Suppression of PFKFB3-driven glycolysis restrains endothelial-to-mesenchymal transition and fibrotic response

Endothelial-to-mesenchymal transition(EndoMT),the process wherein endothelial cells lose endothelial identity and adopt mesenchymal-like phenotypes,constitutes a critical contributor to cardiac *** phenotypic plasticity of endothelial cells can be intricately shaped by alteration of metabolic pathways,but how endothelial cells adjust cellular metabolism to drive EndoMT is incompletely ***,we identified 6-phosphofructo-2-kinase/fructose-2,6-biphosphatase 3(PFKFB3)as a critical driver of EndoMT via triggering abnormal glycolysis and compromising mitochondrial *** suppression of PFKFB3 with salvianolic acid C(SAC),a phenolic compound derived from Salvia miltiorrhiza,attenuates EndoMT and fibrotic ***3-haplodeficiency recapitulates the anti-EndoMT effect of SAC while PFKFB3-overexpression augments the magnitude of EndoMT and exacerbates cardiac ***,PFKFB3-driven glycolysis compromises cytoplasmic nicotinamide adenine dinucleotide phosphate(reduced form,NAdPH)production via hijacking glucose flux from pentose phosphate *** of mitochondrial NAdPH through isocitrate/α-ketoglutarate shuttle replenishes cytoplasmic NAdPH pool but meanwhile impairs mitochondrial respiration by hampering mitochondrial iron-sulfur cluster *** disrupts PFKFB3 stability by accelerating its degradation and thus maintains metabolic homeostasis in endothelial cells,underlying its anti-EndoMT *** findings for the first time identify the critical role of PFKFB3 in triggering EndoMT by driving abnormal glycolysis in endothelial cells,and also highlight the therapeutic potential for pharmacological intervention of PFKFB3(with SAC or other PFKFB3 inhibitors)to combat EndoMT-associated fibrotic responses via metabolic regulation.

Fiber laser and gas metal arc hybrid welding of 440-MPa-grade high-strength marine steel was carried out at different welding *** influence of welding speed on the micro structure and mechanical properties of weld metal was *** weld-metal microstructure mainly consisted of pre-eutectoid ferrite,side-plate ferrite,acicular ferrite and lath bainite at a low welding *** the increase in welding speed,acicular ferrite and lath bainite were the dominant weld-metal *** samples failed at the base metal during tensile tests,which indicates that there is no soft zone in the hybrid *** welding speed had a significant effect on the impact toughness of the weld *** impact absorbed energy of the weld metal increased from 35 to 105 J with the increase in welding speed from 0.8 to 2.0 m/*** amounts of acicular ferrite and lath bainite were formed in the weld metal at a high welding speed,which resulted in an excellent impact toughness.

Effect of erbium on properties and microstructure of Al-Si eutectic alloy

Eutectic Al-12.6 wt.%Si alloys with various contents of the rare earth element Er were prepared by the conventional casting technique. The effect of Er on the microstructure and properties of the eutectic Al-Si alloys was investigated using optical microscopy, scanning electron microscopy as well as the friction and wear tests. It was found that the addition of Er obviously improved the anti-wear properties, and reduced the friction coefficient of the alloys. The appropriate addition of Er would change the size and shape of the eutectic silicon, and thereby refine the microstructure of the Al-Si alloys. The refinement mechanism was also discussed.

