Establishment and application of a surrogate model for human Ebola virus disease in BSL-2 laboratory

The Ebola virus(EBOV)is a member of the Orthoebolavirus genus,Filoviridae family,which causes severe hemorrhagic diseases in humans and non-human primates(NHPs),with a case fatality rate of up to 90%.The development of countermeasures against EBOV has been hindered by the lack of ideal animal models,as EBOV requires handling in biosafety level(BSL)-4 ***,accessible and convenient animal models are urgently needed to promote prophylactic and therapeutic approaches against *** this study,a recombinant vesicular stomatitis virus expressing Ebola virus glycoprotein(VSV-EBOV/GP)was constructed and applied as a surrogate virus,establishing a lethal infection in *** infection with VSV-EBOV/GP,3-week-old female Syrian hamsters exhibited disease signs such as weight loss,multi-organ failure,severe uveitis,high viral loads,and developed severe systemic diseases similar to those observed in human EBOV *** animals succumbed at 2–3 days post-infection(dpi).Histopathological changes indicated that VSV-EBOV/GP targeted liver cells,suggesting that the tissue tropism of VSV-EBOV/GP was comparable to wild-type EBOV(WT EBOV).Notably,the pathogenicity of the VSV-EBOV/GP was found to be species-specific,age-related,gender-associated,and challenge ***,equine anti-EBOV immunoglobulins and a subunit vaccine were validated using this ***,this surrogate model represents a safe,effective,and economical tool for rapid preclinical evaluation of medical countermeasures against EBOV under BSL-2 conditions,which would accelerate technological advances and breakthroughs in confronting Ebola virus disease.

Cavity-enhanced single quantum dots(QDs)are the main approach towards ultra-high-performance solid-state quantum light sources for scalable photonic quantum ***,harnessing the Purcell effect requires precise spectral and spatial alignment of the QDs’emission with the cavity mode,which is challenging for most *** we have successfully integrated miniaturized Fabry-Perot microcavities with a piezoelectric actuator,and demonstrated a bright single-photon source derived from a deterministically coupled QD within this *** the cavity-membrane structures,we have achieved large spectral tunability via strain *** resonance,a high Purcell factor of~9 is *** source delivers single photons with simultaneous high extraction efficiency of 0.58,high purity of 0.956(2)and high indistinguishability of 0.922(4).Together with its compact footprint,our scheme facilitates the scalable integration of indistinguishable quantum light sources on-chip,therefore removing a major barrier to the development of solid-state quantum information platforms based on QDs.

Currently,the enhancement in electromagnetic interference(EMI)performance of polymeric composite generally relies on either improving electrical conductivity(σ)for stronger electromagnetic(EM)reflections or tailoring structure for higher EM ***,we proposed a novel technique called cyclic pulsating pressure enhanced segregating structuration(CPP-SS),which can reinforce these two factors *** structural information was supplied by optical microscopy(OM)and scanning electron microscopy(SEM),both of which confirmed the formation and evolution of segregate structured ultra-high molecular weight polyethylene(UHMWPE)/graphene ***,the result showed that CPP-SS can significantly improve theσof ***,advanced specific EMI shielding efficiency of 31.1 d B/mm was achieved for UHMWPE/graphene composite at 1-mm thickness and a low graphene loading of 5 wt%.Meanwhile,it also confirmed that the intrinsic disadvantage of poor mechanical properties of conventional segregated structure composites can be *** work is believed to provide a fundamental understanding of the structural and performance evolutions of segregated structured composites prepared under CPPSS,and to bring us a simple and efficient approach for fabricating high-performance,strong and light-weight polymeric EMI shields.

