Vibrio parahaemolyticus,a gram-negative halophilic bacterium that naturally inhabits coastal waters,causes gastroenteritis,skin infections,and septicemia in human beings[1].This bacterium produces multiple virulence factors,including thermostable direct hemolysin(TDH),TDH-related hemolysin(TRH),type Ⅲ secretion system 1(T3SS1),T3SS2,type Ⅵ secretion system 1(T6SS1),and T6SS2[1].Furthermore,Ⅴ.parahaemolyticus forms biofilms on the surface,which help it in adapting to unfavorable conditions[2].Mature biofilm formation requires special structures,including lateral and polar flagella,exopolysaccharide(EPS),and type Ⅳ pili[2].

BACKGROUND: Hepatocellular carcinoma (HCC) is one of the most common malignant tumors worldwide, its prognosis is poor, and early detection is of utmost importance. Although alpha-fetoprotein (AFP) is a useful marker for detecting and monitoring HCC development, the false-negative or false-positive rates with AFP alone may be as high as 30%-40% for patients with small HCCs. To enhance the specificity and accuracy of AFP measurements for HCC, we analyzed AFP expression states in livers, detected the hepatoma-specific AFP (HS-AFP) fraction and AFP-mRNA from peripheral blood mononuclear cells, and explored their clinical implications for HCC diagnosis. METHODS: AFP expression and hepatocyte distributions in liver specimens were investigated by an immunohistoche- mical assay. Total RNAs were extracted from circulating blood, synthesized to cDNA through random primers and reverse transcriptase, and fragments of the AFP gene were amplified by a nested-PCR assay. The HS-AFP fraction was separated by lectin-affinity chromatography and its level was detected by radioimmunoassay. RESULTS: The incidence of AFP was 73.3% in HCC tissues and its expression in HCCs with moderate or low differentiation was significantly stronger than that of HCCs with high differentiation (P<0.05). The incidence of AFP gene fragments was 100% in HCCs, and 60% in paracancerous tissues (P<0.01). In the HCC and liver cirrhosis groups, the incidence of HS-AFP was 91.7% and 18% (P<0.01), and of AFP-mRNA was 56.7% and 16% (P<0.01), respectively, and neither was found in ***-AFP or AFP-mRNA was not significantly related to size or number of HCC, but to its differentiation, metastasis, and relapse (P<0.05). CONCLUSIONS: Different AFP expression is present in different parts of HCC tissues. HS-AFP and AFP-mRNA fragments improve sensitivity and specificity, and both are useful markers to diagnose HCC or monitor metastasis and relapse.

Myelination of Schwann cells in the peripheral nervous system is an intricate process involving myelin protein trafficking. Recently, the role and mechanism of the endosomal/lysosomal system in myelin formation were emphasized. Our previous results demonstrated that a small GTPase Rab27a regulates lysosomal exocytosis and myelin protein trafficking in Schwann cells. In this present study, we established a dorsal root ganglion （DRG） neuron and Schwann cell co-culture model to identify the signals associated with Rab27a during myelination. First, Slp2-a, as the Rab27a effector, was endogenously expressed in Schwann cells. Second, Rab27a expression significantly increased during Schwann cell myelination. Finally, Rab27a and Slp2-a silencing in Schwann cells not only reduced myelin protein expression, but also impaired formation of myelin-like membranes in DRG neuron and Schwann cell co-cultures. Our findings suggest that the Rab27a/ Slp2-a complex affects Schwann cell myelination in vitro.

Cervical cancer(CESC)is one of the most common cancers and affects the female genital *** HPV infection status has been determined to be a vital cause of *** infection may induce changes to the immune system and limit the host’s immune *** is therefore essential to improving the overall survival of both locally advanced and recurrent CESC *** 304 relevant samples from TCGA,we assessed immune cell function in CESC patients to better understand the status of both tumor micro-environment cells and immune cells in *** enrichment analysis,pathway enrichment analysis,and PPI network construction were performed to explore the differentially expressed genes(DEGs).The analysis identified 425 DEGs,which included 295 up-regulated genes and 130 down-regulated *** established that upregulation of CCL5 was correlated with significantly better survival,meaning that CCL5 expression could serve as a novel prognostic biomarker for CESC *** further focused on CCL5 as a hub gene in CESC,as it had significant correlations with increased numbers of several types of immune ***-type fractions of M1 macrophages were significantly higher in the high-immune-scores group,which was associated with better overall ***,we concluded that CCL5 is a promising prognostic biomarker for CESC,as well as a novel chemotherapeutic target.
