Taxol inhibits stretch-induced electrophysiological alterations in isolated rat hearts with acute myocardial infarction

Mechanosensitive channels have been determined to work as transducers of mechanoelectric feedback in the heart, which is associated with the generation of arrhythmias. Recent studies have investigated the role of the cytoskeleton in ion channels control. This study explored the ability of taxol to inhibit stretch-induced electrophysiological alterations in the ischemic myocardium. Thirty-two Wistar rats were randomly divided into four groups: normal control group (n=9), taxol group (n=7), myocardial infarction (MI) group (n=9), and MI+taxol group (n=7). After Langendorff perfusion, the isolated hearts were stretched for 5 s by balloon inflation to 0.2 or 0.3 mL. The effects of stretching on 90% monophasic action potential duration (MAPD90), premature ventricular beats (PVB), and ventricular tachycardia (VT) were observed for 30 s. Stretching increased MAPD90 in both the normal control and MI groups, but MAPD90 increased more in the MI group for the same degree of stretch. Taxol (5 μmol L?1) had no effect on MAPD90 under baseline, unstretched conditions, but MAPD90 in the taxol group was slightly increased after stretching compared with the normal control group (P>0.05). However, taxol reduced MAPD90 in infarcted myocardium (P<0.05 at V=0.3 mL). The incidences of PVB and VT in the MI group were higher than in the normal control group (both P<0.01). Taxol had no effect on the occurrence of arrhythmias in normal myocardium, but it inhibited PVB and VT in infarcted hearts (both P<0.01). Thus changes in MAPD and the occurrence of arrhythmias caused by mechanical stretching of the myocardium could be inhibited by taxol in isolated rat hearts during AMI, indicating the involvement of tubulin in mechanoelectric feedback in AMI.

BACKGROUND: Delayed vitamin K deficiency is characterized by acute onset, severe illness and high fatality rate. 33%-50% survivors accompany with other various nervous system sequelas. Therefore, diagnosis and treatment of intracranial hemorrhage in time become a key factor for improving healing rate and reducing fatality rate and incidence of sequela. OBJECTIVE: To investigate the clinical situation, experimental characteristics, CT examination and terminative characteristics of infants with intracranial hemorrhage induced by delayed vitamin K deficiency. DESIGN: Case analysis. SETTING: department of Pediatrics, Zhongshan hospital affiliated to Xiamen university. PARTICIPANTS: A total of 17 infants with intracranial hemorrhage induced by delayed vitamin K deficiency aged 1-3 months including 11 boys and 6 girls were selected from department of Pediatrics, Zhongshan hospital affiliated to Xiamen university from January 1994 to December 2005. All infants had drowsiness, rejective milk, spiting milk, gaze of both eyes, tic, coma, full anterior fontanelle, high muscular tension and cerebral hernia, etc. Experimental examination demonstrated that infants had anemia at various degrees; prothrombin time and partial thromboplastin time were prolonged; platelet count was normal. CT examination indicated that screenages of subarachnoid hemorrhage, subdural hematoma, cerebral parenchyma hemorrhage and intraventricular hemorrage were changed. Hemorrhage was stopped by the application of vitamin K. All patients provided informed consent. METHODS: ① Clinical situation and physical sign of infants were observed after hospitalization and scanned with rapid spiral CT scanning system. The thickness and average space of layers were 8-10 mm and the scanning time was 5 s with window width of 30-80 Hu and window position of 28-35 Hu. ② After hospitalization, four items of blood coagulation was measured with Futura meter and biochemical indexes of blood, such as serum calcium, serum alka

BACKGROUND:Clinically,common bile duct (CBD) stones >2 cm are difficult to remove by endoscopic retrograde cholangiopancreatography (ERCP).To evaluate this observation,the rates of successful clearance of CBD stones and complications were compared between ERCP extraction of CBD stones of >2 cm and hospital from May 2004 to May 2008 were *** with CBD stones of >2 cm in diameter were enrolled in the >2 cm *** matched controls with CBD stones of 2 cm group and 144 patients were in the 2 cm group (77.8% and 58.3%,respectively) than in the 2 cm group was higher than in the 2 cm group than in the 2 cm in diameter was lower but the complication rate was higher than that of stones of <2 cm in diameter.