Efficient translation mediated by the 5'untranslated region(5'UTR)is essential for the robust efficacy of mRNA ***,the N1-methyl-pseudouridine(m1)modification of mRNA can impact the translation efficiency of the 5'*** discovered that the optimal 5'UTR for m1y-modified mRNA(m1y-5'UTR)differs significantly from its unmodified counterpart,high-lighting the need for a specialized tool for designing mly-5'UTRs rather than directly utilizing high-expression endogenous gene 5'*** response,we developed a novel machine learning-based tool,Smart5UTR,which employs a deep generative model to identify superior m1y-5'UTRs in *** tailored loss function and network architecture enable Smart5UTR to overcome limitations inherent in existing *** a result,Smart5UTR can successfully design superior 5'UTRs,greatly benefiting mRNA vaccine ***,Smart5UTR-designed superior 5'UTRs significantly enhanced antibody titers induced by COVID-19 mRNA vaccines against the Delta and Omicron variants of SARS-CoV-2,surpassing the performance of vaccines using high-expression endogenous gene 5'UTRs.

Magnetic domain structures in ultrathin Bi_(2)Te_(3)/CrTe_(2) heterostructures

Chromium tellurium compounds are important two-dimensional van der Waals ferromagnetic materials with high Curie temperature and chemical stability in air,which is promising for applications in spintronic ***,highquality spin-orbital-torque(SOT)device,Bi_(2)Te_(3)/CrTe_(2)heterostructure was epitaxially grown on Al_(2)O_(3)(0001)*** Hall measurements indicate the existence of strong ferromagnetism in this device with the CrTe_(2)thickness down to 10 *** order to investigate its micromagnetic structure,cryogenic magnetic force microscope(MFM)was utilized to measure the magnetic domain evolutions at various temperatures and magnetic *** virgin domain state of the device shows a worm-like magnetic domain structure with the size around 0.6μm-0.8μ*** irregular-shape magnetic domains(>1μm)can be induced and pinned,after the field is increased to coercive field and ramped back to low *** temperature-dependent MFM signals exhibit a nice mean-field-like ferromagnetic transition with Curie temperature around 201.5 K,indicating a robust ferromagnetic *** a device can be potentially implemented in future magnetic memory technology.

High-throughput calculations combining machine learning to investigate the corrosion properties of binary Mg alloys

Magnesium(Mg)alloys have shown great prospects as both structural and biomedical materials,while poor corrosion resistance limits their further *** this work,to avoid the time-consuming and laborious experiment trial,a high-throughput computational strategy based on first-principles calculations is designed for screening corrosion-resistant binary Mg alloy with intermetallics,from both the thermodynamic and kinetic *** stable binary Mg intermetallics with low equilibrium potential difference with respect to the Mg matrix are firstly ***,the hydrogen adsorption energies on the surfaces of these Mg intermetallics are calculated,and the corrosion exchange current density is further calculated by a hydrogen evolution reaction(HER)kinetic *** intermetallics,e.g.Y_(3)Mg,Y_(2)Mg and La_(5)Mg,are identified to be promising intermetallics which might effectively hinder the cathodic ***,machine learning(ML)models are developed to predict Mg intermetallics with proper hydrogen adsorption energy employing work function(W_(f))and weighted first ionization energy(WFIE).The generalization of the ML models is tested on five new binary Mg intermetallics with the average root mean square error(RMSE)of 0.11 *** study not only predicts some promising binary Mg intermetallics which may suppress the galvanic corrosion,but also provides a high-throughput screening strategy and ML models for the design of corrosion-resistant alloy,which can be extended to ternary Mg alloys or other alloy systems.

