Esophageal squamous cell carcinoma（ESCC） is a poor-prognostic cancer type with extensive intra-and inter-patient heterogeneity in both genomic variations and tumor microenvironment（TME）. However, the patterns and drivers of spatial genomic and microenvironmental heterogeneity of ESCC remain largely unknown. Here, we generated a spatial multi-omic atlas by whole-exome, transcriptome, and methylome sequencing of 507 tumor samples from 103 patients. We identified a novel tumor suppressor PREX2,accounting for 22% of ESCCs with frequent somatic mutations or hyper-methylation, which promoted migration and invasion of ESCC cells in vitro. Analysis of the TME and quantification of subclonal expansion indicated that ESCCs undergo spatially directed evolution, where subclones mostly originated from the tumor center but had a biased clonal expansion to the upper direction of the ***, we found upper regions of ESCCs often underwent stronger immunoediting with increased selective fitness, suggesting more stringent immune selection. In addition, distinct TMEs were associated with variable genomic and clinical outcomes. Among them, hot TME was associated with high immune evasion and subclonal heterogeneity. We also found that immunoediting, instead of CD8+T cell abundance, acts as an independent prognostic factor of ESCCs. Importantly, we found significant heterogeneity in previously considered potential therapeutic targets, as well as BRCAness characteristics in a subset of patients, emphasizing the importance of focusing on heterogeneity in ESCC targeted ***, these findings provide novel insights into the mechanisms of the spatial evolution of ESCC and inform precision therapeutic strategies.

Any accurate simulation of regional air quality by numerical models entails accurate and up-to-date emissions data for that *** INTEX-B2006 （I06）,one of the newest emission inventories recently popularly used in China and East Asia,has been assessed using the Community Multiscale Air Quality model and observations from regional atmospheric background stations of *** of the model results with the observations for the species SO2,NO 2,O 3 and CO from the three regional atmospheric background stations of Shangdianzi,Longfengshan and Linan show that the model can basically capture the temporal characteristics of observations such as the monthly,seasonal and diurnal variance *** to the other three species,the simulated CO values were grossly underestimated by about two-third or one-half of the observed values,related to the uncertainty in CO *** to the other two stations,Shangdianzi had poorer simulations,especially for SO2 and CO,which partly resulted from the site location close to local emission sources from the Beijing area;and the regional inventory used was not capable of capturing the influencing factors of strong regional sources on ***,the fact that summer gave poor simulation,especially for SO2 and O 3,might partly relate to poor simulations of meteorological fields such as temperature and wind.

We present in-situ measurements of atmospheric sulfur hexafluoride（SF6） conducted by an automated gas chromatograph–electron capture detector system and a gas chromatography/mass spectrometry system at a regional background site, Shangdianzi,in China, from June 2009 to May 2011, using the System for Observation of Greenhouse gases in Europe and Asia and Advanced Global Atmospheric Gases Experiment（AGAGE）techniques. The mean background and polluted mixing ratios for SF6 during the study period were 7.22 × 10-12（mol/mol, hereinafter） and 8.66 × 10-12, respectively. The averaged SF6 background mixing ratios at Shangdianzi were consistent with those obtained at other AGAGE stations located at similar latitudes（Trinidad Head and Mace Head）, but larger than AGAGE stations in the Southern Hemisphere（Cape Grim and Cape Matatula）. SF6 background mixing ratios increased rapidly during our study period, with a positive growth rate at 0.30 × 10-12year-1. The peak to peak amplitude of the seasonal cycle for SF6 background conditions was 0.07 × 10-12, while the seasonal fluctuation of polluted conditions was 2.16 × 10-12. During the study period, peak values of SF6 mixing ratios occurred in autumn when local surface horizontal winds originated from W/WSW/SW/SWS/S sectors, while lower levels of SF6 mixing ratios appeared as winds originated from N/NNE/NE/ENE/E sectors.

In last few years, therapeutic peptides/proteins are rapidly growing in drug market considering their higher efficiency and lower toxicity than chemical drugs. However, the administration of therapeutic peptides/proteins is mainly limited in parenteral approach. Oral therapy which was hampered by harsh gastrointestinal environment and poorly penetrating epithelial barriers often results in low bioavailability(less than 1%–2%). Therefore, delivery systems that are rationally designed to overcome these challenges in gastrointestinal tract and ameliorate the oral bioavailability of therapeutic peptides/proteins are seriously promising. In this review, we summarized various multifunctional delivery systems, including lipid-based particles, polysaccharide-based particles, inorganic particles, and synthetic multifunctional particles that achieved effective oral delivery of therapeutic peptides/proteins.

Impact of the Vertical Velocity Field on Charging Processes and Charge Separation in a Simulated Thunderstorm

A three-dimensional（3D） charging-discharging cloud resolution model was used to investigate the impact of the vertical velocity field on the charging processes and the formation of charge structure in a strong thunderstorm. The distribution and evolution of ice particle content and charges on ice particles were analyzed in different vertical velocity fields. The results show that the ice particles in the vertical velocity range from 1 to 5 m s-1obtained the most charge through charging processes during the lifetime of the thunderstorm. The magnitude of the charges could reach 1014 n C. Before the beginning of lightning activity,the charges produced in updraft region 2（updraft speed 13 m s-1） and updraft region 1（updraft speed between 5 and 13 m s-1） were relatively significant. The magnitudes of charge reached 1013 n C, which clearly impacted upon the early lightning activity. The vertical velocity conditions in the quasi-steady region（updraft speed between -1 and 1 m s-1） were the most conducive for charge separation on ice particles on different scales. Accordingly, a net charge structure always appeared in the quasi-steady and adjacent regions. Based on the results, a conceptual model of ice particle charging, charge separation, and charge structure formation in the flow field was constructed. The model helps to explain observations of the＂lightning hole＂ phenomenon.

