Investigation on cyanobacterial production of the proposed neurotoxinβ-N-methylamino-L-alanine(BMAA)

β-N-methylamino-L-alanine(BMAA)is an environmental neurotoxin thought to be produced by ***,the cyanobacterial origin of BMAA remains *** detection method and culture conditions of cyanobacteria are often cited as factors behind the discrepancy of published *** showed previously that BMAA was highly toxic to the cyanobacterium Nostoc PCC 7120,and it is taken up via an amino acid transport *** a mutantΔnatAΔbgtA deficient in amino acid transport as a genetic control,we show here that BMAA taken up from the medium can be detected quantitatively at a threshold similar to,or below those reported,but was undetectable in the *** BMAA isomer,2,4-diaminobutanoic acids(DAB),but not BMAA,could be detected in cell free extracts of Nostoc PCC ***-term(20 days)diazotrophic growth or nonlimiting supply of phosphate,conditions reported to enhance BMAA synthesis,did not lead to the detection of *** UPLC-MS/MS signal with a similar retention time to BMAA was found after prolonged diazotrophic incubation,but did not have fragment ions of BMAA after further *** extended to 29 different cyanobacterial strains and 6 natural cyanobacterial bloom samples,none of them was found to produce *** cytotoxicity of BMAA to cyanobacteria,and the lack of a cellular protective mechanism against such toxicity,contradict the presence of a BMAA synthesis pathway in these *** specific methods for BMAA detection in vivo need to be developed to clarify the cyanobacterial origin of BMAA.

Variation in the emission rate of sounds in a captive group of false killer whales Pseudorca crassidens during feedings: possible food anticipatory vocal activity?

This study examines whether a group of captive false killer whales(P seudorca crassidens) showed variations in the vocal rate around feeding times. The high level of motivation to express appetitive behaviors in captive animals may lead them to respond with changes of the behavioral activities during the time prior to food deliveries which are referred to as food anticipatory activity. False killer whales at Qingdao Polar Ocean World(Qingdao, China) showed signifi cant variations of the rates of both the total sounds and sound classes(whistles, clicks, and burst pulses) around feedings. Precisely, from the Transition interval that recorded the lowest vocalization rate(3.40 s/m/d), the whales increased their acoustic emissions upon trainers' arrival(13.08 s/m/d). The high rate was maintained or intensifi ed throughout the food delivery(25.12 s/m/d), and then reduced immediately after the animals were fed(9.91 s/m/d). These changes in the false killer whales sound production rates around feeding times supports the hypothesis of the presence of a food anticipatory vocal activity. Although sound rates may not give detailed information regarding referential aspects of the animal communication it might still shed light about the arousal levels of the individuals during different social or environmental conditions. Further experiments should be performed to assess if variations of the time of feeding routines may affect the vocal activity of cetaceans in captivity as well as their welfare.

Makinoella parva,a new species of the genus Makinoella(Oocystaceae,Trebouxiophyceae,Chlorophyta)

Makinoella Okada is a genus of coccoid green algae belonging to the family Oocystaceae with typical morphological *** to date included only one type *** the genus is rarely reported,there is a lack of taxonomic *** this study,17 strains of this genus were collected from several different places in China,and their taxonomic studies were conducted based on morphological and molecular phylogenetic *** phylogenetic results based on the analysis of ITS,chloroplast genes rbc L and tuf A showed that the genus was divided into two *** of the branches comprised newly collected *** base changes(CBCs)and hemi-compensatory base changes(hemi-CBCs)within the secondary structure of the ITS-2 confirmed the branches as two *** with the type species of Makinoella tosaensis,the cell size of this new branch was only about 50%,and included 2-4 colonial ***,based on the smaller cell size,simpler colony composition,independent phylogenetic position and CBCs and hemi-CBCs,we suggest that the new clade should be designated as a new taxon of Makinoella,namely Makinoella *** the four molecular markers using in this study,the rbc L and newly introduced ITS are recommended for species separation,which can help further studies to revise the species and generic concepts of the family.

Relationship between morphospecies and microcystin-producing genotypes of Microcystis species in chinese freshwaters

Twenty water bodies in China were sampled,and 186 strains of different Microcystis species were isolated,from which eight morpho species were identified and 43 stains containing the mcyB gene were *** analysis based on the mcyB gene indicated that the microcystin(MC)-producing Microcystis in China could be divided into two groups(ⅠandⅡ)and showed significant differences between the two *** maximum sequence similarity was 69.1%.Microcystins(MCs)were measured by high-performance liquid chromatography(HPLC)analysis,and no microcystin-RR(MC-RR)was detected in some strains belonging to GroupⅡ.Compared to other regions of the world,the proportion of chinese MC-producing was different,and the regional differences were more obvious.A whole-cell polymerase chain reactio(PCR)assay was conducted to analyze the proportion of the mcyB gene in the laboratory cultured and field cultured *** proportion of four morphospecies(***,***,***,and ***)that contained the mcyB gene exceeded 50%in the field cultured sample *** with former studies,*** was the mo st likely morphotype that can produce MCs in the *** study provided new insight of Microcystis hazard assessment and field monitoring.