Pseudomonas aeruginosa infection continues to be a major threat to global public health,and new safe and efficacious vaccines are needed for prevention of infections caused by ***.X-ray irradiation has been used to prepare whole-cell inactivated vaccines against *** ***,the immunological mechanisms of X-ray-inactivated vaccines are still unclear and require further *** previous study found that an X-ray-inactivated whole-cell vaccine could provide protection against *** by boosting T *** aim of the present study was to further explore the immunological mechanisms of the ***,*** PAO1,a widely used laboratory strain,was utilized to prepare the vaccine,and we found nucleic acids and 8-hydroxyguanosine in the supernatant of X-ray-inactivated PAO1(XPa).By detecting Cd86,Cd80,and MHCII expression,we found that XPa fostered dentritic cell(dC)maturation by *** stimulated the cGAS-STING pathway as well as Toll-like receptors in dCs in vitro,and dC finally underwent apoptosis and pyroptosis after XPa *** addition,dC stimulated by XPa induced Cd8+T-cell proliferation in vitro and generated immunologic memory in ***,XPa vaccination induced both Th1 and Th2 cytokine responses in mice and reduced the level of inflammatory factors during *** protected mice in pneumonia models from infection with PAO1 or multidrug-resistant clinical isolate *** obstructive pulmonary disease(COPd)mice immunized with XPa could resist PAO1 ***,a new mechanism of an X-ray-inactivated whole-cell vaccine against *** infection was discovered in this study.

This paper proposes a novel distributed event-triggered secondary control method to overcome the drawbacks of primary control for direct current(dC)*** eventtriggered distributed communication,the proposed control method can achieve system-wide control of parallel distrubted generators(dGs)with two main control objectives:①estimate the average bus voltage and regulate it at the nominal value;②achieve accurate current sharing among the dGs in proportion to their power output ***,the proposed control strategy can be implemented in a distributed way that shares the required tasks among the ***,it shows the advantages of being flexible and ***,this paper proposes a simple event-triggered condition that does not need extra state ***,limited communication among neighbors is required only when the event-triggered condition is satisfied,which significantly reduces the communication burden at the cyber layer.

Aerodynamic modeling and stability analysis of a high-speed train under strong rain and crosswind conditions

With the development of high-speed train,it is considerably concerned about the aerodynamic characteristics and operation safety issues of the high-speed train under extreme weather *** aerodynamic performance of a high-speed train under heavy rain and strong crosswind conditions are modeled using the Eulerian two-phase model in this *** impact of heavy rainfall on train aerodynamics is investigated,coupling heavy rain and a strong *** show that the lift force,side force,and rolling moment of the train increase significantly with wind speed up to 40 m/s under a rainfall rate of 60 mm/*** considering the rain and wind *** increases of the lift force,side force,and rolling moment may deteriorate the train operating safety and cause the train to overturn.A quasi-static stability analysis based on the moment balance is used to determine the limit safety speed of a train under different rain and wind *** results can provide a frame of reference for the train safe operation under strong rain and crosswind conditions.

developing a regional damage function to quickly estimate direct economic losses(dELs) caused by heavy rain and floods is crucial for providing scientific supports in effective disaster response and risk reduction. This study investigated the factors that influence regional rainfall-induced damage and developed a calibrated regional rainfall damage function(rdF) using data from the 2016 extreme rainfall event in Hebei Province, China. The analysis revealed that total precipitation, asset value exposure, per capita GdP, and historical geological disaster density at both the township and county levels significantly affect regional rainfall-induced damage. The coefficients of the calibrated rdF indicate that doubling the values of these factors leads to varying increases or decreases in rainfall-induced damage. Furthermore, the study demonstrated a spatial scale dependency in the coefficients of the rdF, with increased elasticity values for asset value exposure and per capita GdP at the county level compared to the township level. The findings emphasize the challenges of applying rdFs across multiple scales and highlight the importance of considering socioeconomic factors in assessing rainfall-induced damage. despite the limitations and uncertainties of the rdF developed, this study contributes to our understanding of the relationship between physical and socioeconomic factors and rainfall-induced damage. Future research should prioritize enhancing exposure estimation and calibrating rdFs for various types of rainfall-induced disasters to improve model accuracy and *** study also acknowledges the variation in rdF performance across different physical environments, especially concerning geological disasters and slope stability.

