Pharmacognostic Identification of Hedyotis auricularia and Mitracarpus villosus

基于质量源于设计(QbD)理念的喘贴灵提取工艺研究

目的基于质量源于设计(QbD)理念设计并优化喘贴灵水提工艺。方法采用单因素实验筛选喘贴灵水提最佳实验点,在此基础上,以盐酸麻黄碱转移率、芥子碱硫氰酸盐转移率和浸膏得率为评价指标。选择液料比、提取时间及pH值作为主要考察因素,进行Box-Behnken设计(BBD)。建立关键质量属性(CPPs)与关键工艺参数(CQAs)的数学模型,考察CQAs与CPPs的交互作用,建立喘贴灵提取工艺设计空间并进行优化,保障该工艺的稳定性。使用高效液相色谱法(HPLC)分析喘贴灵提取液中主要有效成分。结果通过单因素筛选实验,选择液料比为8∶1,提取时间为90 min,pH值为4以及提取次数3次为最佳实验点进行后续实验。BBD方差分析结果显示回归模型的P值小于0.05,且失拟值大于0.01,表明所建模型具有较好的预测能力。喘贴灵水提工艺CPPs的操作空间结合实际生产的情况,最佳提取工艺为加水倍数9倍,提取时间72 min,pH值4.5。HPLC结果显示喘贴灵提取液中含盐酸麻黄碱、芥子碱硫氰酸盐、延胡索乙素、甲基丁香酚、桂皮醛和6-姜酚的峰面积分别占喘贴灵提取液总峰面积的0.94%、14.32%、0.78%、31.23%、0.34%和0.44%。结论基于QbD理念建立的喘贴灵水提工艺设计空间,有利于实际生产操作,保障其工艺稳定性。

Synthesis,Crystal Structure and Its Application in Living Cells Imaging of Benz(c)acridine-1,2,3-triazole Derivatives

A highly selective iron ions fluorescent probe based on the benz(c)acridine-1,2,3-triazole derivatives was produced by multi-step reactions. 1-(7-Benz[c]acridinyl)-4-(4-methylphenyl)-1,2,3-triazole(4 a), C_(26)H_(18)N_(4), was structurally determined by single-crystal X-ray diffraction. It crystallizes in the trigonal system, space group R-3 with a = 36.230(10), b = 36.230(10), c = 7.993(3)Å, β = 90°, V = 9086(6)Å^(3), Z = 18, D_(c)= 1.271 g/cm^(3), F(000) = 3636, μ = 0.602 mm^(-1), the final R = 0.0865 and wR = 0.1619 for 3951 observed reflections(I > 2σ(I)). X-ray analysis indicated that all four rings of benz(c)acridine are in the same plane, and the 1,2,3-triazole ring and the corresponding linked benzene(C(1)–C(6)–C(7)–C(8)–C(9)–N(1)) are approximately perpendicular with a dihedral angle of 106.5°. The crystal packing of the compound was stabilized by two weak interactions between C(11)–H(10)…N(4) and C(18)–H(18)…N(1). In fluorescence spectra studies, compound 4 c was exhibited good selectivity and sensitivity towards iron ions in DMSO: mops buffer solution. Furthermore, 4 c was successfully used for imaging iron ions in living He La cervical cancer cells.

Pharmacognostic Identification of Borreriae Strictae Herba and Borreriae Latifoliae Herba

Research Progress of Dendrobium officinale Kimura et Migo

The effects of light on the quality and related genes of Dendrobium officinale were summarized, and the main chemical components and pharmacological effects of D. officinale were analyzed. The biomass accumulation of D. officinale is related to light quality and light intensity. Monochromatic light quality can make polysaccharide accumulate, and mixed light quality can promote cell proliferation and component accumulation of D. officinale. Under different light intensity, the polysaccharide content of D. officinale is different, and it changes with the change of light intensity. When a certain intensity is exceeded, the polysaccharide content decreases with the increase of light intensity. The chemical components of D. officinale are polysaccharides, alkaloids, amino acids, volatile components, etc. The analysis of the growth and components of D. officinale provides reference and basis for improving the quality and chemical components of D. officinale and promotes the research and development of related products of D. officinale.

Study on the Microscopic Identification of Chenopodium album L.

[Objectives]This study was conducted to identify the microscopic characteristics of Chenopodium album L.[Methods]The microscopic identification method was adopted.[Results]The xylem vessels and fiber bundles of the roots are arranged into 3-4 intermittent ring belts in a concave-convex pattern,alternately with the parenchymal cell ring belts;and the xylem rays vary in *** cross section of the stems is polygonal;there are parenchyma cells at the corners of the cortex;there are many vascular bundles of varying sizes;and calcium oxalate clusters are common in the medulla and *** are 2 to 4 vascular bundles in the main leaf vein,peltate;and fiber bundles exist below the *** powder is characterized by numerous clusters of calcium oxalate,which are uniform in size,sharp at edges and corners.[Conclusions]This study can provide reference for the identification and quality standard of the crude drug.

