Th1-response plays a crucial role in determining pathogenesis of organ-specific autoimmune diseases. It is believed that both IL-12 and INF-a are initiators to regulate Th1-response. In our experimental autoimmune uveitis (EAU) model, both Lewis and Fischer 34. rats share the same MHC class II molecules, while Lewis rat is EAU susceptible and Fischer 34. rat is EAU resistant. However, under the same condition of immunization, if pertussis toxin (PTX) was injected intraperitoneally as an additional adjuvant, Fischer 34. rat can develop EAU. In this study we investigate which mechanisms are involved in the induction of EAU in CFA + R16 + PTX treated (CRP- treated) Fischer 34. rats. In vivo and in vitro data demonstrated that Th1-cytokine, IFN- mRNA expression was significantly increased in disease target tissue-eyes and in draining lymph node cells of CRP-treated Fischer 34. rat. When IL-12 and IFN-a mRNA expression were compared in the experimental groups, only IFN-a mRNA expression was associated with EAU development. To distinguish the sources of IFN-a producing cells, it was observed that IFN-a expression was mainly produced by macrophages. It was further confirmed that normal macrophage from Fischer 34. rat was able to produce significant IFN-a in the presence of PTX. The data strongly suggested that IFN-a might be involved in initiating Th1-cell differentiation and in turn contribute to the induction of EAU. High IFN-a expression induced by PTX may represent a novel pathway to initiate Th1 response in Fischer 34. rat.

Our previous work showed that the cartilage proteoglycan aggrecan could induce an erosive polyarthritis and spondylitis in BALB/c mice and the GI globular domain of the aggrecan (GI) contained the arthritogenic region. To elucidate whether autoreactive T cells to G1 are expressed in rheumatoid arthritis patients, we analyzed the frequency of human G1-specific T cells in the peripheral blood of five rheumatoid arthritis patients and tried to establish G1-reactive T cell lines from these rheumatoid arthritis patients. The results showed that the G1-specific T cells in PBL were detectable at the range of 4.97 ±0.5 ×10-6 in peripheral blood lymphocytes. We have also generated 15 G1-specific T lymphocyte lines from these pateints with a standard split-well method. All these cells expressed fine specificity to human recombinant G1, but not to unrelated antigen. All the 15 lines expressed a panT cell marker and 13 of them selectively used the αβ T cell receptor. Two of them used rye T cell receptor. The 13 of these T cell lines was CD4.positive. One line expressed CD8. One line expressed both CD4.and CD8. Moreover, 14.out of 15 lines expressed the Th-1 cytokine profile, characterized by interferon-γpositivity and IL-4. negativity. No Th-2 type cell line was generated. These data provide strong evidence in favor of the presence of autoreactive T cells in the rheumatoid arthritis pateints. What is the mechanism(s) that these autoreactive T cells attack self-target and whether these G1-specific, Th-1 type T cell lines can induce arthritis in immune deficiency mice are currently under investigation.

The fully discrete lumped mass finite element method is proposed for vibration analysis of elastic plate-plate *** the space directions,the longitudinal displacements on plates are discretized by conforming linear elements,and the transverse displacements are discretized by the Morley *** means of the second order central difference for discretizing the time derivative and the technique of lumped masses,a fully discrete lumped mass finite element method is obtained,and two approaches to choosing the initial functions are also *** error analysis for the method in the energy norm is established,and some numerical examples are included to validate the theoretical analysis.

A time delay equation for sea-air oscillator model is studied. The aim is to create an asymtotic solving method of nonlinear equation for the ENSO model. And based on a class of oscillators of ENSO model, employing the variational iteration method, the approximate solution of corresponding problem is obtained. It is proven from the results that the method of variational iteration method can be used for analyzing the sea surface temperature anomaly in the equatorial eastern Pacific of the atmosphere-ocean oscillation for ENSO model.

A finite element method is introduced to solve the general elastic multi-structure problem, in which the displacements on bodies, the longitudinal displacements on plates and the longitudinal displacements on beams are discretized using conforming linear elements, the rotational angles on beams are discretized using conforming elements of second order, the transverse displacements on plates and beams are discretized by the Morley elements and the Hermite elements of third order, respectively. The generalized Korn's inequality is established on related nonconforming element spaces, which implies the unique solvability of the finite element method. Finally, the optimal error estimate in the energy norm is derived for the method.

To lay background for studying rejection mechanisms in xenotransplantation and developing the strategies for intervention, class I genes of swine leukocyte antigens (SLA) of three Chinese pig strains Bm, Gz and Yn were cloned and sequenced. The cDNA of the class I loci P1 and P14.were amplified by RT-PCR and subjected to insert into sequencing vectors. All six allelic sequences we examined, each two for one Chinese strain, are not identical to those reported, which allows these novel sequences receiving their accession numbers AY1024.7- AY1024.2 from GenBank. This study further reveals that the homologies of MHC class I genes in their primary structures and the deduced amino acids between Chinese pigs (SLA) and human (HLA-A*0201) are better than those between pigs and mice (H-2Db/H-2Kb). The comparison also indicates that the amino acid residues critical for recognition by human KIRs are altered in the swine class I molecules. A monoclonal antibody, LC-1, recognizing lung cancer associated common antigens was obtained in authors’ laboratory. Its single chain Fv fragment (ScFv) named LC-1 ScFv was constructed based on recombinant phage displayed techniques. For expression on cell membrane, LC-1 ScFv was cloned into pDisplay vector, which directed the cloned gene to express as cell membrane bound protein. The resulting plasmid was sequenced and then introduced by the lipofectin method into a lung adenocarcinoma cell line SPC-A-1. G4.8 resistant cells were obtained by G4.8 selection. After transfection, LC-1 ScFv expression was observed by Western blot analysis and the expression of cognate antigens was down-regulated as shown in ELISA assay. SPC-A-1-pDisplay-ScFv cells grew in vitro at lower speed than the control intact cells and the cells transfected with vacant vector. Flow cytometry analysis detected a substantial increase in G1 phase and decrease in S phase in population of SPC-A-1-pDisplay-ScFv cells compared to SPC-A -1 and SPC-A1-pDisplay cells. Semi-quantitative RT-PCR analysis showed that c-myc expression was down-regulated in SPC-A-1-pDisplay-ScFv cells. It seems that the antigens recognized by LC-1 may be in some way involved in a growth stimulating pathway and the antibody blocking of the function of the antigens shut down the pathway and thus down-regulate the expression of c-myc and growth of the cells.

The sympathetic nervous system plays a cardinal role in regulating cardiac function through releasing the neurotransmitter norepinephrine (NE). In comparison with ceniral nervous system, the molecular mechanism of NE uptake in myocardium is not clear. In present study we proved that in rat the CNS type of NE transporter (NET) was also expressed in middle cervical-stellate ganglion complex (MC-SG complex) which is considered to control the activity of heart, but not expressed in myocardium. The results also showed that NET expression level in right ganglion was significantly higher than in the left, rendering the greater capacity of NE uptake in right ventricle, a fact which may contribute to the maintenance of right ventricular function under pathologic state.
