experimentally shown on a straightforward all-dielectric bimodal structure,leading to a remarkable optical path reduction compared to other conventional interferometers.Moreover,we experimentally demonstrate the significant performance improvement provided by the proposed bimodal photonic crystal interferometer in the creation of an ultra-compact optical modulator and a highly sensitive photonic sensor.
