Aim:To investigate the apoptosis-inducing effect of oridonin,a diterpenoid iso-lated from Rabdosia rubescens,in the human cervical carcinoma HeLa cell ***:A morphological analysis,nuclear condensation,and fragmentation ofchromatin were monitored using Hoechst 33342 *** viability was as-sessed using the 3-(4,5-dimethylthiazol-(2)-yl)-2,5-diphenyl tetrazolium bromide(MTT) *** apoptosis and the apoptosis-related activation in the HeLa cellline were evaluated by flow cytometry and Western ***:Oridoninsuppressed the proliferation of the HeLa cell line in a dose-and *** treatment downregulated the activation of protein kinase B(Akt),the expression of forkhead box class O (FOXO) transcription factor,andglycogen synthase kinase 3 (GSK3).Oridonin also induced the release of cyto-chrome c accompanied by the activation of caspase-3 and poly-adenosine diphos-phate-ribose polymerase *** addition,Z-D(OMe)-E(OMe)-V-D(OMe)-FMK (z-DEVD-fmk),an inhibitor of caspases,prevented caspase-3 activation andabrogated oridonin-induced cell ***,oridonin treatment of the HeLa cellline downregulated the expression of the inhibitor of the apoptosis ***:Our results showed that oridonin-induced apoptosis involved sev-eral molecular *** may suppress constitutively activated targetsof phosphatidylinositol 3-kinase (Akt,FOXO,and GSK3) in the HeLa cell line,inhibiting the proliferation and induction of caspase-dependent apoptosis.

Glycogen synthase kinase-3β positively regulates the proliferation of human ovarian cancer cells

Although glycogen synthase kinase-3(GSK-3)might act as a tumor suppressor since its inhibition is expected tomimic the activation of Wnt-signaling pathway,GSK-3β may contribute to NF-κ3 activation in cancer cells leading toincreased cancer cell proliferation and *** we report that GSK-3β activity was involved in the proliferationof human ovarian cancer cell both in vitro and in *** of GSK-3 activity by pharmacological inhibitors sup-pressed proliferation of the ovarian cancer *** constitutively active form of GSK-3β induced entryinto the S phase,increased cyclin D1 expression and facilitated the proliferation of ovarian cancer ***,GSK-3 inhibition prevented the formation of the tumor in nude mice generated by the inoculation of human ovariancancer *** findings thus suggest that GSK-3β activity is important for the proliferation of ovarian cancer cells,implicating this kinase as a potential therapeutic target in ovarian cancer.

Effect of IL-1βand TNF-αon the Expression of MonocyteChem otactic Protein-1 in Endom etriotic Cells

To investigate the clinical significance of monocyte chemotactic protein 1 (MCP 1) produced by endometriotic tissues, the endometriotic tissues were taken from 15 patients with endometriosis. MCP 1 mRNA and MCP 1 protein were determined by dot blot analysis and enzyme linked immunosorbent assay (ELISA) in endometriotic cells cultured with or without interleukin 1β (IL 1β, 2 μg/L), tumor necrosis factor α (TNF α, 20 g/L). After exposure to IL 1β or TNF α, the expression of MCP 1 mRNA in the endometriotic cells (8.635± 0.826, 7.031±0.970, respectively) were significantly higher than that in the control group (4.482±0.435, P cantly higher than 1.74±0.16 μg/L in control ( P <0.01). The results suggested that IL 1β and TNF α could up regulate the expression of MCP 1 in endometriotic cells, which might be related to the development of endometriosis.

To explore the roles of mitochondria tRNA leu(UUR) gene mutation at nucleotide 3243 and the activity of cytochrome c oxidase in pathogenesis of preeclampsia, 57 patients with preeclampsia and 60 normotension pregnancy women were screened for tRNA leu(UUR) nt3243 A→G mutation with the method of polymers chain reaction (PCR) and restriction fragment length polymorphism. Cytochrome c oxidase activity was determined by measuring the rate of cyanide sensitive oxidation of reduced cytochrome c using luminosity photographer. The results showed that cytochrome c oxidase activity was significantly lower in the preeclampsia group (0.30±0.39/min, n = 32) than that in the controls (0.73±0.54/min, n = 26, P <0.01). The mitochondria DNA mutation at position 3243 was not found in our series. The results suggested that the decreased activity of cytochrome c oxidase might impair the energy production, leading to the mitochondria dysfunction and placenta dysfunction in preeclampsia patients. Mitochondria dysfunction may be involved in the pathogenesis of preeclampsia. The mutation of mitochondria DNA may not be the common contributor of preeclampsia in our series.

