Computer-assisted sperm morphometry fluorescence-based analysis has potential to determine progeny sex

This study was designed to determine the ability of computer-assisted sperm morphometry analysis （CASA-Morph） with fluorescence to discriminate between spermatozoa carrying different sex chromosomes from the nuclear morphometrics generated and different statistical procedures in the bovine species. The study was divided into two experiments. The first was to study the morphometric differences between X- and Y-chromosome-bearing spermatozoa （SX and SY, respectively）. Spermatozoa from eight bulls were processed to assess simultaneously the sex chromosome by FISH and sperm morphometry by fluorescence-based CASA-Morph. SX cells were larger than SY cells on average （P 〈 0.001） although with important differences between bulls. A simultaneous evaluation of all the measured features by discriminant analysis revealed that nuclear area and average fluorescence intensity were the variables selected by stepwise discriminant function analysis as the best discriminators between SX and SY. In the second experiment, the sperm nuclear morphometric results from CASA-Morph in nonsexed （mixed SX and SY） and sexed （SX） semen samples from four bulls were compared. FISH allowed a successful classification of spermatozoa according to their sex chromosome content. X-sexed spermatozoa displayed a larger size and fluorescence intensity than nonsexed spermatozoa （P 〈 0.05）. We conclude that the CASA-Morph fluorescence-based method has the potential to find differences between X- and Y-chromosome-bearing spermatozoa in bovine species although more studies are needed to increase the precision of sex determination by this technique.

A comparative study of the morphometry of sperm head components in cattle, sheep, and pigs with a computer-assisted fluorescence method

The aim of this study was to compare the sperm nuclear and acrosomal morphometry of three species of domestic artiodactyls; cattle （Bos taurus）, sheep （Ovis aries）, and pigs （Sus scrofa）. Semen smears of twenty ejaculates from each species were fixed and labeled with a propidium iodide-Pisum sativum agglutinin （PI/PSA） combination. Digital images of the sperm nucleus, acrosome, and whole sperm head were captured and analyzed. The use of the PI/PSA combination and CASA-Morph fluorescence-based method allowed the capture, morphometric analysis, and differentiation of most sperm nuclei, acrosomes and whole heads, and the assessment of acrosomal integrity with a high precision in the three species studied. For the size of the head and nuclear area, the relationship between the three species may be summarized as bull 〉 ram 〉 boar. However, for the other morphometric parameters （length, width, and perimeter）, there were differences in the relationships between species for sperm nuclei and whole sperm heads. Bull sperm acrosomes were clearly smaller than those in the other species studied and covered a smaller proportion of the sperm head. The acrosomal morphology, small in the bull, large and broad in the sheep, and large, long, and with a pronounced equatorial segment curve in the boar, was species-characteristic. It was concluded that there are clear variations in the size and shape of the sperm head components between the three species studied, the acrosome being the structure showing the most variability, allowing a clear distinction of the spermatozoa of each species.

Li2SnO3 branched nano- and microstructures with intense and broadband white-light emission

Exploiting the synergy between microstructure, morphology and dimensions by suitable nanomaterial engineeri ng, can effectively upgrade the physical properties and material performances. Li2SnO3 elongated nano- and microstructures in form of belts, wires, rods and branched structures have been fabricated by a vapor-solid method at temperatures ranging from 700 to 900℃ using metallic Sn and Li2CO3 as precursors. The achievement of these new morphologies can face challenging applications for Li2SnO3, not only in the field of energy storage, but also as building blocks in optoelectronic devices. The micro? and nanostructures grown at 700 and 800℃ correspond to monoclinic Li2SnO3, while at 900℃ complex Li2S nO/SnO2 core-shell microstructures are grow n, as con firmed by X-ray diffractio n and Rama n spectroscopy. Transmission electron microscopy reveals structural disorder related to stacking faults in some of the branched structures, which is associated with the prese nee of the low-temperature phase of L12S nO3. The luminescent resp onse of these structures is domi nated by in tense emissi ons at 2, 2.5 and 3 eV, almost completely covering the whole range of the visible light spectrum. As a result, white-light emission is obtained without the need of phosphors or complex quantum well heterostructures. Enhanced functionality in applications such as in light-emitting devices could be exploited based on the high luminescenee intensity observed in some of the analysed Li2SnO3 structures.