Correlation between the distribution pattern of virulence genes and virulence of Aeromonas hydrophila strains

Nine strains of Aeromonas hydrophila isolated from diseased fish or soft-shelled tortoise were tested for the presence of three virulence genes including the genes encoding aerolysin,hemolysin,and extracellular serine protease(i.e.,aerA,hlyA,and ahpA,respectively).These genes were investigated using polymerase chain reaction(PCR)with specific primers for each *** the pathogenicities to Carrassius auratus ibebio of these strains were also *** results demonstrated that the distribution patterns of aerA,hlyA,and ahpA were different in these strains.6/9 of *** strains were aerA positive,8/9 of strains hlyA positive,7/9 of strains ahpA positive,***,the assay for pathogenesis showed that two strains(*** XS91-4-1 and C2)were strong virulent,two strains(*** ST78-3-3 and 58-20-9)avirulent and the rest middle virulent was to the *** conclusion,there are significant correlation between the distribution pattern of the three virulence genes and the patho-genicity to Carrassius auratus *** strong virulent *** strains were aerA+hlyA+ahpA+genotype,and all aerA+hlyA+ahpA+strains were *** with the genotype of aerA−hlyA−ahpA+have middle *** the present study,we found for the first time that all *** isolated from the ahpA positive were virulent to Carrassius auratus ***,there was a positive correlation between the virulence of *** and the presence of aerA and ahpA.

PCR-RFLP analysis of mitochondrial DNA ND5/6 region among 3 subspecies of common carp (Cyprinus carpio L.)and its application to genetic discrimination of subspecies

Mitochondrial DNA ND5/6 region was studied by PCR-RFLP analysis among ten representative strains belonging to three subspecies (Cyprinus carpio carpio, Cypri-nus carpio haematopterus and Cyprinus carpio rubrofuscus) of common carp (Cyprinus carpio L.). A total of 2.4 kb frag-ment was amplified and subjected to restriction endonucle-ase analysis with nine restriction endonucleases *** results indicated that each subspecies owned one hyplo-type and four restriction enzymes (DdeⅠ, HaeⅢ, TaqⅠand MboⅠ) produced diagnostic restriction sites which could be used for discriminating the three subspecies and as molecu-lar genetic markers for assistant selective breeding of com-mon carp.

Diverse and variable sex determination mechanisms in vertebrates

Sex is prevalent in nature and sex determination is one of the most fundamental biological processes, while the way of initiating female and male development exhibits remarkable diversity and variability across vertebrates. The knowledge on why and how sex determination mechanisms evolve unusual plasticity remains limited. Here, we summarize sex determination systems,master sex-determining genes and gene-regulatory networks among vertebrates. Recent research advancements on sex determination system transition are also introduced and discussed in some non-model animals with multiple sex determination mechanisms. This review will provide insights into the origin, transition and evolutionary adaption of different sex determination strategies in vertebrates, as well as clues for future perspectives in this field.

A novel male-specific SET domain-containing gene setdm identified from extra microchromosomes of gibel carp males

Various genes have been screened on extra chromosomes, but their molecular characterization,expression pattern and biological function are still unclear. Here, we utilized a male-specific sequence of polyploid gibel carp(Carassius gibelio) to identify a novel male-specific SET(Su(var)3–9, Enhancerof-zeste, Trithorax) domain-containing gene setdm on extra microchromosomes of gibel carp *** setdm was characterized in molecule and expression aspects, in which its expression was specific to testis, and had relative high transcription during middle/late stages of testis ***, prominent expression of Setdm in spermatogenic cells was observed in testis through immunofluorescence co-localization analysis. These results suggest that biological function of setdm might be related to testis development and spermatogenesis of gibel carp. Additionally, the homeologous gene setdmf of setdm, was also characterized, and its expression was gonad-specific, in which its expressed product was detected to mainly distribute in gametogenic cells of testis and ovary, and to have dynamic expression pattern similar to that of setdm. Based on the current results, we propose that the novel male-specific setdm on extra microchromosomes might be functional divergence gene of the gonad-specific setdmf. Therefore, these findings will help us to further understand evolutionary fate and functional role of genes on extra microchromosomes.
