Aim:To determine the inhibitory effect of the synthetic STAT3.siRNA on theexpression of STAT3.gene in human laryngeal cancer cell lines Hep2 and to inves-tigate the effect of STAT3.siRNA on growth and apoptosis in Hep2 ***:A pair of DNA templates coding siRNA against STAT3.mRNA was synthesized toreconstruct plasmid of pSilencer1.0-U6 ***2 cells were trans-fected with RPMI-1640 media (untreated),plasmid (empty),and STAT3.siRNA,*** blot and Western blot analysis of STAT3.and pTyr-STAT3.xpression in Hep2 cells and Western blot analysis of Bcl-2 expression in the Hep2cell was performed 72 h after ***,flow cytometry,and AO/EBassay were used for determination of cells proliferation and apoptosis in ***:pTyr-STAT3.was markedly expressed in untreated Hep2 cells andthe vector-treated Hep2 cells,whereas pTyr-STAT3.expression was significantlyreduced in STAT3.siRNA-transfected Hep2 cells,indicating that STAT3.siRNAinhibited the activity of *** of Hep2 cells with STAT3.siRNAsignificantly inhibited STAT3.expression at both mRNA and protein level in Hep2cells and the inhibition was characterized by time-dependent ***-ment of Hep2 cells with STAT3.siRNA resulted in dose-dependent growth inhibi-tion of Hep2,this significantly increased apoptotic cell rate,and decreased Bcl-2expression level in Hep2 ***3.siRNA had an effect on induction of eitherearly or late stage ***:This study demonstrates that STAT3.iRNA effectively inhibits STAT3.gene expression in Hep2 cells leading to growthsuppression and induction of apoptosis in Hep2 *** use of siRNA tech-nique may provide a novel therapeutic approach to treat laryngeal cancer andother malignant tumors expressing constitutively activated STAT3.

Functions and call relations are extracted into nodes and edges,respectively,by which a novel topological model is *** directional edges and the corresponding weight values express the call relations and tightness *** introducing two concepts of function fault-tolerant capability and software fault intensity and by designing a allocation rule on fault-tolerant capability,a cascading fault model is built to explore fault propagation *** on practical software networks show that a weak fault intensity,a small number of initial faults,and a strong fault-tolerant capability can slow down the spreading *** functions with more call relations and more closer tightness contribute more to the stability of the whole system.

The optimized molecular structures and vibrational frequencies of 4-mercaptopyridine(4MPY) monomer and its dihydrate were studied by means of the density functional theory(DFT), viz. B3.YP method with the 6-3.1++G(d,p) basis set. On the basis of the calculations, the assignments of the vibrational spectra of the monomer and the dihydrate were performed, and so were investigated the changes in the structure and the vibrational spectrum of the dihydrate as well as the intermolecular force resulting in the formation of the dihydrate. The calculated results show that each of the water molecule planes is vertical to the pyridine ring plane in the dihydrate that is formed via the H-bonds between 4MPY and water molecules. Furthermore, the structure and the vibrational spectrum of 4MPY can be consi- derably affected by the water molecules.

Roles of Salicylic Acid-responsive Cis-acting Elements and W-boxes in Salicylic Acid Induction of VCH3.Promoter in Transgenic Tobaccos