Repression of the Auxin Response Pathway Increases Arabidopsis Susceptibility to Necrotrophic Fungi

In plants, resistance to necrotrophic pathogens depends on the interplay between different hormone systems, such as those regulated by salicylic acid （SA）, jasmonic acid （JA）, ethylene, and abscisic acid. Repression of auxin signaling by the SA pathway was recently shown to contribute to antibacterial resistance. Here, we demonstrate that Arabidopsis auxin signaling mutants axrl, axr2, and axr6 that have defects in the auxin-stimulated SCF （Skpl-Cullin- F-box） ubiquitination pathway exhibit increased susceptibility to the necrotrophic fungi Plectosphaerella cucumerina and Botrytis cinerea. Also, stabilization of the auxin transcriptional repressor AXR3 that is normally targeted for removal by the SCF-ubiquitin/proteasome machinery occurs upon P. cucumerina infection. Pharmacological inhibition of auxin transport or proteasome function each compromise necrotroph resistance of wild-type plants to a similar extent as in non-treated auxin response mutants. These results suggest that auxin signaling is important for resistance to the necrotro- phic fungi P. cucumerina and B. cinerea. SGTlb （one of two Arabidopsis SGT1 genes encoding HSP90/HSC70 co-chaperones） promotes the functions of SCF E3-ubiquitin ligase complexes in auxin and JA responses and resistance conditioned by certain Resistance （R） genes to biotrophic pathogens. We find that sgtlb mutants are as resistant to P. cucumerina as wild-type plants. Conversely, auxin/SCF signaling mutants are uncompromised in RPP4-triggered resistance to the obligate biotrophic oomycete, Hyaloperonospora parasitica. Thus, the predominant action of SGTlb in R gene-conditioned resis- tance to oomycetes appears to be at a site other than assisting SCF E3-ubiquitin ligases. However, genetic additivity of sgtlb axrl double mutants in susceptibility to H. parasitica suggests that SCF-mediated ubiquitination contributes to lim- iting biotrophic pathogen colonization once plant-pathogen compatibility is established.

Bohm’s approach to quantum mechanics: Alternative theory or practical picture?

Since its inception Bohmian mechanics has been generally regarded as a hidden-variable theory aimed at providing an objective description of quantum phenomena. To date, this rather narrow conception of Bohm’s proposal has caused it more rejection than acceptance. Now, after 65 years of Bohmian mechanics, should still be such an interpretational aspect the prevailing appraisal? Why not favoring a more pragmatic view, as a legitimate picture of quantum mechanics, on equal footing in all respects with any other more conventional quantum picture? These questions are used here to introduce a discussion on an alternative way to deal with Bohmian mechanics at present, enhancing its aspect as an efficient and useful picture or formulation to tackle, explore, describe and explain quantum phenomena where phase and correlation (entanglement) are key elements. This discussion is presented through two complementary blocks. The first block is aimed at briefly revisiting the historical context that gave rise to the appearance of Bohmian mechanics, and how this approach or analogous ones have been used in different physical contexts. This discussion is used to emphasize a more pragmatic view to the detriment of the more conventional hidden-variable (ontological) approach that has been a leitmotif within the quantum foundations. The second block focuses on some particular formal aspects of Bohmian mechanics supporting the view presented here, with special emphasis on the physical meaning of the local phase field and the associated velocity field encoded within the wave function. As an illustration, a simple model of Young’s two-slit experiment is considered. The simplicity of this model allows to understand in an easy manner how the information conveyed by the Bohmian formulation relates to other more conventional concepts in quantum mechanics. This sort of pedagogical application is also aimed at showing the potential interest to introduce Bohmian mechanics in undergraduate quantum

Dear Editor Abscisic acid （ABA） is the main phytohormone involved in adaptive crop responses to drought. ABA signaling relies on the family of pyrabactin resistance 1/PYRl-like/regulatory components of ABA receptors （PYR/PYL/RCAR）, which upon ABA binding form high-affinity ternary complexes with clade A protein phosphatases type 2C （PP2Cs） and inhibit them （Cutler et al., 2010）. This results in the activation of an SnRK2-dependent phosphorylation cascade affecting a high number of targets in the plant cells, regulating stomatal aper- ture and the expression of stress-responsive genes （Cutler et al., 2010）.