The rate of neutralized charge by lightning（RNCL） is an important parameter indicating the intensity of lightning *** total charging rate（CR）,the CR of one kind of polarity（e.g.,negative） charge（CROP）,and the outflow rate of charge on precipitation（ORCP） are proposed as key factors impacting RNCL,based on the principle of conservation of one kind of polarity charge in a *** this paper,the impacts of updraft on CR and CROP are analyzed by using a 3D cloud resolution model for a strong storm that occurred in Beijing on 6 September *** results show that updraft both promotes and inhibits RNCL at the same time.（1） Updraft always has a positive influence on *** correlation coefficient between the updraft volume and CR can reach *** of the updraft facilitates strengthening of RNCL through this positive influence.（2） Strengthening of the updraft also promotes reinforcement of *** correlation coefficient between the updraft volume and CROP is high（about0.9）,but this promotion restrains the strengthening of RNCL because the strengthening of CROP will,most of the time,inhibit the increasing of RNCL.（3） Additionally,increasing of ORCP depresses the strengthening of *** terms of magnitude,the peak of ORCP is equal to the peak of *** precipitation mainly appears after the lightning activity finishes,the depression effect of ORCP on RNCL can be ignored during the active lightning period.

Interaction between Colon Cancer Cells and Human Liver Sinusoidal Endothelial Cells Promotes Liver Metastasis of Tumor Cells

OBJECTIVE To investigate the effect of co-culture between colon cancer cells （SW1116） and human liver sinusoidal endothelial cells （HLSECs） on cancer cell metastasis, and to provide a novel model for studying the mechanism of colon cancer liver metastasis. METHODS HLSECs and SW1116 were co-cultured for 21 rounds in vitro. Transwell migration, gelatin-zymography, CCK-8 proliferation and colony formation assays were used to examine the invasion, proliferation, and colony forming ability of cancer cells. Assays were carried out to examine tumor growth ability and liver metastasis. The associated molecular change was examined by western blotting. RESULTS After 21 selection rounds, colon cancer cells SWl 1161）21 displayed a clear boundary. Compared with the 5W1116 cells, SW1116P21 cells had a greater invasive ability, cell proliferation and colony formation in soft agar. A gelatin-zymography assay showed that the ability of SW1116P21 cells to secrete matrix metalloproteinase-2/9 was significantly greater than that of SWl116 cells. Additionally, the capacity for subcutaneous tumor formation of SW1116P21 was significantly increased. It was found that mice injected with SW1116P21 cells developed significantly more visually observable liver nodules than mice injected with SW1116 cells. Western blotting showed increased vimentin expression and decreased E-cadherin expression in the SW1116P21 cells, compared with the SWl 116 cells. CONCLUSION The interaction between SW1116 and HLSECs may promote tumor cell invasion, proliferation and colony formation in vitro, and tumor formation and liver metastasis in vivo. An epithelial-mesenchymal transition occurs in SWl 116P21 cells, which contributes to the change in the characteristics of tumor cells.

A wide variety of algorithms have been developed to monitor aerosol burden from satellite images. Still, few solutions currently allow for real-time and efficient retrieval of aerosol optical thickness （AOT）, mainly due to the extremely large volume of computation necessary for the numeric solution of atmospheric radiative transfer equations. Taking into account the efforts to exploit the SYNergy of Terra and Aqua Modis （SYNTAM, an AOT retrieval algorithm）, we present in this paper a novel method to retrieve AOT from Moderate Resolution Imaging Spectroradiometer （MODIS） satellite images, in which the strategy of block partition and collective communication was taken, thereby maximizing load balance and reducing the overhead time during inter-processor communication. Experiments were carried out to retrieve AOT at 0.44, 0.55, and 0.67μm of MODIS/Terra and MODIS/Aqua data, using the parallel SYNTAM algorithm in the IBM System Cluster 1600 deployed at China Meteorological Administration （CMA）. Results showed that parallel implementation can greatly reduce computation time, and thus ensure high parallel efficiency. AOT derived by parallel algorithm was validated against measurements from ground-based sun-photometers; in all cases, the relative error range was within 20%, which demonstrated that the parallel algorithm was suitable for applications such as air quality monitoring and climate modeling.

Attenuated phenotypes and analysis of a herpes simplex virus 1strain with partial deletion of the UL7, UL41 and LAT genes

We previously constructed a herpes simplex virus 1(HSV-1) UL7 mutant virus(M1) and showed that a partial deletion mutation of the UL7 gene led to a lower proliferative rate and an attenuated phenotype. Using the M1 mutant, we further modified the UL41 gene, which encodes another tegument protein, and the latency-associated transcript(LAT) gene. Observations of the resulting mutants with modified UL7 and UL41(M2) or UL7, UL41 and LAT(M3) genes indicated attenuated phenotypes, with lower proliferative ratios in various cells, non-lethal infections in mice and lower viral loads in nervous tissues compared with the wild-type strain. Furthermore, no LAT stable intron could be detected in the trigeminal ganglion of M3-infected animals. The results obtained with the three HSV-1 mutants indicate that the M3 mutant is an attenuated strain with low pathogenicity during both acute and latent infections. Together, the results support the use of the M3 mutant as a candidate for the development of an HSV-1 vaccine.
