Cytokinins are mobile phytohormones that regulate plant growth,development,and environmental *** major cytokinin species include isopentenyl adenine(iP),trans-zeatin(tz),cis-zeatin(cz),and dihydrozeatin(Dz).The spatial distributions of different cytokinin species in different organelles,cells,tissues,and organs are primarily shaped by biosynthesis via isopentenyltransferases(IPT),cytochrome P450 monooxygenase,and 50-ribonucleotide phosphohydrolase and by conjugation or catabolism via glycosyltransferase or cytokinin oxidase/*** bind to histidine receptor kinases in the endoplasmic reticulum or plasma membrane and relay signals to response regulators in the nucleus via shuttle proteins known as histidine phosphotransfer *** movements of cytokinins from sites of biosynthesis to sites of signal perception usually require long-distance,intercellular,and intracellular *** the past decade,ATP-binding cassette(ABC)transporters,purine permeases(PUP),AzA-GUANINE RESISTANT(AzG)transporters,equilibrative nucleoside transporters(ENT),and Sugars Will Eventually Be Exported transporters(SWEET)have been characterized as involved in cytokinin transport *** review begins by introducing the spatial distributions of various cytokinins and the subcellular localizations of the proteins involved in their metabolism and *** focus on an inventory of the characterized transporters involved in cytokinin compartmentalization,including long-distance,intercellular,and intracellular transport,and the regulation of the spatial distributions of cytokinins by environmental *** directions for cytokinin research are also discussed.

The primary cilium,an important microtubule-based organelle,protrudes from nearly all the vertebrate *** motility of cilia is necessary for various developmental and physiological ***(PIs)and its metabolite,Ptd Ins(4,5)P2,have been revealed to contribute to cilia assembly and *** an important kinase of the PI pathway and signaling,phosphatidylinositol 4-kinaseβ(PI4 KB)is the one of the most extensively studied phosphatidylinositol 4-kinase ***,its potential roles in organ development remain to be *** investigate the developmental role of Pi4 kb,especially its function on zebrafish ciliogenesis,we generated pi4 kb deletion mutants using clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated protein 9 *** homozygous pi4 kb mutants exhibit an absence of primary cilia in the inner ear,neuromasts,and pronephric ducts accompanied by severe edema in the eyes and other ***,smaller otic vesicle,malformed semicircular canals,and the insensitivity on sound stimulation were characteristics of pi4 kb *** the protein level,both in vivo and in vitro analyses revealed that synthesis of Pi4 p was greatly reduced owing to the loss of Pi4 *** addition,the expression of the Pi4 kb-binding partner of neuronal calcium sensor-1,as well as the phosphorylation of phosphatidylinositol-4-phosphate downstream effecter of Akt,was significantly inhibited in pi4 kb *** together,our work uncovers a novel role of Pi4 kb in zebrafish inner ear development and the functional formation of hearing ability by determining hair cell ciliogenesis.

RNA interference(RNAi)has emerged as a powerful technology for pest ***,we have shown that plastid-mediated RNAi(PM-RNAi)can be utilized to control the Colorado potato beetle,an insect pest in the Chrysomelidae family;however,whether this technology is suitable for controlling pests in the Coccinellidae remained *** coccinellid 28-spotted potato ladybird(Henosepilachna vigintioctopunctata;HV)is a serious pest of solanaceous *** this study,we identified three efficient target genes(β-Actin,SRP54,and SNAP)for RNAi using in vitro double-stranded RNAs(dsRNAs)fed to HV,and found that dsRNAs targetingβ-Actin messenger RNA(dsACT)induced more potent RNAi than those targeting the other two *** next generated transplastomic and nuclear transgenic potato(Solanum tuberosum)plants expressing HV *** dsACT stably accumulated to up to 0.7%of the total cellular RNA in the transplastomic plants,at least three orders of magnitude higher than in the nuclear transgenic ***,the transplastomic plants also exhibited a significantly stronger resistance to HV,killing all larvae within 6 *** data demonstrate the potential of PM-RNAi as an efficient pest control measure for HV,extending the application range of this technology to Coccinellidae pests.

Global patterns of taxonomic and phylogenetic diversity of flowering plants: Biodiversity hotspots and coldspots

Species diversity of angiosperms(flowering plants) varies greatly among *** patterns of variation in species diversity are shaped by the interplay of ecological and evolutionary ***,using a comprehensive data set for regional angiosperm floras across the world,we show geographic patterns of taxonomic(species) diversity,phylogenetic diversity,phylogenetic dispersion,and phylogenetic deviation(i.e.,phylogenetic diversity after accounting for taxonomic diversity) across the *** diversity is strongly and positively correlated with taxonomic diversity;as a result,geographic patterns of taxonomic and phylogenetic diversity across the world are highly *** with high taxonomic and phylogenetic diversity are located in tropical regions whereas areas with low taxonomic and phylogenetic diversity are located in temperate regions,particularly in Eurasia and North America,and in northern ***,phylogenetic dispersion is,in general,higher in tropical regions and lower in temperate ***,the geographic pattern of phylogenetic deviation differs substantially from those of taxonomic and phylogenetic diversity and phylogenetic *** a result,hotspots and coldspots of angiosperm diversity identified based on taxonomic and phylogenetic diversity and phylogenetic dispersion are incongruent with those identified based on phylogenetic *** of these metrics may be considered when selecting areas to be protected for their biodiversity.

Integrin-interacting protein Kindlin-2 induces mammary tumors in transgenic mice

Kindlin-2, an integrin-interacting protein, regulates breast cancer progression. However, currently, no animal model to study the role of Kindlin-2 in the carcinogenesis of mammary gland is available. We established a Kindlin-2 transgenic mouse model using a mammary gland-specific promoter, mammary tumor virus(MMTV) long terminal repeat(LTR). Kindlin-2 was overexpressed in the epithelial cells of the transgenic mice. The mammary gland ductal trees were found to grow faster in MMTV-Kindlin-2 transgenic mice than in control mice during puberty. Kindlin-2 promoted mammary gland growth as indicated by more numerous duct branches and larger lumens, and more alveoli were formed in the mammary glands during pregnancy under Kindlin-2 overexpression. Importantly, mammary gland-specific expression of Kindlin-2 induced tumor formation at the age of 55 weeks on average. Additionally, the levels of estrogen receptor and progesterone receptor were decreased, whereas human epidermal growth factor receptor 2 and β-catenin were upregulated in the Kindlin-2-induced mammary tumors. These findings demonstrated that Kindlin-2 induces mammary tumor formation via activation of the Wnt signaling pathway.