Safety Limit Test of Heavy Metals and Pesticide Residues in the Zhuang Ethnic Medcine Herba Polygoni Chinensis from 13 Producing Areas

[Objectives]This study was conducted to determine the contents of such five heavy metals as lead(Pb),cadmium(Cd),arsenic(As),mercury(Hg)and copper(Cu)and the contents of nine kinds of organochlorine pesticide residues including total BHC(α-BHC,β-BHC,γ-BHC,δ-BHC)and total DDT(pp-DDE,pp-DDD,op-DDT,pp-DDT)and pentachloronitrobenzene in the Zhuang medicine Herba Polygoni Chinensis from 13 producing areas.[Methods]The five heavy metals were determined in accordance with the second method of general rule 2341,the inductively coupled plasma mass spectrometry method,in part 4 of the 2020 edition of chinese Pharmacopoeia;and the nine organochlorines were determined in accordance with the first method of general rule 2341,organochlorine pesticide residue determination method(chromatography),in part 4 of the 2020 edition of chinese Pharmacopoeia.[Results]In the Herba Polygoni Chinensis from 13 origins,the lead contents were in the range of 0.50-3.28 mg/kg;the cadmium contents were in the range of 0.06-4.21 mg/kg;the arsenic contents were in the range of 0.16-6.9 mg/kg;the copper contents were in the range of 10.5-45.1 mg/kg;the mercury contents were in the range of 0.00-0.003 mg/kg;and the nine organochlorines including the total benzene hexachloride(α-BHC,β-BHC,γ-BHC,δ-BHC),total DDT(pp-DDE,pp-DDD,op-DDT,pp-DDT)and pentachloronitrobenzene were all not *** to part 4 of the 2020 edition of chinese Pharmacopoeia,the Cu and As of the Herba Polygoni Chinensis produced in Hezhou exceeded the standards,the Cu of the Herba Polygoni Chinensis produced in Wuzhou exceeded the standard,and the As of the Herba Polygoni Chinensis produced in Yulin City and Guilin City exceeded the standard.[Conclusions]This study provides safety limit test data for the development and utilization of Herba Polygoni Chinensis of the Zhuang medicine.

Analysis of the Volatile Components of Polygonum chinense *** with Different Solvents by GC-MS

[Objectives]This study was conducted to investigate the volatile components of Polygonum chinense L.[Methods]The volatile components of the methanol extract,ethyl acetate extract,chloroform extract and petroleum ether extract of *** were analyzed and determined by gas chromatography.[Results]The volatile components of the methanol extract from *** were the most,and phenolic acids and ketones accounted for a relatively high proportion,among which pyrogallic acid had the highest *** ethyl acetate extract contained the second most volatile components,mostly acids and esters,of whichβ-sitosterol had the highest content;the chloroform and petroleum ether extracts had relatively few types of volatile components,most of which were alkanes,and the content ofγ-sitosterol shared by the two was the *** common substances of the four extracts were palmitic acid and phytol.[Conclusions]This study provides a theoretical basis for the quality evaluation of *** and a scientific basis for its resource development.

Research trends and development status of TCM network pharmacology: a bibliometric study

Objective: To analyze the research status, development context and trend of traditional chinese medicine (TCM) network pharmacology from birth to the present. Methods: In this study, bibliographic data and references of articles on TCM network pharmacology were collected from Web of Science Core Collection (WoSCC), and software including CiteSpace, VOSviewer and Microsoft Excel were applied for the bibliometric analysis of periodicals and the quantity of literature, and for the construction and analysis of the co-occurrence network of countries, journals, research institutes and authors, clusters of co-cited references, and time distribution network of keywords. Results: 249 articles were included in this study. The time span was from 2011 to 2018, and the annual volume of publications showed an increasing trend year by year. The 77 journals contain relevant literature, and the average impact factor of the journal is 3.215. chinese Journal of Natural Medicines, Scientific Reports, PLoS One have high influence in the field of TCM network pharmacology. Universities and research institutes including China Academy of chinese Medical Science and Northwest Agriculture & Forestry University are the major forces in this field. Wang Yonghua and Li Shao are authors with high influence in this field, with a considerable number of relevant documents and citations. According to the clustering results of co-cited references, research topics can be classified into 13 development contexts such as “compound danshen formula”,“computational tool”, and “new multi-compound drug discovery”. Hotspots of TCM network pharmacology include “drug discovery”,“mechanism” and “cardiovascular disease”, and relevant research methods and research contents including drugs and diseases tend to be constantly enriched. Effects of Leaf Extract of Phyllanthus reticulatus from guangxi on Human Gastric Cancer SGC-7901 Cells in vitro

[Objectives]To observe the effect of drug-containing serum from different extracts of Phyllanthus reticulatus leaf on the proliferation of human gastric cancer SGC-7901 cells and the effect of in vivo on the life extension rate of mice with H22 ascites tumor,and to investigate the effects of chemical components such as Corilagin,ethyl gallate,ethyl brevifolincarboxylate,and gallic acid on the proliferation of human gastric cancer SGC-7901 cells.[Methods]MTT method was used to observe the inhibitory effect of drug-containing serum of different extracts of *** leaf on human gastric cancer SGC-7901 cells in vitro to determine its active *** active site was used as the research object to establish a Kunming mouse ascites tumor model,to investigate the effect on the life extension rate of mice with H22 ascites tumor,to further separate the monomer components from the effective fraction and investigate its effect on human gastric cancer SGC-7901 cells.[Results]Compared with the 10%blank serum control group,10%drug-containing serum of ethyl acetate and n-butanol in *** leaf had significant inhibitory effect on the proliferation of human gastric cancer SGC-7901 cells,and the inhibition rate was 34.99%and 28.68%,respectively(P<0.05).Compared with the blank group,the survival time of the high dose group of ethyl acetate in *** leaf was significant(P<0.05).Gallic acid,Corilagin,and Brevifolincarboxylic acid ethyl ester had a significant inhibitory effect on the proliferation of human gastric cancer SGC-7901 cells,with IC 50 of 26.52,70.45,and 158.86μg/mL,*** gallate had no inhibitory effect on the proliferation of human gastric cancer SGC-7901 cells,and its IC 50 was 251.96μg/mL.[Conclusions]The drug-containing serum of ethyl acetate and n-butanol extract of *** can inhibit the proliferation of human gastric cancer SGC-7901 *** acetate of *** leaves can increase the life extension rate of mice with H