Most redox-regulated chloroplast enzymes are reduced during the day and oxidized during the *** the reduction mechanism of light-dependent enzymes is well known,the mechanism mediating their oxidation in the dark remains *** thiol-dependent peroxidases,2-Cys peroxiredoxins (Prxs),play a key role in light-dependent reduction of chloroplast *** transfer reducing equivalents of thiols to hydrogen peroxide,suggesting the participation of these peroxidases in enzyme oxidation in the ***,we have addressed this issue by analyzing the redox state of well-known redox-regulated chloroplast enzymes in response to darkness in Arabidopsis thaliana mutants deficient in chloroplastlocalized Prxs (2-Cys Prxs A and B,Prx ⅡE,and Prx Q).Mutant plants lacking 2-Cys Prxs A and B,and plants overexpressing NADPH-dependent thioredoxin (Trx) reductase C showed delayed oxidation of chloroplast enzymes in the *** contrast,the deficiencies of Prx ⅡE or Prx Q exerted no *** vitro assays allowed the reconstitution of the pathway of reducing equivalents from reduced fructose 1,6-bisphosphatase to hydrogen peroxide mediated by Trxs and 2-Cys *** together,these results suggest that 2-Cys Prxs participate in the short-term oxidation of chloroplast enzymes in the dark.

Farm-scale evaluation of the impact of CrylAb Bt maize on canopy nontarget arthropods： a 3-year study

The cultivation of CrylAb-expressing genetically modified MON810 （Bt maize） has led to public concern in Europe, regarding its impact on nontarget arthro- pods （NTAs）. We have assessed the potential effects of DKC 6451 YG （MON810） maize on canopy NTAs in a farm-scale study performed in Central Spain during 3 years. The study focused on hemipteran herbivores （leafhoppers and planthoppers） and hymenopteran parasitic wasps （mymarids） collected by yellow sticky traps, which accounted for 72% of the total number of insects studied. The dynamics and abundance of these groups varied among years, but no significant differences were found between Bt and non-Bt maize, indicating that Bt maize had no negative effect on these taxa. Nonetheless, the CrylAb toxin was detected in 2 different arthropods collected from Bt maize foliage, the cicadellids Zyginidia scutellaris and Empoasca spp. A retrospective power analysis on the arthropod abundance data for our field trials has determined that Z. scutellaris and the family My- maridae have high capacity to detect differences between the Bt maize and its isogenic counterpart. The use of these canopy NTAs as surrogates for assessing environmental impacts of Bt maize is discussed.

Influence of metal support in-plane symmetry on the corrugation of hexagonal boron nitride and graphene monolayers

Predicting the properties of two-dimensional （2D） materials as graphene and hexagonal boron nitride （h-BN） monolayers after their growth on any given substrate is a major challenge. While the influence of the electron configuration of the atoms of the underlying surface is well-understood, the effect of substrate geometry still remains unclear. The structural properties of h-BN monolayers grown on a rectangularly packed Rh（110） surface were characterized in situ by ultrahigh vacuum scanning tunneling microscopy and were compared to those that this material exhibits when grown on substrates showing different crystallographic orientations. Although the h-BN monolayer grown on Rh（110） was dominated by a unique quasiunidimensional moir6 pattern, suggesting considerable interface interaction, the moir6 corrugation was unexpectedly smaller than those reported for strongly interacting interfaces with hexagonal-terminated substrates, owing to differences in the possible binding landscapes at interfaces with differently oriented substrates. Moreover, a rule was derived for predicting how interface corrugation and the existence and extent of subregions within moir6 supercells containing favorable sites for orbital mixing between h-BN monolayers and their supports depend on substrate symmetry. These general symmetry considerations can be applied to numerous 2D materials, including graphene, thereby enabling the prediction of how substrate choice determines the properties of these materials. Furthermore, they could also provide new routes for tuning 2D material properties and for developing nanotemplates showing different geometries for Krowing adsorbate superlattices.
