Self-sufficient nanoparticles with dual-enzyme activity trigger radical storms and activate cascade-amplified antitumor immunologic responses

Radiotherapy(RT) can potentially induce systemic immune responses by initiating immunogenic cell death(ICD) of tumor ***,RT-induced antitumor immunologic responses are sporadic and insufficient against cancer ***,we construct multifunctional self-sufficient nanoparticles(MARS) with dual-enzyme activity(GOx and peroxidase-like) to trigger radical storms and activate the cascade-amplified systemic immune responses to suppress both local tumors and metastatic *** addition to limiting the Warburg effect to actualize starvation therapy,MARS catalyzes glucose to produce hydrogen peroxide(H_(2)O_(2)),which is then used in the Cu^(+)-mediated Fenton-like reaction and RT *** and chemodynamic therapy produce reactive oxygen species in the form of radical storms,which have a robust ICD impact on mobilizing the immune ***,when MARS is combined with RT,potent systemic antitumor immunity can be generated by activating antigen-presenting cells,promoting dendritic cells maturation,increasing the infiltration of cytotoxic T lymphocytes,and reprogramming the immuno suppre ssive tumor ***,the synergistic therapy of RT and MARS effectively suppresses local tumor growth,increases mouse longevity,and results in a 90% reduction in lung metastasis and postoperative ***,we provide a viable approach to treating cancer by inducing radical storms and activating cascade-amplified systemic immunity.

A new type of leak-floor flip bucket

Flip buckets are commonly used to discharge flow away from a hydraulic structure into the downstream to dissipate energy.A new leak-floor flip bucket is presented,making the ski-jump water jet a typical long-narrow *** on the model experiments and numerical simulation,the flow pattern,formation process and mechanism of the leak-floor flip bucket are *** results show that cross section flow shape develops from the"Y-type"to"|-type",and this is because the natural pressure difference is generated when water flows through the leak-floor area and moves transversely from both sides to the *** from the slit-type flip bucket with sidewall contraction,the leak-floor flit bucket makes the water jet narrow and long without high pressure on the side walls of the flip *** the same jet length condition,the maximum sidewall pressure of the slit-type is 4.67 times that of the leak-floor flip *** effects of flow discharge on the jet length are less significant for the leak-floor bucket than for the slit-type bucket.

Essential amino acids(EAAs)deprivation is a potential antitumor approach because EAAs are critical for tumor *** efficiently inhibit tumor growth,continuous deprivation of EAAs is required,how-ever,continuous deprivation without precise control will introduce toxicity to normal ***,a programmable double-unlock nanocomplex(ROCK)was prepared,which could self-supply phenylalanine ammonia-lyase(PAL)to tumor cells for phenylalanine(Phe)*** was double-locked in physiological conditions when administered *** ROCK actively targeted to tumor cells by integrinαvβ3/5 and CD44,ROCK was firstly unlocked by cleavage of protease on tumor cell membrane,exposing CendR and R8 to enhance ***,hyaluronic acid was digested by hyaluronidase overexpressed in endo/lysosome of tumor cells,in which ROCK was secondly unlocked,resulting in pro-moting endo/lysosome escape and PAL plasmid(pPAL)*** pPAL could sustainably express PAL in host tumor cells until the self-supplied PAL precisely and successfully deprived Phe,thereby block-ing the protein synthesis and killing tumor cells ***,our precise Phe deprivation strategy effectively inhibited tumor growth with no observable toxicity to normal cells,providing new insights to efficiently remove intratumoral nutrition for cancer therapy.