Glucocorticoids-based prodrug design: Current strategies and research progress

Attributing to their broad pharmacological effects encompassing anti-inflammation,antitoxin, and immunosuppression, glucocorticoids(GCs) are extensively utilized in the clinic for the treatment of diverse diseases such as lupus erythematosus, nephritis,arthritis, ulcerative colitis, asthma, keratitis, macular edema, and leukemia. However, longterm use often causes undesirable side effects, including metabolic disorders-induced Cushing's syndrome(buffalo back, full moon face, hyperglycemia, etc.), osteoporosis,aggravated infection, psychosis, glaucoma, and cataract. These notorious side effects seriously compromise patients' quality of life, especially in patients with chronic ***, glucocorticoid-based advanced drug delivery systems for reducing adverse effects have received extensive attention. Among them, prodrugs have the advantages of low investment, low risk, and high success rate, making them a promising *** this review, we propose the strategies for the design and summarize current research progress of glucocorticoid-based prodrugs in recent decades, including polymer-based prodrugs, dendrimer-based prodrugs, antibody-drug conjugates, peptide-drug conjugates,carbohydrate-based prodrugs, aliphatic acid-based prodrugs and so on. Besides, we also raise issues that need to be focused on during the development of glucocorticoid-based prodrugs. This review is expected to be helpful for the research and development of novel GCs and prodrugs.

Seismic Risk Model for the Beijing–Tianjin–Hebei Region, China:Considering Epistemic Uncertainty from the Seismic Hazard Models

This study presents a probabilistic seismic risk model for the Beijing–Tianjin–Hebei region in China. The model comprises a township-level residential building exposure model, a vulnerability model derived from the Chinese building taxonomy,and a regional probabilistic seismic hazard model. The three components are integrated by a stochastic event-based method of the OpenQuake engine to assess the regional seismic risk in terms of average annual loss and exceedance probability curve at the city, province, and regional levels. The novelty and uniqueness of this study are that a probabilistic seismic risk model for the Beijing–Tianjin–Hebei region in China is developed by considering the impact of site conditions and epistemic uncertainty from the seismic hazard model.

Developing cost-effective,robust and stable non-precious metal catalysts for oxygen reduction reaction(ORR) is of paramount importance for electrochemical energy conversion devices such as fuel cells and metal-air *** Fe-N-C single atom catalysts(SACs) have been hailed as the most promising candidate due to the optimal binding strength of ORR intermediates on the Fe-N_(4) sites,they suffer from serious mass transport limitations as microporous templates/substrates,i.e.,zeolitic imidazolate frameworks(ZIFs),are usually employed to host the active *** by this challenge,we herein develop a hydrogen-bonded organic framework(HOF)-assisted pyrolysis strategy to construct hierarchical micro/mesoporous carbon nanoplates for the deposition of atomically dispersed Fe-N_(4) *** a design is accomplished by employing HOF nanoplates assembled from 2-aminoterephthalic acid(NH_(2)-BDC) and p-phenylenediamine(PDA) as both soft templates and C,N *** from the structural merits inherited from HOF templates,the optimized catalyst(denoted as Fe-N-C SAC-950) displays outstanding ORR activity with a high half-wave potential of 0.895 V(*** hydrogen electrode(RHE)) and a small overpotential of 356 mV at 10 mA cm^(-2) for the oxygen evolution reaction(OER).More excitingly,its application potential is further verified by delivering superb rechargeability and cycling stability with a nearly unfading charge-discharge gap of 0.72 V after 160 *** dynamics(MD) simulations reveal that micro/mesoporous structure is conducive to the rapid mass transfer of O_(2),thus enhancing the ORR *** situ Raman results further indicate that the conversion of O_(2) to~*O_(2)-the rate-determining step(RDS) for Fe-N-C *** work will provide a versatile strategy to construct single atom catalysts with desirable catalytic properties.

This paper studies the optimization problem of heterogeneous networks under a timevarying *** agent only accesses to one local objective function,which is *** improved algorithm with noisy measurement of local objective functions' sub-gradients and additive noises among information exchanging between each pair of agents is designed to minimize the sum of objective functions of all *** weaken the effect of these noises,two step sizes are introduced in the control *** graph theory,stochastic analysis and martingale convergence theory,it is proved that if the sub-gradients are uniformly bounded,the sequence of digraphs is balanced and the union graph of all digraphs is joint strongly connected,then the designed control protocol can force all agents to find the global optimal point almost *** last,the authors give some numerical examples to verify the effectiveness of the stochastic sub-gradient algorithms.
