Surveillance recommendations for gastric cancer(GC)in current guidelines focused on advanced precancerous lesions and were based on precise diagnosis of severity/extent of baseline *** aimed to develop a less endoscopy-related equipment-dependent risk-stratification tool,and assessed whether mild-precursor-lesion patients can be safely exempt from *** the multicenter communitybased cohort,75,051 participants receiving baseline endoscopy were enrolled during 2015–2017 and followed-up until *** incidence rates(CIRs)of GC for precancerous-conditions were calculated by Kaplan-Meier method and compared by Log-rank *** Cox regression models were used to detect potential factors for progression towards GC.A risk score was calculated as counts of selected *** independent cohort,including 26,586 participants was used for external *** a median follow-up of 6.25 years,CIRs of GC were 0.302%,0.436%,and 4.756%for normal group,non-neoplastic(atrophic gastritis/intestinal metaplasia)and neoplastic lesions(low-grade/high-grade dysplasia),respectively(Ptrend<0.001).Four predictors,including male,≥60 years,smoking,and limited vegetable consumption,were selected for ***-risk patients(≥3 risk factors)with non-neoplastic lesions showed higher GC risks(adjusted HR=7.73,95%CI:4.29–13.92),and their four-year CIR reached the one-year CIR of neoplastic *** categorizing non-neoplastic lesions by histological grade,both patients with moderate-to-severe lesions(aHR=3.07,95%CI:1.67–5.64)and high-risk patients with mild lesions(aHR=7.29,95%CI:3.58–14.86)showed higher *** trends were observed in validation ***-risk mild-precursor-lesion patients should receive surveillance within 3–5 years after baseline *** study provides evidence on supplementing current guideline recommendations.

Six-fold swelling expansion microscopy assisted cell painting for high-content phenotypic screening

Imaging-based phenotypic screening uses cellular phenotypes to describe the drug performance， which generally focuses on single cellular feature， lacking of a comprehensive characterization. Here， we propose a high content phenotypic screening method based on expansion microscopy (ExM) assisted Cell Painting， which enables multi-channel imaging with approximately 50 nm resolution. As a demonstration， we applied this method to a phenotypic screening involving five drugs. The morphological attributes of three subcellular structures were summarized to consist a “fingerprint” describing the drug effect. The proposed method can provide comprehensive and detailed clues for drug evaluation， enriching the content of phenotypic screening.

γ-Aminobutyric acid(GABA),a natural non-protein amino acid,plays an irreplaceable role in regulating the life activities of ***,the separation and purification of food-grade GABA from fermentation broth is still a great *** research utilized monosodium glutamate as a substrate for the production of high-purity GABA via an integrated process incorporating fermentation,purification,and ***,147 g·L^(-1) GABA with a yield of 99.8%was achieved through fed-batch fermentation by Lactobacillus brevis ***,three integrated purification methods by ethanol precipitation were compared,and crude GABA with a purity of 89,85%was obtained by the optimized ***,GABA crystals with a purity of 98.69%and a yield of 60%were further obtained through a designed crystallization ***,the GABA industrial production process model was established by Superproper Designer V10 software,and material balance and economic analysis were carried *** used in the process was recovered with a recovery of 98.79%through Aspen simulated extractive *** the fixed investment(equipment purchase and installation costs)for an annual production of 80 t GABA will be about 833000 USD;the total annual production cost(raw material cost and utility cost)will be about 641000 *** annual sale of GABA may be at the range of 2400000-4000000 USD and the payback period will be about 1-2 *** integrated process provides a potential way for the industrial-scale production of food-grade GABA.

Kinetic modeling of gamma-aminobutyric acid production by Lactobacillus brevis based on pH-dependent model and rolling correction

Gamma-aminobutyric acid(GABA)is a natural non-protein functio nal amino acid,which has potential for fermentation industrial production by Lactobacillus *** work investigated the batch fermentation process and developed a kinetic model based on substrate restrictive model established by experimental data from L25(5~6)orthogonal *** this study,the OD600 value of fermentation broth was fixed to constant after reaching its maximum because the microorganism death showed no effect on the enzyme activity of glutamate decarboxylase(GAD).As pH is one of the key parameters in fermentation process,a pH-dependent kinetic model based on radial basis function was developed to enhance the practicality of the ***,as to decrease the deviations between the simulated curves and the experimental data,the rolling correction strategy with OD600 values that was measured in real-time was introduced into this work to modify the ***,the accu racy of the rolling corrected and pH-dependent model was validated by good fitness between the simulated curves and data of the initial batch fermentation(pH 5.2).As a result,this pH-dependent kinetic model revealed that the optimal pH for biomass growth is 5.6-5.7 and for GABA production is about 5,***,the developed model is practical and convenient for the instruction of GABA fermentation production,and it has instructive significance for the industrial scale.

Immunosuppressive microenvironments present critical problems in clinical *** regulate the tumor immune microenvironment for enhancing antitumor effect,a combination of immune checkpoint inhibitors(ICIs)with chemotherapeutics has been applied *** this study,miriplatin(MiPt),the lipidic derivative of 5-fluorouracil(Fu-OA),as well as the programmed death ligand 1(PD-L1)target si RNA(siPD-L1)were integrated into Lip-Pt/Fu@siPD-L1 nanoparticles(NPs)for *** vitro results showed that Lip-Pt/Fu@siPD-L1 NPs could exhibit effective siRNA gene silencing and promote the phagocytosis of tumor cells by ***,in vivo results revealed that LipPt/Fu@siPD-L1 NPs showed significantly higher anti-tumor efficiency than that of the physical mixing of Mi Pt,5-fluorouracil,and Lip@siPD-L1 NPs(delivery of siPD-L1 by liposomes).The best anti-tumor efficiency of Lip-Pt/Fu@siPD-L1 NPs resulted from the synergistic immunotherapeutic effects of Mi Pt and siPD-L1 based on the inhibition of CD47 expression and the downregulation of PD-L1 in tumor cells,which elicited a robust anti-tumor immune response through the activation of macrophage phagocytosis and immune checkpoint *** Lip-Pt/Fu@siPD-L1 NPs provide a potential strategy for tumor chemo-immunotherapy.
