Characteristics of hepatic nuclear-transcription factor-kappa B expression and quantitative analysis in rat hepatocarcinogenesis

BACKGROUND: Hepatocellular carcinoma (HCC) is one of the most common malignant tumors. We analyzed the expression of miclear-transcription factor-kappa B (NF-kappa B) during hepatocarcinogenesis in order to evaluate its dynamic expression and its clinical value in the development and diagnosis of HCC. METHODS: Hepatoma models were induced by oral administration of 2-acetamidoflurene (2-FAA) to male Sprague-Dawley rats. Morphological changes were observed after hematoxylin and eosin staining. The cellular distribution of NF-kappa B expression during different stages of cancer development was investigated by immunohistochemistry, and the level of NF-kappa B expression in liver tissues was quantitatively analyzed by ELISA. The gene fragments of hepatic NF-kappa B were amplified by nested-polymerase chain reaction assay. RESULTS: Hepatocytes showed vacuole-like degeneration during the early stages, then had a hyperplastic nodal appearance during the middle stages, and finally progressed to tubercles of cancerous nests with high differentiation. The NF-kappa B-positive material was buff-colored, fine particles localized in the nucleus, and the incidence of NF-kappa B-positive cells was 81.8% in degeneration, 83.3% in precancerous lesions, and 100% in cancerous tissues. All of these values were higher than those in controls (P<0.01). Hepatic NF-kappa B expression and hepatic NF-kappa B-mRNA were also higher during the course of HCC development (P<0.01). CONCLUSION: The NF-kappa B signal transduction pathway is activated during the early stages of HCC development, and its abnormal expression may be associated with the occurrence of HCC.

A scheme for character restoration of Jiagu rubbings based on Poisson distribution and fractal geometry is proposed in this paper. The distribution characteristics of the connected regions on Jiagu rubbings is analyzed, and all the connected regions are recognized according to differences between these and their mathematical expectations. Regions are recognized as noise regions and filled if the differences are negative, while stroke regions are reserved. The attributes of the character image edges on Jiagu rubbings are also analyzed. The fractal dimensions of the character edge of Jiagu rubbings are calculated by means of statistics. The feature points are obtained using the cosine of the angle between the two vectors of the point and the consecutive two points. Different compression transformations with weighted coordinates are performed on different strokes and segments of the characters so as to smoothen the character edge of Jiagu rubbings images. Experimental results show that our scheme could successfully clear noise regions and polish the character edge of Jiagu rubbings.

Study on the Inhibitory Mechanism of Sophora japonica N-hexane Extract on Microcystis aeruginosa

[Objective] The research aimed to analyze the inhibitory mechanism of Sophora japonica n-hexane extract which significantly inhibited Microcystis aeruginosa in the prior research.[Method] *** n-hexane extract was used to treat *** inspecting chlorophyll a content,protein content,cell membrane permeability and superoxide dismutase（SOD） activity,the inhibitory mechanism of *** n-hexane extract on *** was analyzed initially.[Result] *** n-hexane extract destroyed the cell membrane system of ***,and increased the cell membrane *** contents of chlorophyll a and protein respectively declined to 10% and 50% of that in the control group after cultivated for 7 d,which indicated the photosynthetic reaction system of *** was *** addition,under the effect of *** n-hexane extract,SOD activity of *** increased in the early period and decreased in the latter period.[Conclusion] The possible inhibitory mechanism of *** n-hexane extract on *** was destroying the cell membrane to increase the membrane permeability;destroying the photosynthetic reaction system to decrease the contents of photosynthetic pigment and protein;making SOD activity showing the phased variation.