A salicylic acid (SA)-inducible VCH3.promoter was recently identified from grapevine (Vitisarnurensis) that contains two inverse SA-responsive cis-acting elements and four *** furtherdemonstrate the roles of these elements,four fragments with lengths from-1187,-892,-589,-276 to+7 bp were fused with the β-glucuronidase (GUS) reporter géne and transferred to Nicotiana tobacum,together with another four VCH3.promoter fragments with mutation in the two inverse *** functions,of each promoter fragment were-examined by analysis of GUS activity in thetransgenic tobacco root treated with *** GUS activity was shown in the roots of transgenictobaccos with the VCH3.(-1187)-GUS construct containing two SA-responsive cis-acting elements andfour ***,GUS activity directed by the VCH3.(-892)-GUS construct,containing one SA cis-acting element and four W-boxes,was reduced by up to 3.% compared with that in tobaccos transformedwith the VCH3.(-1187)-GUS construct,indicating that the SA cis-acting element plays an important role inSA induction of the VCH3.*** the m2VCH3.(-1187)-GUS nor the mVCH3.(-892)-GUSconstruct,with mutation on the SA-responsive elements,abolished the expression of GUS activity,demon-strating that the W-boxes in the VCH3.promoter are also involved in SA *** arialysis ofGUS activity directed by each of the eight VCH3.promoter fragments showed that GUS was expressedspecifically in vascular *** was concluded that both the SA-responsive cis-acting elements and the W-boxes are important for the SA induction of the VCH3.*** promoter might have a potential usein plant genetic engineering.

Safe and Objective Assay of Enterovirus 71 Neutralizing Antibodies via Pseudovirus

Current serum neutralization assays based on the inhibition of the cytopathic effect（Nt-CPE） need to ma nipulate live viruses, which are time-consuming, labor-intensive, and have the potential exposure to infectious agents, so a safe and objective assay via pseudovirus for the fast and efficient detection of enterovirus 71（EV71） neutralizing antibodies was developed. First, we generated EV71 pseudovirus containing firefly luciferase gene in place of the capsid gene P1 in EV71 genome. Vero cells infected with 200 CCID50（50% cell culture infective dose） of EV71 pseudovirus for 24 h were found to have the best performance. Seval sera were measured by EV71 pseudoparticle neutralization assay（Nt-PPN） and the conventional serological method Nt-CPE. Neutralizing antibody titers measured by Nt-PPN and those obtained by Nt-CPE demonstrate a high correlation between the two methods. Overall, the PPN assay represents a valid alternative to conventional serological methods for the evaluation of EV71 neutralizing anti bodies. This method can be used for detecting neutralizing antibodies of other picornaviruses, such as hepatitis A vi rus（HAV） and coxsackievirus 16（CVA16）, and make it possible to determine whether there is cross-reactivity be tween EV71 and CVA16.

BACKGROUND: Activation of N-methyl-D-aspartate receptor (NMDAR) is a key link of exitotoxicity at the phase of cerebral ischemic injury. Because NMDAR is a main way to mediate internal flow of Ca2+ among glutamic acid receptors, over-excitation can cause neuronal apoptosis. Calcitonin gene related peptide has a strongly biological activity. On one hand, it can protect ischemic neurons through inhibiting the expression of NMDAR1 mRNA; on the other hand, it can play the protective effect through down-regulating the expression of NMDAR1 mRNA by exogenous calcitonin gene related peptide. OBJECTIVE: To observe the expression of NMDAR1 and the regulatory effect of calcitonin gene related peptide on the expression of NMDAR1 mRNA and protein in the cerebral cortex of rats with focal cerebral ischemia/reperfusion (I/R). DESIGN: Randomized controlled animal study. SETTING: china medical university. MATERIALS: A total of 216 healthy male Wistar rats, general grade, weighing 250-280 g, were selected in this study. Twelve rats were randomly selected to regard as control group; meanwhile, other 204 rats were used to establish middle cerebral artery occlusion/reperfusion (MACO) models. The main reagents were detailed as follows: calcitonin gene related peptide (Sigma Company); calcitonin gene related peptide kit (Boster Company); antibody Ⅰ, Ⅱ and antibody β-actin Ⅰ, Ⅱ of NMDAR1 mRNA and chemiluminescence reagent (Santa Cruz Company, USA). METHODS: The experiment was carried out in the Laboratory of Neurobiology of china medical university from August 2005 to June 2006. ① Right MCAO models of rats were established to cause focal ischemia and scored based on Zea Longa five-grade scale. If the scores were 1, 2 and 3.after wakefulness, the MACO models were established successfully and involved in the experiment.
