The Chinese herb Ephedra(also known as Mahuang)has been extensively utilized for the prevention and treatment of coronavirus-induced diseases,including coronavirus disease 2019(COVID-19).However,the specific anti-SARS-CoV-2 compounds and mechanisms have not been fully *** main protease(M^(pro))of SARS-CoV-2 is a highly conserved enzyme responsible for proteolytic processing during the viral life cycle,making it a critical target for the development of antiviral *** study aimed to identify naturally occurring covalent inhibitors of SARS-CoV-2 M^(pro)from Ephedra and to investigate their covalent binding *** results demonstrated that the non-alkaloid fraction of Ephedra(ENA)exhibited a potent inhibitory effect against the SARS-CoV-2 M^(pro)effect,whereas the alkaloid fraction did ***,the chemical constituents in ENA were identified,and the major constituents'anti-SARS-CoV-2 M^(pro)effects were *** the tested constituents,herbacetin(HE)and gallic acid(GA)were found to inhibit SARS-CoV-2 M^(pro)in a time-and dose-dependent *** combination displayed a significant synergistic effect on this key ***,various techniques,including inhibition kinetic assays,chemoproteomic methods,and molecular dynamics simulations,were employed to further elucidate the synergistic anti-M^(pro)mechanisms of the combination of HE and ***,this study deciphers the naturally occurring covalent inhibitors of SARS-CoV-2 M^(pro)from Ephedra and characterizes their synergistic anti-M^(pro)synergistic effect,providing robust evidence to support the anti-coronavirus efficacy of Ephedra.

Diabetic nephropathy(DN)is a severe complication of diabetes,characterized by changes in kidney structure and *** natural product rosmarinic acid(RA)has demonstrated therapeutic effects,including anti-inflammation and anti-oxidative-stress,in renal damage or *** this study,we characterized the heterogeneity of the cellular response in kidneys to DN-induced injury and RA treatment at single cell *** results demonstrated that RA significantly alleviated renal tubular epithelial injury,particularly in the proximal tubular S1 segment and on glomerular epithelial cells known as podocytes,while attenuating the inflammatory response of macrophages,oxidative stress,and cytotox-icity of natural killer *** findings provide a comprehensive understanding of the mechanisms by which RA alleviates kidney damage,oxidative stress,and inflammation,offering valuable guidance for the clinical application of RA in the treatment of DN.

Mitochondrial dysfunction is a hallmark feature of cellular senescence and organ ***,we asked whether the mitochondrial antiviral signaling protein(MAVS),which is essential for driving antiviral response,also regulates human stem cell *** answer this question,we used CRISPR/Cas9-mediated gene editing and directed differentiation techniques to generate various MAVS-knockout human stem cell *** found that human mesenchymal stem cells(hMSCs)were sensitive to MAVS deficiency,as manifested by accelerated senescence *** uncovered that the role of MAVS in maintaining mitochondrial structural integrity and functional homeostasis depends on its interaction with the guanosine triphosphatase optic atrophy type 1(OPA1).Depletion of MAVS or OPA1 led to the dysfunction of mitochondria and cellular senescence,whereas replenishment of MAVS or OPA1 in MAVS-knockout hMSCs alleviated mitochondrial defects and premature senescence *** together,our data underscore an uncanonical role of MAVS in safeguarding mitochondrial homeostasis and antagonizing human stem cell senescence.

The effects of root oxidization ability and P fertilization on As mobility in soils, and subsequently As uptake, translocation and speciation in rice plants were investigated. Results show that root oxidation significantly influences As mobility in *** TD71 with higher radial oxygen loss(ROL) induces more Fe plaque formation and sequesters more As and P in iron plaque and rhizoshphere soil, leading to the reduction of As accumulation in rice plants. Additionally, P addition mobilizes As in soil solution, and increases As accumulation in rice plants. Arsenic speciation results show that the majority of As species in husks detected is inorganic As, accounting for 82%-93% of the total As, while in grains the majority of As is inorganic As and dimethyl arsenic(DMA), with DMA accounting for 33%-64% of the total As. The fraction of inorganic As decreases while fraction of DMA increases, with increasing As and P concentrations. The study further elucidates the mechanisms involved in effects of ROL on As tolerance and accumulation in rice.

Productivity and international energy price shocks are reflected in PPI and CPI via industrial ***’s in-depth participation into the global value chains has increasingly lengthened its industrial production *** question is how the changing length of production chains will affect CPI and PPI,as well as CPI-PPI correlation?By constructing a global input-output price model,this paper offers a theoretical discussion on the impact of production chain length on the CPI-PPI *** findings suggest that the price shock of international bulk commodities has a greater impact on china’s PPI than that on *** effects on both china’s PPI and CPI estimated by using the single-country input-output model are higher than the results estimated with the global input-output ***,the difference between CPI and PPI variations estimated with the global input-output model is greater than the result estimated with the single-country input-output model,which supports the view that the lengthening of production chains,especially international production chains,leads to a divergence between CPI and *** results based on cross-national panel data also suggest that the lengthening of production chains has reduced the CPI-PPI correlation for countries,*** lengthening of production chains has increased the PPI-CPI *** is to say,policymakers should target not just CPI in maintaining price stability,but instead focus on the stability of both PPI and *** can be made to proactively adjust the price index system,and formulate the industrial chain price index.

Epigenetic clocks are accurate predictors of human chronological age based on the analysis of DNA methylation(DNAm)at specific CpG ***,a systematic comparison between DNA methylation data and other omics datasets has not yet been ***,available DNAm age predictors are based on datasets with limited ethnic *** address these knowledge gaps,we generated and analyzed DNA methylation datasets from two independent Chinese cohorts,revealing age-related DNAm ***,a DNA methylation aging clock(iCAS-DNAmAge)and a group of DNAm-based multi-modal clocks for Chinese individuals were developed,with most of them demonstrating strong predictive capabilities for chronological *** clocks were further employed to predict factors influencing aging *** DNAm aging clock,derived from multi-modal aging features(compositeAge-DNAmAge),exhibited a close association with multi-omics changes,lifestyles,and disease status,underscoring its robust potential for precise biological age *** findings offer novel insights into the regulatory mechanism of age-related DNAm changes and extend the application of the DNAm clock for measuring biological age and aging pace,providing the basis for evaluating aging intervention strategies.

Hemihydrate calcium sulfate whiskers(HH-CSWs)were hydrothermally synthesized in a sulfuric acid solution at 120℃ for different holding times(20,40,and 60 min).The phase structures and morphologies were characterized by XRD and SEM,*** XRD pattern of the sample under 60 min was refined via the Rietveld fitting *** structure models of the HH-CSW sample under a 60-min holding time was established based on Rietveld fitting *** difference in the positions of diffraction peaks was *** as-prepared holding time increased the intensity and aspect ratio of the diffraction peaks of both *** the prepared HH-CSW structure,Ca-O polyhedron is a 12-sided polyhedron similar to that in the gypsum structure;the Ca atom is located in two positions in the one-unit cell;the H_(2)O channel along the c-axis similar is O-shaped and bigger than that in hexagonal and monoclinic CaSO_(4)·0.5H_(2)O ***,although the prepared HH-CSW crystals’structure are similar to that of the reported hexagonal and monoclinic CaSO_(4)·0.5H_(2)O structures,they are not the *** formation mechanism of HH-CSW from flue gas desulfurization(FGD)gypsum is discussed based on the analysis of gypsum structure.
