Hypoxia-preconditioned bone marrow-derived mesenchymal stem cells protect neurons from cardiac arrest-induced pyroptosis

Cardiac arrest can lead to severe neurological impairment as a result of inflammation,mitochondrial dysfunction,and post-cardiopulmonary resuscitation neurological *** preconditioning has been shown to improve migration and survival of bone marrow–derived mesenchymal stem cells and reduce pyroptosis after cardiac arrest,but the specific mechanisms by which hypoxia-preconditioned bone marrow–derived mesenchymal stem cells protect against brain injury after cardiac arrest are *** this end,we established an in vitro co-culture model of bone marrow–derived mesenchymal stem cells and oxygen–glucose deprived primary neurons and found that hypoxic preconditioning enhanced the protective effect of bone marrow stromal stem cells against neuronal pyroptosis,possibly through inhibition of the MAPK and nuclear factor κB ***,we transplanted hypoxia-preconditioned bone marrow–derived mesenchymal stem cells into the lateral ventricle after the return of spontaneous circulation in an 8-minute cardiac arrest rat model induced by *** results showed that hypoxia-preconditioned bone marrow–derived mesenchymal stem cells significantly reduced cardiac arrest–induced neuronal pyroptosis,oxidative stress,and mitochondrial damage,whereas knockdown of the liver isoform of phosphofructokinase in bone marrow–derived mesenchymal stem cells inhibited these *** conclude,hypoxia-preconditioned bone marrow–derived mesenchymal stem cells offer a promising therapeutic approach for neuronal injury following cardiac arrest,and their beneficial effects are potentially associated with increased expression of the liver isoform of phosphofructokinase following hypoxic preconditioning.

To compare the impact of the scrotal vs inguinal orchidopexy approach on the testicular function of infants with cryptorchidism,a randomized controlled trial was conducted involving boys who were 6–12 months old at surgery and were diagnosed with clinically palpable,inguinal undescended *** June 2021 and December 2021,these boys at fujian Maternity and Child Health Hospital(Fuzhou,China)and fujian Children’s Hospital(Fuzhou,China)were *** randomization with a 1:1 allocation ratio was *** primary outcome was testicular function assessed by testicular volume,serum testosterone,anti-Müllerian hormone(AMH),and inhibin B(InhB)*** outcomes included operative time,amount of intraoperative bleeding,and postoperative *** 577 screened patients,100(17.3%)were considered eligible and enrolled in the *** the 100 children who completed the 1-year follow-up,50 underwent scrotal orchidopexy and 50 underwent inguinal *** testicular volume,serum testosterone,AMH,and InhB levels in both groups increased markedly after surgery(all P0.05).No differences between the scrotal and inguinal groups were noted regarding the operative time(P=0.987)and amount of intraoperative bleeding(P=0.746).The overall complication rate(2.0%)of the scrotal group was slightly lower than that of the inguinal group(8.0%),although this difference was not statistically significant(P>0.05).Both scrotal and inguinal orchiopexy exerted protective effects on testicular function in children with cryptorchidism,with similar operative status and postoperative *** orchiopexy is an effective alternative to inguinal orchiopexy in children with cryptorchidism.

The effect of age and abstinence time on semen quality:a retrospective study

This study analyzed the effects of male age and abstinence time on semen quality and explored the best abstinence time for Chinese males among different age *** parameters,including sperm kinetics,morphology,and DNA fragmentation index(DFI),were reviewed from 2952 *** were divided into six age groups(≤25 years,26–30 years,31–35 years,36–40 years,41–45 years,and>45 years)and were divided into six groups according to different abstinence time(2 days,3 days,4 days,5 days,6 days,and 7 days).The differences in semen quality between the groups were compared,and the effect of age and abstinence time on semen quality was *** differences were observed in semen volume,progressive motility(PR),and DFI among the age groups(all P0.05).There were significant differences in semen volume,PR,and DFI among different abstinence time groups(all P0.05).Pearson analysis showed that male age and abstinence time were both significantly correlated with sperm kinetics and DFI(both P0.05).The box plots and histograms of men’s age,abstinence time,and semen quality show that most semen quality parameters differ significantly between the 2 days and 7 days abstinence groups and other groups at different *** for the sperm morphology parameters,sperm kinetic parameters and sperm DFI are linearly related to male age and abstinence time.