One-step growth of the interconnected carbon nanotubes/graphene hybrids from cuttlebone-derived bi-functional catalyst for lithium-ion batteries

Carbon nanotubes/graphene hybrid materials with excellent physicochemical properties can be widely ap-plied in the fields of energy storage,electrocatalysis,sensing,*** the self-stacking and achiev-ing covalent interaction between carbon nanotubes and graphene are important to ensure a stable hi-erarchical architecture and effective mass ***,we propose a one-step strategy to synthesize 3D interconnected carbon nanotubes/graphene hybrids on the easy-to-remove biomass-derived *** calcined natural cuttlebone as bi-functional catalyst precursor can simultaneously grow carbon nan-otubes and graphene by one-step chemical vapor deposition without the addition of extra metal catalysts,while the interconnected structure can act as the porous template for graphene *** simultane-ous growth process can obtain covalent bonding between carbon nanotubes and graphene,while the crystalline quality and interlayer space can be adjusted by different carbon sources and growth parame-ters(e.g.,temperature).The one-step grown carbon nanotubes/graphene hybrids with seamless interfaces and hierarchical interconnected 3D structure can effectively enhance the electron transfer as well as the electrolyte infiltration *** utilized as lithium-ion batteries(LIBs)anode,a high specific ca-pacity(544 mAh g^(-1) at 0.1 A g^(-1)),good rate capability(200 mAh g^(-1) at 6.4 A g^(-1) with an ultrashort charge time of 113 s),and excellent cyclic stability can be *** simple and one-step carbon nanotubes/graphene hybrids fabrication strategy can be easily scale-up and applied in various fields.

Monitoring physiological signals of the human body can provide extremely important information for sports healthcare,preventing injuries and providing efficient guidance for individual ***,the signals related to human healthcare involve both subtle and vigorous signals,making it difficult for a sensor to satisfy the full-scale monitoring at the same ***,a novel conductive elastomer featuring homogeneously micropyramid-structured PDMS/CNT composite is used to fabricate highperformance piezoresistive sensors by a drop-casting *** from the significant increase in the contact area of microstructure during deformation,the flexible sensor presents a broad detection range(up to 185.5 kPa),fast response/recovery time(44/13 ms),ultrahigh sensitivity(242.4 kPa–1)and excellent durability over 8,000 *** a proof of concept,the as-fabricated pressure sensor can be used for body-area sports healthcare,and enable the detection of full-scale pressure *** the fabulous sensing performance,the sensor may potentially become promising in personal sports healthcare and telemedicine monitoring.

High energy density Li-CO_(2)batteries have attracted much attention owing to the"two birds with one stone"feature in fixing greenhouse gas CO_(2)and providing renewable ***,poor reversibility of the discharge product Li_(2)CO_(3)is one of the main problems that limit its application,resulting in poor cycling stability and severe ***,copper indium sulfide(CIS),a semiconducting non-precious metal sulfide,is fabricated as cathode catalysts for high-performance Li-CO_(2)*** with the synergistic effect of bimetallic valence bonding and coordinated electron transfer,Li-CO_(2)batteries using CIS cathodes exhibit high full specific discharge capacity,excellent rate capability and cycle stability,namely it delivers a high specific full discharge capacity of 8878μAh cm^(-2),runs steadily from 10 to 100μA cm^(-2),and performs a stable long-term cycling behavior(>1050 h)under a high energy efficiency of 84%and a low charge voltage of approximately 3.4 V at 20μA cm^(-2)within 100μAh cm^(-2).In addition,a flexible Li-CO_(2)pouch cell is constructed to reveal the potential of employing CIS to fabricate flexible high energy storage devices in practical *** work shows a promising development pathway toward next-generation sustainable energy storage devices.

Analysis of rockburst mechanism and warning based on microseismic moment tensors and dynamic Bayesian networks

One of the major factors inhibiting the construction of deep underground projects is the risk posed by rockbursts.A study was conducted on the access tunnel of the Shuangjiangkou hydropower station to determine the evolutionary mechanism of microfractures within the surrounding rock mass during rockburst development and develop a rockburst warning *** study area was chosen through the combination of field studies with an analysis of the spatial and temporal distribution of microseismic(MS)*** moment tensor inversion method was adopted to study rockburst mechanism,and a dynamic Bayesian network(DBN)was applied to investigating the sensitivity of MS source parameters for rockburst warnings.A MS multivariable rockburst warning model was proposed and validated using two case *** results indicate that fractures in the surrounding rock mass during the development of strain-structure rockbursts initially show shear failure and are then followed by tensile *** effectiveness of the DBN-based rockburst warning model was demonstrated using self-validation and K-fold *** magnitude and source radius are the most sensitive factors based on an investigation of the influence on the parent and child nodes in the model,which can serve as important standards for rockburst *** proposed rockburst warning model was found to be effective when applied to two actual projects.