The molecular mechanism underlying angiogenesis in hepatocellular carcinoma: the imbalance activation of signaling pathways

OBJECTIVE: To explore the effect of two dominating signaling pathways, VEGF/KDR and angiopoietins/Tie2, on the formation of new blood vessel in hepatocellular carcinoma (HCC) growth and metastasis. METHODS: RT-PCR and Western blot were employed to evaluate the VEGF/KDR and angiopoietins/Tie2 expression in samples from 23 patients with HCC. Meanwhile, microvessel density (MVD) was determined as a marker of angiogenesis by counting CD34 positive cells with the method of immunohistochemistry. RESULTS: The two pathways were activated in all HCC samples. The expressions of vascular endothelial growth factor (VEGF) and angiopoietin-2 (Ang2) were significantly higher (P4 positive cells. Although significant difference in the expression of kinase insert domain containing receptor (KDR) and Ang1/Tie2 was not observed in all groups, their distinct high levels were seen in hepatoma and its margin compared with normal and cirrhotic liver. VEGF and Ang2 expressions were seen up-regulated in HCC with vascular invasion and satellite lesion. CONCLUSIONS: The two signaling pathways, VEGF/KDR and angiopoietins/Tie2 are activated in the process of angiogenesis in HCC and modulate the formation of new blood vessels. The imparity of the two signaling pathways' activation is to benefit HCC metastasis. In the two pathways, VEGF and Ang2 may play an important role in the process of angiogenesis, and are necessary indicators for the prognosis and metastasis of HCC. This study provides another clue for the exploration of anti-angiogenic agents.

EGFR Inhibitor Enhances Cisplatin Sensitivity of Human Glioma Cells

Epidermal growth factor receptor （EGFR） is fotmd to express at high levels in a variety of solid tumors including gliomas. This study was to examine the effect of an EGFR-tyrosine kinase inhibitor （AG1478） alone or in combination with cisplatin （CDDP） on the growth of glioma cells （U87）. U87 glioma cells were treated with AG1478 （10 μmol/L） or CDDP （25 μmol/L） as a single agent or in combination for 24 or 48 h. The expression of EGFR and the components in its downstream signaling pathway [extracellular signal-regulated kinase （ERK）, protein kinase B （AKT）] in U87 glioma cells was detected by Western blotting. Cell growth, cell cycle distribution and cell apoptosis were determined by MTT method and flow cytometry, respectively. The results showed that CDDP could induce the activation of EGFR and the components in its downstream signaling pathways in a concentration-dependent manner. The combined treatment of AG1478 with CDDP could inhibit the proliferation of U87 glioma cells, arrest the-cell cycle and promote cell apoptosis. In the EGFR signaling pathway, AG1478 decreased the phosphorylation of ERK, AKT and EGFR in U87 glioma cells. It was：concluded that the combined treatment of AG1478 and CDDP may exert synergistic inhibitory effects on the growth of glioma cells by suooressing the activities of EGFR, AKT and ERK.

Thrombolytic efficacy of native recombinant staphylokinase on femoral artery thrombus of rabbits

Aim:To investigate the thrombolytic efficacy,ideal dosage and administration ofnative recombinant staphylokinase(r-SAK).Methods:Forty New Zealand rab-bits were randomly assigned into the control,r-SAK low-dose,medial-dose,high-dose,single bolus,allied therapy,recombinant streptokinase(r-SK)and uroki-nase(UK)*** right femoral artery thrombosis models were made byballoon injury,and 120 min after the injury,the thrombolytic agents were infusedthrough the rabbits’ parallel-ear ***:(1)2 h after balloon injury,thepulse pressures of the right femoral arteries reduced to 0 or less than 10% of thatof left femoral arteries in all groups;(2)after thrombolytic therapy,the pulsepressures in some of the femoral arteries markedly enhanced to more than 50% ofthat of left femoral arteries;(3)the reopening rates in the r-SAK medial and high-dose groups were significantly higher than that of the *** reopening rateof the same dose native r-SAK was significant higher than that of UK and r-SK;(4)the patency score of the right femoral arteries tended to be better in the r-SAKmedial and high-dose groups than that of the low-dose group,and the time toreopening in the allied therapy group tended to be ***:(1)r-SAKhas a definite thrombolytic effect on the femoral artery thrombus of rabbits;(2)single bolus is an effective manner of r-SAK therapy,and r-SAK allied therapywith heparin may shorten the time to recanalization;(3)the efficacy of the samedose native r-SAK was superior to that of r-SK and UK.
