Mechanism by which Rab5 promotes regeneration and functional recovery of zebrafish Mauthner axons

Rab5 is a GTPase protein that is involved in intracellular membrane trafficking. It functions by binding to various effector proteins and regulating cellular responses, including the formation of transport vesicles and their fusion with the cellular membrane. Rab5 has been reported to play an important role in the development of the zebrafish embryo;however, its role in axonal regeneration in the central nervous system remains unclear. In this study, we established a zebrafish Mauthner cell model of axonal injury using single-cell electroporation and two-photon axotomy techniques. We found that overexpression of Rab5 in single Mauthner cells promoted marked axonal regeneration and increased the number of intra-axonal transport vesicles. In contrast, treatment of zebrafish larvae with the Rab kinase inhibitor CID-1067700markedly inhibited axonal regeneration in Mauthner cells. We also found that Rab5 activated phosphatidylinositol 3-kinase(PI3K) during axonal repair of Mauthner cells and promoted the recovery of zebrafish locomotor function. Additionally, rapamycin, an inhibitor of the mechanistic target of rapamycin downstream of PI3K, markedly hindered axonal regeneration. These findings suggest that Rab5 promotes the axonal regeneration of injured zebrafish Mauthner cells by activating the PI3K signaling pathway.

Catalyst–support interaction plays a crucial role in improving the catalytic activity of oxygen evolution reaction(OER).Here we modulate the catalyst–support interaction in polyaniline-supported Ni_(3)Fe oxide(Ni_(3)Fe oxide/PANI)with a robust hetero-interface,which significantly improves oxygen evolution activities with an overpotential of 270 mV at 10 mA cm^(-2)and specific activity of 2.08 mA cm_(ECSA)^(-2)at overpotential of 300 mV,3.84-fold that of Ni_(3)Fe *** is revealed that the catalyst–support interaction between Ni_(3)Fe oxide and PANI support enhances the Ni–O covalency via the interfacial Ni–N bond,thus promoting the charge and mass transfer on Ni_(3)Fe *** the excellent activity and stability,rechargeable Zn-air batteries with optimum Ni_(3)Fe oxide/PANI are assembled,delivering a low charge voltage of 1.95 V to cycle for 400 h at 10 mA cm^(-2).The regulation of the effect of catalyst–support interaction on catalytic activity provides new possibilities for the future design of highly efficient OER catalysts.

We present the AS-Index, a new index structure for exact string search in disk resident databases. AS-Index relies on a classical inverted file structure, whose main innovation is a probabilistic search based on the properties of algebraic signatures used for both n-grams hashing and pattern search. Specifically, the properties of our signatures allow to carry out a search by inspecting only two of the posting lists. The algorithm thus enjoys the unique feature of requiring a constant number of disk accesses, independently from both the pattern size and the database size. We conduct extensive experiments on large datasets to evaluate our index behavior. They confirm that it steadily provides a search performance proportional to the two disk accesses necessary to obtain the posting lists. This makes our structure a choice of interest for the class of applications that require very fast lookups in large textual databases. We describe the index structure, our use of algebraic signatures, and the search algorithm. We discuss the operational trade-offs based on the parameters that affect the behavior of our structure, and present the theoretical and experimental performance analysis. We next compare the AS-Index with the state-of-the-art alternatives and show that 1） its construction time matches that of its competitors, due to the similarity of structures, 2） as for search time, it constantly outperforms the standard approach, thanks to the economical access to data complemented by signature calculations, which is at the core of our search method.

Given a set of signals, a classical construction of an optimal truncatable basis for optimally representing the signals, is the principal component analysis(PCA for short)approach. When the information about the signals one would like to represent is a more general property, like smoothness, a different basis should be considered. One example is the Fourier basis which is optimal for representation smooth functions sampled on regular grid. It is derived as the eigenfunctions of the circulant Laplacian operator. In this paper,based on the optimality of the eigenfunctions of the Laplace-Beltrami operator(LBO for short), the construction of PCA for geometric structures is regularized. By assuming smoothness of a given data, one could exploit the intrinsic geometric structure to regularize the construction of a basis by which the observed data is represented. The LBO can be decomposed to provide a representation space optimized for both internal structure and external observations. The proposed model takes the best from both the intrinsic and the extrinsic structures of the data and provides an optimal smooth representation of shapes and forms.

The structural, elastic, and thermodynamic properties of cubic-Fe2TiA1 under high temperatures and pressures are investigated by performing ab initio calculation and using the quasi-harmonic Debye model. Some ground state properties such as lattice constant, bulk modulus, pressure derivative of the bulk modulus, and elastic constants are in good agreement with the available experimental results and theoretical data. The thermodynamic properties of Fe2TiA1 such as thermal expansion coefficient, Debye temperature, and heat capacity in ranges of 0 K-1200 K and 0 GPa-250 GPa are also obtained. The calculation results indicate that the heat capacities at different pressures all increase with temperature increasing and are close to the Dulong-Petit limit at higher temperatures, Debye temperature decreases with temperature increasing, and increases with pressure rising. The cubic-FezTiA1 is stable mechanically under 250 GPa. Moreover, under lower pressure, thermal expansion coefficient rises rapidly with temperature increasing, and the increasing rate becomes slow at higher pressure.

Hierarchical mechanical behavior of cobalt supracrystals related to nanocrystallinity

Here, we report on hierarchical mechanical behavior of 500-nm-thick Conanocrystal 3D superlattices （supracrystals） induced by either the crystalline structure （nanocrystallinity） or the length of the coating agent of Co nanocrystals. Increasing the nanocrystal shape anisotropy of Co nanocrystals through the control of their nanocrystallinities induces a higher level of ordering with both translational and orientational alignment of nanocrystals within the supracrystals. The hierarchy in ordering at various scales, i.e., from the atomic lattice within the nanocrystals to the nanocrystal superlattices within supracrystals, is correlated with marked changes in the Young＇s modulus of supracrystals： From 0.7 ±0.4 to 1.7 ±0.5 and to 6.6 ±1.5 GPa as the crystalline structure of Co nanoparticles changes from amorphous-Co to ε-Co and to hexagonal compact packing （hcp）-Co, respectively. Moreover, for supracrystals of 7 nm amorphous Co nanoparticles, the Young＇s modulus decreases by one order of magnitude from 0.7 ±0.4 to 0.08 ±0.03 GPa upon reducing the alkyl chain length of the ligands coating the Co nanoparticles from C18 （oleic acid） to C12 （lauric acid）. The hierarchical mechanical behavior is rationalized using a dimensional model of the stress-strain relationship in supracrystals.