New insights into the DPR mechanism of elastic energy released by polymer gel for enhanced oil recovery

Polymer gel was widely used as water shutoff agent in mature oil *** the results of single-phase plugging experiments show that the plugging rate of the polymer gel to the oil phase is lower than that of the water ***,the disproportionate permeability reduction(DPR)mechanism of polymer gels still remains *** this paper,we used four gel formulations including polyethyleneimine(PEI)and phenol-formaldehyde crosslinked gel with and without adding laponite to investigate the effect of gel elastic property on the water shutoff *** result of sand pack flooding experiments shown that the gel with higher elastic modulus has better effects on decreasing water cut and increasing oil *** adding laponite,the elastic modulus of phenol-formaldehyde crosslinked gel increased from 64.2 Pa to 192 Pa,and the elastic modulus of PEI crosslinked gel increased from 27.4 Pa to 36.5 *** to the phenol-formaldehyde-HAPM gel,the oil recovery of laponite-phenol-formaldehyde-HPAM gel increased by 5.2%and the maximum water cut decreased by 8.3%.Besides,comparing with PEI-HPAM gel,the oil recovery of laponite-PEI-HPAM gel increased by 2.7%and the water cut dropped by 27.8%.In the meanwhile,the laponite-phenol-formaldehyde-HPAM gel with higher elastic modulus obviously swells in the formation water but almost remains constant in oil at 105℃.The mass of gel soaked in the formation water increased from 42 g to 96 g and the gel volume increased by 300%within 48 *** study improves the understanding of the DPR mechanism of polymer gel for water shutoff.

Stress Corrosion Cracking of High-strength Drill Pipe in Sour Gas Well

In high sour gas reservoir drilling process, it happens occasionally that high-strength drill pipe suffers brittle fracture failure due to stress corrosion cracking, and poses serious hazard to drilling safety. To solve this problem, this paper studied the stress corrosion cracking mechanism and infl uencing factors of highstrength drill pipe in sour environment with hydrogen permeation experiments and tensile tests. We simulated practical conditions in laboratory and evaluated the stress corrosion cracking performance of the high-strength drill pipe under conditions of high stress level. For the problems occurring in use of high-strength drill pipe on site, the paper proposed a technical measure for slower stress corrosion cracking.

A tactical nanomissile mobilizing antitumor immunity enables neoadjuvant chemo-immunotherapy to minimize postsurgical tumor metastasis and recurrence

Neoadjuvant chemotherapy has become an indispensable weapon against high-risk resectable cancers,which benefits from tumor ***,the utility of chemotherapeutics alone as a neoadjuvant agent is incapable of generating durable therapeutic benefits to prevent postsurgical tumor metastasis and ***,a tactical nanomissile(TALE),equipped with a guidance system(PD-L1 monoclonal antibody),ammunition(mitoxantrone,Mit),and projectile bodies(tertiary amines modified azobenzene derivatives),is designed as a neoadjuvant chemo-immunotherapy setting,which aims at targeting tumor cells,and fast-releasing Mit owing to the intracellular azoreductase,thereby inducing immunogenic tumor cells death,and forming an in situ tumor vaccine containing damage-associated molecular patterns and multiple tumor antigen epitopes to mobilize the immune *** formed in situ tumor vaccine can recruit and activate antigen-presenting cells,and ultimately increase the infiltration of CD8^(+)T cells while reversing the immunosuppression ***,this approach provokes a robust systemic immune response and immunological memory,as evidenced by preventing 83.3%of mice from postsurgical metastasis or recurrence in the B16-F10 tumor mouse ***,our results highlight the potential of TALE as a neoadjuvant chemo-immunotherapy paradigm that can not only debulk tumors but generate a long-term immunosurveillance to maximize the durable benefits of neoadjuvant chemotherapy.
