To the Editor:Current guidelines recommend that for patients with acute coronary syndrome(ACS)undergoing percutaneous coronary intervention(PCI),dual antiplatelet therapy(DAPT),preferably involving powerful P2Y_(12) receptor inhibitors,including ticagrelor or prasugrel,should be used for at least 12 months.^([1–3])However,there is a lack of relevant evidence regarding the feasibility of de-escalation treatment and specific DAPT treatment regimens for ACS patient populations with high ischemic risk.

Osteonecrosis of the femoral head(ONFH)is mainly caused by a decrease in the vascular supply to the subchondral bone of the femoral head,leading to the death of bone cells,degeneration and necrosis of the subchondral bone,and eventually collapse of the femoral head.^([1])Early and accurate diagnosis is the key to successful hip-preserving therapy,and the size/location of necrosis is the main determinant for ONFH treatment.Previous studies have shown that many hip-preserving therapies achieved satisfactory results in ONFH patients with Association Research Circulation Osseous(ARCO)I–II.When ONFH progresses to ARCO IV with clinical manifestations of osteoarthrosis,total hip arthroplasty(THA)may be the only option.

Computer-generated holography technology has been widely applied,and as research in this field deepens,the demand for memory and computational power in small AR and VR devices continues to increase.This paper presents a hologram generation method,i.e.,a symmetrically high-compressed look-up table method,which can reduce memory usage by50%.In offline computing,half of the basic horizontal and vertical modulation factors are stored,halving the memory requirements without affecting inline speed.Currently,its potential extends to various holographic applications,including the production of optical diffraction elements.

BACKGROUND We report a rare case of primary clinical presentation featuring elevated creatine kinase(CK)levels in a neonate,which is associated with the LAMA2 gene.In this case,a heterozygous mutation in exon5 of the LAMA2 gene,c.715C>G(resulting in a change of nucleotide number 715 in the coding region from cytosine to gua-nine),induced an amino acid alteration p.R239G(No.239)in the patient,repre-senting a missense mutation.This observation may be elucidated by the neonatal creatine monitoring mechanism,a phenomenon not previously reported.CASE SUMMARY We analysed the case of a neonate presenting solely with elevated CK levels who was eventually discharged after supportive treatment.The chief complaint was identification of increased CK levels for 15 d and higher CK values for 1 d.Ad-mission occurred at 18 d of age,and despite prolonged treatment with creatine and vitamin C,the elevated CK levels showed limited improvement.Whole exo-me sequencing revealed the presence of a c.715C>G mutation in LAMA2 in the newborn,correlating with a clinical phenotype.However,the available informa-tion offers insufficient evidence for clinical pathogenicity.CONCLUSION Mutations in LAMA2 are associated with the clinical phenotype of increased neonatal CK levels,for which no specific treatment exists.Whole genome sequen-cing facilitates early diagnosis.

In recent years,the field of endoscopy has experienced significant technological advancements.These developments have facilitated the efficient use of endoscopy for precise examination and minimally invasive treatment of various lesions.A key parameter considered during both endoscopic diagnosis and treatment is the size of the lesion.In colon polypectomy,the size of the polyp is linked to the probability of malignancy and the likelihood of complications.Similarly,in the case of early gastric cancer,larger tumors are often associated with an increased likelihood of lymph node metastasis,and whether to perform endoscopic submucosal dissection(ESD)is partially dependent on the size of the lesion.Therefore,accurate measurement of lesion size may significantly impact the diagnosis and treatment plan for the patient.However,at present,there is no standardized method for quantitative measurement during endoscopy.This lack of a universal technique hinders the ability of clinical guidelines to provide precise recommendations for treatment.