PCR enzyme linked immune sorbent assay (ELISA) was developed for detection of RT PCR amplified cytokeratin 19 (CK19) mRNA and carcinoembryonic antigen (CEA) mRNA. The non radioactive hybridization was performed in a streptavidin coated microwell with digoxigenin labeled PCR products and with biotin labeled capture probe. PCR ELISA was proved to be expedient, simple, sensitive and safe for identification of CK19 , CEA RT PCR products. These results were proven by sequencing.

Association of the MTHFR C677T polymorphism and bone mineral density in postmenopausal women:a meta-analysis

Osteoporosis is a condition characterized by low bone mineral density （BMD） and micro-architectural changes in the bone *** risk of osteoporosis is partly determined by genetic *** role of C677T polymorphism of methylenetetrahydrofolate reductase （MTHFR） gene has been investigated in postmenopausal ***,the relationship between MTHFR polymorphism and BMD is still *** carried out a meta-analysis of 5,833 subjects to evaluate the association of MTHFR and BMD in postmenopausal *** of MEDLINE,Web of Science,Scopus and CNKI were retrieved for all publications relating to MTHFR polymorphism and BMD in postmenopausal *** eligible studies were selected for *** these articles studied the association of MTHFR polymorphism and BMD of the femoral neck and lumbar spine in postmenopausal *** analysis suggested that postmenopausal women with the TT genotype had lower femoral neck BMD than the women with the CC/CT genotype,and the weighted mean difference （WMD） was-0.01 g/cm 2 [95% confidence interval （CI）：（-0.01,-0.01）,P 0.01].However,BMD of the lumbar spine of postmenopausal women with the TT genotype was not significantly different from that of women with the CC/CT *** the random effects model,the WMD between the TT and TC/CC genotype was-0.01 g/cm 2 [95% CI：（-0.04,0.01）,P=0.32].The C677T polymorphism of the MTHFR gene is associated with BMD of the femoral neck in postmenopausal *** with the TT genotype of the MTHFR gene have lower BMD,suggesting that the TT genotype may be a risk factor for postmenopausal osteoporosis.

Increased Connexin 43 Expression Improves the Migratory and Proliferative Ability of H9c2 Cells by Wnt-3a Overexpression

The change of connexin 43 （Cx43） expression and the biological behaviors of Cx43 in rat heart cell line H9c2, expressing Wnt-3a （wingless-type MMTV integration site family, member 3A） were evaluated in the present study. Plasmid pcDNA3.1/Wnt-3a was constructed and transferred into H9c2 cells. The cell model Wnt-3a+-H9c2 steadily expressing Wnt-3a was obtained. Compared with H9c2 and pcDNA3.1-H9c2 cells, the expression of Cx43 in Wnt-3a+-H9c2 cells was clearly increased, the proliferation of Wnt-3a+-H9c2 cells was significantly changed, and cell migration abilities were also improved （P<0.05）. In comparison with H9c2 and pcDNA3.1-H9c2 cells, the G2phase of the cell cycle increased by 11% in Wnt-3a+-H9c2 cells. Thus, Wnt-3a overexpression is associated with an increase in Cx43 expression and altered migratory and proliferative activity in H9c2 cells. Cx43 might be one of the downstream target genes regulated by Wnt-3a.

Changes of Cytokines Levels in Peritoneal Fluids of Patients with Endometriosis and Its Effect on Reproductive Activity

To study the changes of cytokines in peritoneal fluids of patients with endometriosis and their effects on reproductive activity, levels of tumor necrosis factor (TNF) and interleukin 6 (IL-6) in peritoneal fluids and peritoneal macrophages' culture supernatant were studied by using enzyme linked immunoassay (ELISA) in 14 infertile patients with endometriosis (EMT group) and 11 infertile women with normal pelvis (control group). The effects of peritoneal fluids in patients with endometriosis in vitro on sperm motility and development of 2-cell mouse embryos were also studied. The results showed that the levels of TNF and IL-6 in peritoneal fluids and peritoneal macrophages' supernatant in EMT group were elevated significantly as cohipared with thai in the control group (P cantly in EMT group (P6-cells was 32. 5 % in EMT group, while 47. 6 % in control group after 48 h co-culture with peritoneal fluid (P <0. 01). It is likely that the elevation of peritoneal fluid cytokines in patients with endometriosis may play a role in infertility associated with endometriosis.