Factors affecting patency time and semen quality in a single-armed microsurgical vasoepididymostomy

Although microsurgical vasoepididymostomy(MVE)is an effective treatment for epididymal obstructive azoospermia,some patients may experience delayed patency or suboptimal semen parameters after ***,research into patency time,semen quality postpatency,and associated influencing factors remains *** study aimed to address these issues by evaluating 181 patients who underwent at least one-sided MVE employing asingle-armed longitudinal intussusception vasoepididymostomy technique,with a follow-up period of over 12 months for 150 *** overall patency rate was 75.3%,with 86.0%of patients achieving patency within 6 months following ***,factors such as age,history of epididymitis,duration of surgery,side of anastomosis,sperm motility in epididymal fluid,and the site of anastomosis showed no correlation with patency ***,our univariate and multivariate linear regression analysis indicated that only the site of anastomosis was positively correlated with and could independently predict postoperative total motile sperm ***,the site of anastomosis might serve as a predictor for optimal postoperative semen quality following the MVE procedure.

Background:Carbon(C),nitrogen(N),and phosphorus(P)stoichiometry is a key indicator of nutrient utilization in plants,and C/N/P ratios are related to the life histories and adaptation strategies of tree ***,no consensus has been reached on how leaf stoichiometric characteristics are affected by forest type and stand *** relationships between leaf stoichiometry and geographical,meteorological,and soil factors also remain poorly ***:Leaf and soil were sampled from forest stands of different age groups(young,middle-aged,near-mature,and mature)in two forest types(Chinese fir(Cunninghamia lanceolata)forests and evergreen broadleaved forests).The relationships between leaf C,N,and P stoichiometric parameters and geographical,meteorological,and soil factors were analysed by using redundancy analysis(RDA)and stepwise linear regression ***:Leaf C concentrations peaked in the near-mature stands with increasing age irrespective of forest *** N and P concentrations fluctuated with a rising trend in Chinese fir forests,while decreased first and increased later from young to mature phases in natural evergreen broadleaved *** fir forests were primarily limited by N and P,while natural evergreen broadleaved forests were more susceptible to P *** C,N,and P stoichiometric characteristics in Chinese fir forests were mainly affected by the soil total P concentration(SP),longitude(LNG),growing season precipitation(GSP)and mean temperature in July(JUT).The leaf C concentration was mainly affected by GSP and JUT;leaf N and P concentrations were both positively correlated with LNG;and leaf P was positively correlated with *** evergreen broadleaved forests,however,leaf stoichiometric parameters displayed significant correlations with latitude(LAT)and mean annual precipitation(MAP).Conclusions:Leaf stoichiometry differed among forest stands of different age groups and forest *** C,N,and P stoichiometry was pri

The cause of asthenozoospermia(AZS)is not well understood because of its complexity and *** some gene mutations have been identified as contributing factors,they are only responsible for a small number of *** spokes(RSs)are critical for adenosine triphosphate-driven flagellar beating and axoneme stability,which is essential for flagellum *** this study,we found novel compound heterozygous mutations in leucine-rich repeat-containing protein 23(LRRC23;c.1018C>T:p.Q340X and c.881_897 Del:p.R295Gfs*32)in a proband from a nonconsanguineous family with AZS and male ***-Quik staining and scanning electron microscopy revealed no abnormal sperm *** blotting and immunofluorescence staining showed that these mutations suppressed LRRC23 expression in sperm ***,transmission electron microscopy showed the absence of RS3 in sperm flagella,which disrupts stability of the radial spoke complex and impairs *** in vitro fertilization and embryo transfer,the proband’s spouse achieved successful pregnancy and delivered a healthy *** conclusion,our study indicates that two novel mutations in LRRC23 are associated with AZS,but successful fertility outcomes can be achieved by in vitro fertilization-embryo transfer techniques.

Ecofriendly Hydroxyalkyl Cellulose Additives for Efficient and Stable MAPbI_(3)-Based Inverted Perovskite Solar Cells

Perovskite solar cells(PSCs)have been demonstrated to be one of the most promising technologies in the field of renewable ***,the presence of the defects in the perovskite films greatly limits the efficiency and the stability of the *** additive engineering is one of the most effective approaches to overcome this *** of the successful additives are extracted from the petroleum-based materials,while the research on the biomass-based additives is still lagging *** this paper,two ecofriendly hydroxyalkyl cellulose additives,i.e.,hydroxyethyl cellulose(HEC)and hydroxylpropyl cellulose(HPC),are investigated on the performance of the MAPbl_(3)-based inverted *** to the strong interaction between the hydroxyl groups of the cellulose and the divalent cations of the perovskite,these additives enhance the crystal grain orientation and significantly repair the defects of the perovskite *** as the additives,these two cellulose derivatives show a strong passivation ability,which significantly reduces the trap density and improves the optoelectronic feature of the *** with the average power conversion efficiency(PCE)of the control device(19.19%),an enhancement of~10%is achieved after the addition of *** optimized device(PCE=21.25%)with a long-term stability(10:80 h,PCE=20.93%)is achieved by the incorporation of the HEC additives into the precursor *** is the best performance among the PSCs with the cellulose additives up to *** research provides a novel choice to develop a cost-effective and renewable additive for the PSCs with high efficiency and excellent long-term stability.