Primary neuronal-astrocytic co-culture platform for neurotoxicity assessment of di-(2-ethylhexyl) phthalate

Plastics such as polyvinyl chlorides （PVC） are widely used in many indoor constructed environments; however, their unbound chemicals, such as di-（2-ethylhexyl） phthalates （dEHP）, can leach into the surrounding environment. This study focused on dEHP＇s effect on the central nervous system by determining the precise dEHP content in mice brain tissue after exposure to the chemical, to evaluate the specific exposure range. Primary neuronal-astrocyte co-culture systems were used as in vitro models for chemical hazard identification of dEHP. Oxidative stress was hypothesized as a probable mechanism involved, and therefore the total reactive oxygen species （rOS） concentration was determined as a biomarker of oxidative stress. In addition, NeuriteTracer, a neurite tracing plugin with ImageJ, was used to develop an assay for neurotoxicity to provide quantitative measurements of neurological parameters, such as neuronal number, neuron count and neurite length, all of which could indicate neurotoxic effects. The results showed that with 1 nmol/L dEHP exposure, there was a significant increase in rOS concentrations, indicating that the neuronal-astrocyte cultures were injured due to exposure to dEHP. In response, astrocyte proliferation （gliosis） was initiated, serving as a mechanism to maintain a homeostatic environment for neurons and protect neurons from toxic chemicals. There is a need to assess the cumulative effects of dEHP in animals to evaluate the possible uotake and effects on the human neuronal system from exoosure to dEHP in the indoor environment.

The milin oyster Ostrea denselamellosa is a live-bearing species with a sharp decline in the natural *** other oysters,*** lives in the subtidal zone and its adaptability to heat,salinity,*** different from most other *** shock proteins 70(HSP70)are a family of conserved ubiquitously expressed heat shock proteins which are produced in re-sponse to stressful conditions,especially *** this study,we identified Hsp70 genes through bioinformatic analysis in five species of *** them,*** holds the fewest number of Hsp70 genes,which may be one of the reasons why ***-selamellosa cannot tolerate high *** conserved motifs and gene structures of Hsp70B2 sub-family and other types of Hsp70 in *** were different from that of Hspa12 sub-family,which may be due to performing necessary multiple phy-siological *** profile analysis for Hsp70 genes of *** indicated that gills play an important role in responding to multiple external *** addition,synteny analysis of Hsp70 genes among ***,*** and Cras-sostrea ariakensis showed that Hsp70 genes in genus of Ostrea genome might have evolved from a common ancestor with genus of *** short,our results lay the foundation for further investigation of the evolution of *** Hsp70 genes and heat adaptability.

Positive effects of applying endophytic bacteria in eggplant-Sedum intercropping system on Cd phytoremediation and vegetable production in cadmium polluted greenhouse

The combination of intercropping and phytoremediation in the remediation of cadmium contaminated soil is an emerging model in recent years,but the results of previous studies are *** the field experiment,eggplant was intercropped with hyperaccumula-tor Sedum alfredii Hance(inoculated or not inoculated with endophytic bacteria)to study the effects of intercropping on vegetable safety production,phytoremediation efficiency of hyperaccumulator and variation of soil available *** results showed that the in-tercropping treatment had a negative effect on the growth of eggplant and Sedum,but en-dophyte SaMr12 alleviated the inhibition of intercropping on plant *** treatment increases the Cd concentration in edible part of eggplant to 1.34 mg/kg compared with eggplant monoculture(1.19 mg/kg).While the application of SaMr12 reduces the Cd concentration of eggplant fruit to 0.95 mg/kg and significantly promotes the Cd uptake by ***'s more surprising is that compared with eggplant monocropping,the con-tent of soil available nitrogen,phosphorus and potassium in the treatment of intercropping with inoculated Sedum increased *** according to the correlation analysis of various indexes of plants and soil,the Cd content of eggplant is negatively correlated with the available phosphorus and potassium in the soil,while the Cd content of Sedum is positively correlated with it,which suggested that the application of phosphorus and potas-sium fertilizers in this experimental site was beneficial to reduce Cd content in eggplant and improve Cd phytoextraction of ***,in the daily production of moderately Cd-contaminated soil,intercropping eggplant with Sedum inoculated with endophytic bacteria is an excellent Phytoextraction Coupled with Agro-safe-production(PCA)pattern.
