Aim HSP is a common immunoglobulin A(IgA),especially lgA1-mediated vasculitis in *** has been showed that apoptosis of vascular endothelial cells played an important role in the pathogenesis of *** this study,we investigated the apoptosis of vein endothelial cells in HSP patients. Methods lgA1 was isolated from sera at the acute stage of childhood HSP and the apoptosis rate of human umbilical vein endothelial cells(HUVEC) induced by lgA1 was analyzed using flow cytometry and TUNEL ***-2,Bax and P53 mRNA and protein expression was determined by Real-time quantitative RT-PCR and western blotting methods. Results The results showed that isolated lgA1 markedly induced the apoptosis death of HUVEC compared with normal control *** the bax and P53 expression was up-regulated and the Bcl-2 expression was down-regulated in HUVEC co-cultured with lgA1 isolated from HSP patients for 24h. Conclusion These findings suggested that lgA1 isolated from HSP patients had an effect on the apoptosis of HUVEC;moreover bax/bcl-2 and p53 signaling pathways played a key role in the process of apoptosis in HUVEC induced by lgA1.

Objective: To investigate the factors affecting progno- sis of patients with primary hepatocellular carcinoma (PHC) after resection. Methods: From 1976 to 1992, 213 patients with PHC treated with hepatectomy were followed up for more than 5 years. Thirty-one of the patients who had sur- vived for more than 5 years were compared with 56 patients who had survived for less than 5 years. Results: Early detection of tumor, radical resection, number of tumors, capsule formation, operation safe distance, presence of portal tumor embolus, pre- sence of cirrhosis and tumor size were important fac- tors affecting the prognosis of patients with PHC. The proportion of small liver cancer in the patients who had survived for more than 5 years was greatly larger than that in the control group. Conclusions: Early detection of tumor and radical re- section are of value in raising the 5-year survival rate. Much remains to be investigated about the rela- tionship between safe margin distance and prognosis of patients with PHC.

However the expression and role of ASICs in gastrointestinal tract of HSP patients was still *** study was designed to examine the expression and localization of ASICs in gastric mucosa from HSP patients Methods Endoscopic biopsies of gastric mucosa were obtained from 25 patients during over a year from January 2010 to June *** expression of ASICs was done by using immunochemical techniques. Results The results showed that there was a significant increase in the mean relative optical density of ASIC-2 and ASIC-3,but not ASIC-1 ain the lining epithelium and glandular tubes of gastric mucosa from HSP patients. Conclusion This finding suggested that ASICs may be related to the pathogenesis of gastrointestinal manifestations in HPS patients.

Epidermal growth factor receptor （EGFR） is fotmd to express at high levels in a variety of solid tumors including gliomas. This study was to examine the effect of an EGFR-tyrosine kinase inhibitor （AG1478） alone or in combination with cisplatin （CDDP） on the growth of glioma cells （U87）. U87 glioma cells were treated with AG1478 （10 μmol/L） or CDDP （25 μmol/L） as a single agent or in combination for 24 or 48 h. The expression of EGFR and the components in its downstream signaling pathway [extracellular signal-regulated kinase （ERK）, protein kinase B （AKT）] in U87 glioma cells was detected by Western blotting. Cell growth, cell cycle distribution and cell apoptosis were determined by MTT method and flow cytometry, respectively. The results showed that CDDP could induce the activation of EGFR and the components in its downstream signaling pathways in a concentration-dependent manner. The combined treatment of AG1478 with CDDP could inhibit the proliferation of U87 glioma cells, arrest the-cell cycle and promote cell apoptosis. In the EGFR signaling pathway, AG1478 decreased the phosphorylation of ERK, AKT and EGFR in U87 glioma cells. It was：concluded that the combined treatment of AG1478 and CDDP may exert synergistic inhibitory effects on the growth of glioma cells by suooressing the activities of EGFR, AKT and ERK.

AIM:To investigate the diverse characteristics of different pathological gradings of gastric adenocarcinoma (GA) using tumor-related ***:GA tissues in different pathological gradings and normal tissues were subjected to tissue *** of 15 major tumor-related genes were detected by RNA in situ hybridization along with 3' terminal digoxin-labeled anti-sense single strandedoligonucleotide and locked nucleic acid modifying probe within the tissue *** data obtained were processed by support vector machines by four different feature selection methods to discover the respective critical gene/gene subsets contributing to the GA activities of different pathological ***:In comparison of poorly differentiated GA with normal tissues,tumor-related gene TP53 plays a key role,although other six tumor-related genes could also achieve the Area Under Curve (AUC) of the receiver operating characteristic independently by more than 80%.Comparing the well differentiated GA with normal tissues,we found that 11 tumor-related genes could independently obtain the AUC by more than 80%,but only the gene subsets,TP53,RB and PTEN,play a key *** the gene subsets,Bcl10,UVRAG,APC,Beclin1,NM23,PTEN and RB could distinguish between the poorly differentiated and well differentiated *** of a single gene could obtain a valid ***:Different from the traditional point of view,the well differentiated cancer tissues have more alterations of important tumor-related genes than the poorly differentiated cancer tissues.

BACKGROUND: Active immunotherapy has been successful in preventing many infectious diseases, and is being explored for its anti-tumor use. Purified antigens, peptides, gene-based systems and antigens contained in whole cells or cell lysates are used in specific active immunotherapy for cancer, known as cancer vaccines. Cancer vaccines do not directly kill tumor cells, but prime a specific humoral and/or cellular immune response against the tumor. Up to date, many kinds of cancer vaccines have been tested in the world and have shown their own advantages. Heat shock protein (HSP)-based cancer vaccine is one of the outstanding representatives. In this paper, we review recent advances in HSP-based cancer vaccines. DATA SOURCES: An English-language literature search was conducted using MEDLINE (1990-2005) on HSP, cancer vaccines and other related subjects. RESULTS: Several kinds of HSP-based cancer vaccines which have been explored worldwide, include tumor derived HSP-pepdde complex cancer vaccines, artificially reconstituted HSP-peptide complex cancer vaccines, HSPpeptide fusion protein cancer vaccines and HSP-based DNA cancer vaccines, etc. Many HSP-based cancer vaccines are being tested in clinical trials, and some are being tested in phase Ⅲ clinical trials at present. CONCLUSION: The available results in preclinical tests and clinical trials indicate that HSP-based cancer vaccines are promising in cancer therapy.