Endoplasmic reticulum stress and mitochondrial dysfunction play important roles in Parkinson s disease,but the regulato ry mechanism remains elusive.Prohibitin-2(PHB2)is a newly discove red autophagy receptor in the mitochondrial inner membrane,and its role in Parkinson’s disease remains unclear.Protein kinase R(PKR)-like endoplasmic reticulum kinase(PERK)is a factor that regulates cell fate during endoplasmic reticulum stress.Parkin is regulated by PERK and is a target of the unfolded protein response.It is unclear whether PERK regulates PHB2-mediated mitophagy thro ugh Parkin.In this study,we established a 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP)-induced mouse model of Parkinson’s disease.We used adeno-associated virus to knockdown PHB2 expression.Our res ults showed that loss of dopaminergic neurons and motor deficits were aggravated in the MPTP-induced mouse model of Parkinson’s disease.Ove rexpression of PHB2 inhibited these abnormalities.We also established a 1-methyl-4-phenylpyridine(MPP+)-induced SH-SY5Y cell model of Parkinson’s disease.We found that ove rexpression of Parkin increased co-localization of PHB2 and microtubule-associated protein 1 light chain 3,and promoted mitophagy.In addition,MPP+regulated Parkin involvement in PHB2-mediated mitophagy through phosphorylation of PERK.These findings suggest that PHB2 participates in the development of Parkinson’s disease by intera cting with endoplasmic reticulum stress and Parkin.

The application of microorganisms as probiotics is limited due to lack of safety evaluation.Here,a novel multi-stress-tolerant yeast Meyerozyma guilliermondii GXDK6 with aroma-producing properties was identified from marine mangrove microorganisms.Its safety and probiotic properties were assessed in accordance with phenotype and whole-genome sequencing analysis.Results showed that the genes and phenotypic expression of related virulence,antibiotic resistance and retroelement were rarely found.Hyphal morphogenesis genes(SIT4,HOG1,SPA2,ERK1,ICL1,CST20,HSP104,TPS1,and RHO1)and phospholipase secretion gene(VPS4)were annotated.True hyphae and phospholipase were absent.Only one retroelement(Tad1-65_BG)was found.Major biogenic amines(BAs)encoding genes were absent,except for spermidine synthase(JA9_002594),spermine synthase(JA9_004690),and tyrosine decarboxylase(inx).The production of single BAs and total BAs was far below the food-defined thresholds.GXDK6 had no resistance to common antifungal drugs.Virulence enzymes,such as gelatinase,DNase,hemolytic,lecithinase,and thrombin were absent.Acute toxicity test with mice demonstrated that GXDK6 is safe.GXDK6 has a good reproduction ability in the simulation gastrointestinal tract.GXDK6 also has a strong antioxidant ability,β-glucosidase,and inulinase activity.To sum up,GXDK6 is considered as a safe probiotic for human consumption and food fermentation.

α-Fe_(2)O_(3)/epoxy resin composite superhydrophobic coating was prepared withα-Fe_(2)O_(3) nanoparticles and epoxy resin by spin coating method.The coating without epoxy resin has higher contact angle(CA)and lower ice adhesion strength(IAS),but the mechanical properties are poor.Theα-Fe_(2)O_(3)/epoxy resin composite superhydrophobic coating exhibits good mechanical durability.In addition,compared with the bare aluminum substrate,the Ecorr of the composite coating is positive and the Jcorr is lower.The inhibition efficiency of the composite coating is as high as 99.98%in 3.5 wt%NaCl solution.The difference in the microstructure caused by the two preparation methods leads to the changes in mechanical properties and corrosion resistance of composite superhydrophobic coating.

Ribonucleotide reductase M2(RRM2)is a small subunit in ribonucleotide reduc-tases,which participate in nucleotide metabolism and catalyze the conversion of nucleotides to deoxynucleotides,maintaining the dNTP pools for DNA biosynthesis,repair,and replication.RRM2 performs a critical role in the malignant biological behaviors of cancers.The structure,regulation,and function of RRM2 and its inhibitors were discussed.RRM2 gene can produce two transcripts encoding the same ORF.RRM2 expression is regulated at multiple levels during the processes from transcription to translation.Moreover,this gene is associated with resistance,regulated cell death,and tumor immunity.In order to develop and design inhibitors of RRM2,appropriate strategies can be adopted based on different mechanisms.Thus,a greater appreciation of the characteristics of RRM2 is a benefit for understanding tumorigenesis,resistance in cancer,and tumor microenvironment.Moreover,RRM2-targeted therapy will be more attention in future therapeutic approaches for enhancement of treatment effects and amelioration of the dismal prognosis.
