Background:Human epidermal growth factor receptor 2(HER2)overexpression is related to anti-HER2 therapy in many ***48-antibody-drug conjugate(ADC)has shown promising efficacy in patients with HER2-positive locally advanced or metastatic urothelial carcinoma(UC).The characteristic expression and scoring systems of HER2 are nonexistent in *** aimed to explore HER2 status and its correlation with the efficacy of HER2-targeting ADC therapy in ***:A total of 137 and 43 patients were enrolled in cohort 1 and cohort 2,respectively,from March 2009 to December *** patients in cohort 2 were enrolled in a phase II study of *** samples were tested for HER2 status using immunohistochemistry(IHC)and/or fluorescence in situ hybridization(FISH).The 2018 ASCO/CAP HER2 scoring system was adopted and modified to score HER2 expression in ***:The HER2-positive(IHC 2+or 3+)rate was 24.1%(33/137).In HER2 IHC 2+or 3+patients,the HER2 gene amplification rate was 31%(13/42).The objective response rates(ORRs)in RC48-ADC-treated patients with IHC 3+,IHC 2+and FISH+,IHC 2+and FISH-were 58.8%,66.7%and 40%,*** ORR showed a trend toward a better benefit for RC48-ADC therapy in patients with HER2 amplification than in those without amplification(61.5%vs.44.8%,P=0.059).The heterogeneity of HER2 expression in the primary tumor was 55.5%(15/27),and the ORR was not significantly different between patients with tumor heterogeneity and ***:IHC testing should be performed to assess the HER2 status before the initiation of HER2-ADC *** was a trend toward a better benefit for patients with HER2 amplification,and tumor heterogeneity did not influence the drug efficacy.

Achieving high‐precision extraction of sea islands from high‐resolution satellite remote sensing images is crucial for effective resource development and sustainable ***,achieving such accuracy for sea island extraction presents significant challenges due to the presence of extensive background interference.A more widely applicable noise‐tolerant matched filter(NTMF)scheme is proposed for sea island extraction based on the MF *** NTMF scheme effectively suppresses the background interference,leading to more accurate and robust sea island *** further enhance the accuracy and robustness of the NTMF scheme,a neural dynamics algorithm is supplemented that adds an error integration feedback term to counter noise interference during internal computer operations in practical *** comparative experiments were conducted on various remote sensing images of sea islands under different noisy working conditions to demonstrate the superiority of the proposed neural dynamics algorithm‐assisted NTMF *** experiments confirm the ad-vantages of using the NTMF scheme for sea island extraction with the assistance of neural dynamics algorithm.

Based on a comprehensive review of domestic and foreign literature, this article discusses the technical difficulties and development status of enhanced geothermal system(EGS) concerning the thermal energy extraction of deep hot dry rock(HDR) reservoirs and proposes suggestions for the research focus of numerical simulation of HDR reservoir stimulation. Additionally, it summarizes the existing methods and mainstream working fluids for HDR reservoir stimulation. The article emphasizes the significance of factors such as well location, production well depth, artificial fracture orientation, and complexity in optimizing the thermal production efficiency of the EGS. Furthermore, this article delves into a detailed discussion on the influence of fracture spacing, fracture permeability,fracture length, fluid injection rate, and injected fluid temperature on the performance of the EGS. In light of the thermo-hydro-mechanical coupling challenges associated with high-temperature reservoirs, it is suggested that future research efforts should focus on investigating the impact of thermo-induced stresses on the stability of the artificial fracture network within the EGS during long-term(>30 years) circulation of hot and cold fluids.

Background:The current standard of care for non-bulky diffuse large B-cell lymphoma(DLBCL)patients with an International Prognostic Index(IPI)of 0 is four cycles of rituximab plus cyclophosphamide,doxorubicin,vincristine and prednisone(R-CHOP)but whether the same efficacy can be achieved with reduced chemotherapy regimen of four cycles for non-bulky DLBCL patients with an IPI of 1 remains *** study compared four cycles versus six cycles of chemotherapy in non-bulky low-risk DLBCL patients with negative interim positron emission tomography with computed tomography(PET-CT,Deauville 1-3),irrespective of age and other IPI risk factors(IPI 0-1).Methods:This was an open-label,randomized,phaseⅢ,non-inferiority *** aged 14-75 years with newly diagnosed low-risk DLBCL,according to IPI,achieving PET-CT confirmed complete response(CR)after four cycles of R-CHOPwere randomized(1:1)between four cycles of rituximab(4R-CHOP+4R arm)or two cycles of R-CHOP plus two cycles of rituximab(6R-CHOP+2R arm).The primary endpoint was 2-year progression-free survival(PFS),conducted in the intention-to-treat *** was assessed in patients with at least one cycle of assigned *** non-inferiority margin was-8%.Results:A total of 287 patients were included in the intention-to-treat analysis,the median follow-up was 47.3 months,and the 2-year PFS rate was 95%(95%confidence interval[CI],92%to 99%)and 94%(95%CI,91%to 98%)for the 4R-CHOP+4R and 6R-CHOP+2R *** absolute difference in 2-year PFS between the two arms was 1%(95%CI,-5%to 7%),supporting the non-inferiority of 4R-CHOP+*** 3-4 neutropenia was lower in the last four cycles of rituximab alone in the 4R-CHOP+4R arm(16.7%versus 76.9%),with decreased risk of febrile neutropenia(0.0%versus 8.4%)and infection(2.1%versus 14.0%).Conclusions:For newly diagnosed low-risk DLBCL patients,interim PET-CT after four cycles of R-CHOP was effective in identifying patients with Deauville 1-3 who would have a goo

Chronic obstructive pulmonary disease(COPD)is a common chronic respiratory disease and the third leading cause of death *** in next-generation sequencing technology have improved microbiome analysis,which is increasingly recognized as an important component of disease *** to the gut,the lung is a biosphere containing billions of microbial *** lung microbiome plays an important role in regulating and maintaining the host immune *** microbiome composition,metabolites of microorganisms,and the interactions between the lung microbiome and the host immunity profoundly affect the occurrence,development,treatment,and prognosis of *** this review,we drew comparisons between the lung microbiome of healthy individuals and that of patients with ***,we summarize the intrinsic interactions between the host and the overall lung microbiome,focusing on the underlying mechanisms linking the microbiome to the host innate and adaptive immune response ***,we discuss the possibility of using the microbiome as a biomarker to determine the stage and prognosis of COPD and the feasibility of developing a novel,safe,and effective therapeutic target.
