Moxibustion upregulates hippocampal progranulin expression

Acute kidney injury(AKI)is a common clinical complication associated with high mortality in *** cells and cytokines have recently been described to play essential roles in AKI *** dendritic cells(pDCs)are a unique DC subset that specializes in type Ⅰ interferon(IFN)***,we showed that pDCs rapidly infiltrated the kidney in response to AKI and contributed to kidney damage by producing IFN-α.Deletion of pDCs using DTR^(BDCA2) transgenic(Tg)mice suppressed cisplatin-induced AKI,accompanied by marked reductions in proinflammatory cytokine production,immune cell infiltration and apoptosis in the *** contrast,adoptive transfer of pDCs during AKI exacerbated kidney *** further identified IFN-α as the key factor that mediated the functions of pDCs during AKI,as IFN-α neutralization significantly attenuated kidney ***,IFN-α produced by pDCs directly induced the apoptosis of renal tubular epithelial cells(TECs)in *** addition,our data demonstrated that apoptotic TECs induced the activation of pDCs,which was inhibited in the presence of an apoptosis ***,similar deleterious effects of pDCs were observed in an ischemia reperfusion(IR)-induced AKI ***,increased expression of IFN-α in kidney biopsies was observed in kidney transplants with *** together,the results of our study reveal that pDCs play a detrimental role in AKI via IFN-α.

疼痛包括感觉分辨和情绪体验两个基本成分。对疼痛感觉分辨成分的研究,在基因、分子、细胞和系统水平已获得重要进展,但对于疼痛的情绪、情感成分的研究相对滞后。越来越多的临床观察显示,疼痛特别是慢性疼痛所伴随的负性情绪状态,严重影响患者的身心健康。本文简要总结了痛厌恶情绪研究领域的主要进展,着重阐述了前扣带皮层(anterior cingulate cortex,ACC)参与痛厌恶情绪过程的神经机制,特别是ACC神经元NMDA受体和ERK-CREB信号通路的关键性作用。多种调控分子如突触相关蛋白SIP30和雌激素可通过突触前和突触后机制调控兴奋性氨基酸释放、NMDA受体功能和ACC锥体神经元突触可塑性参与痛厌恶情绪的形成。

Selective ischemic-hemisphere targeting Ginkgolide B liposomes with improved solubility and therapeutic efficacy for cerebral ischemia-reperfusion injury

The chronic neuropathic pain-associated psychiatric disorders have seriously disturbed the quality of patients’ life, such as depression and anxiety. Neuroinflammation in hippocampus plays an important role in the neuropathic pain-associated depressive and anxiety disorders, but the underline mechanism remains unclear. NLRP1 inflammasome, which controls the production of pro-inflammatory cytokines, was broadly involved in the neuroinflammation-related diseases. In the present study, we show that the hippocampal NLRP1 inflammasome are crucial for the development of depressive behaviors after neuropathic pain. In the hippocampus, NLRP1 inflammasome is significantly activated after the rats subjected to the chronic constriction injury（CCI）-induced neuropathic pain. Inhibiting the NLRP1 inflammasome activation not only attenuated the depression-like behaviors but also significantly suppressed the production of mature IL-1β in hippocampus of CCI rats. The double-stranded RNA-dependent protein kinase（PKR, also known as EIF2 AK2）, has been recently shown to be a pivotal regulator for inflammasome activation. In the hippocampus of CCI rats, the PKR was simultaneously activated in neurons. Functional inhibition of PKR suppressed the NLRP1 inflammasome activation and effectively attenuated the CCI-induced depression-like behaviors. These results indicate that the hippocampal PKR/NLRP1 inflammasome pathway contributed to the depression-like behaviors of rats with chronic neuropathic pain. Thus, interrupting this pathway might provide a novel therapeutic strategy for neuropathic pain-associated depressive disorders.

Chinese medicine as Complementary Therapy for Female Infertility

Chinese medicine (CM) has been used in clinical treatment for thousands of years in China, Japan, Korea, and other countries. CM is at present attracting many attentions around the world for reproductive health care and disease prevention, including treatment of female infertility. This review focuses on the CM treatment for female infertility patients, and supplies a summary on the efficacy, safety, and mechanism of some Chinese herbal medicines, herbal medicine-derived active compounds, and acupuncture. A large number of researches have reported that CM could alleviate or even cure female infertility by regulating hormone, improving reproductive outcome of in vivo fertilization, affecting embryonic implantation, curing polycystic ovarian syndrome, endometriosis, pelvic inflammatory disease, relieving mental stress, and regulating immune system. Meanwhile, a few studies claimed that there was little adverse reaction of CM in randomized controlled trials. However, up to present there is a lack of adequate evidences with molecular mechanistic researches and randomized controlled trials to prove the CM as an effective and safe treatment for infertility. Thus, utility of CM as a complementary medicine will be a feasible method to improve the outcome of female infertility treatment.

A General Description for Chinese medicine in Treating Premature Ovarian Failure

Premature ovarian failure（POF） is a kind of gynecological disease that causes amenorrhea, infertility, menopause and urogenital symptoms. Currently hormone replacement therapy（HRT） is the most popular choice for women with POF to get rid of menopausal syndrome. However, as the popularization of Chinese herbs made Chinese medicine（CM） shine new lights, physicians are able to treat POF with both meno-herbs and integrated therapy. HRT has its own indications and contraindications. For example, unexplained vaginal bleeding, acute liver damage, liver dysfunction, vascular embolization, and breast cancer are all contraindications of HRT, and CM is taken by more physicians as an adjuvant therapy. This review, including a range of common Chinese herbs and formulations according to the existing literature, provides a general description of CM treating POF from the aspects of mechanisms and clinical application. It also highlights acupuncture as a unique physiotherapy for POF. Although the validity of CM has been supported by the evidence of many preclinical trials, clinical trials and meta-analysis, the adverse events with CM therapy still exist and no guarantee has been made for its safety. This review concludes the updated information for CM treating POF contributing to further studies.e
