＜正＞Objective To investigate the neuroprotective actions of pioliotazone（Piog） and the effects of Piog-mediated PPARγ activation on NF-B signaling pathway in rat model of permanent focal cerebral *** Permanent middle cerebral artery occlusion（pMCAO） model was induced by using intraluminal filament technique in ***（0.5, 1 and 2 mg/kg） was administrated intraperitoneally（i.p.） twice at 24 h before and at the time of ischemia,or administrated intraperitoneally（i.p.） only once 10 min after the onset of *** neuroprotection of Piog was analyzed by scoring neurological deficits,assessing brain infarction volume with 2,3,5-triphenyltetrazolium chloride（TTC） and determining the brain water content at 24 h after ischemia.A selective PPARγ antagonist GW9662 150,300 and 600 nmol/kg was administrated intracerebral ventricularly（i.c.v.） 30 min before *** then Piog（1 mg/kg） was administrated intraperitoneally （i.p.） 10 min after the onset of *** volume was assessed with 2,3,5-triphenyltetrazolium chloride （TTC） at 24 h after *** blot analysis was employed to determine alterations in IκBy,p-ERK in ischemic striatum at the indicated *** sections were immunostained for NF-kB p65 in ischemic ***-PCR was used to measure p53 mRNA levels in ischemic *** Piog（0.5,1 and 2 mg/kg） reduced infarction volume in a dose-dependent manner（P＜0.05,P＜0.01） when administered twice at 24 h before and at the time of ischemia,or only once at 10 min after the onset of *** 0.5,1 and 2 mg/kg also significantly ameliorated motor behavior deficits,diminished the increase in brain water content（P＜0.01）.Pretreatment with the irreversible PPARγantagonist GW9662 300 and 600nmol/kg abolished piog’s neuroprotection against ischemia（P＜0.01）.Western blot analysis revealed that Piog（0.5,1 and 2 mg/kg） markedly up-regulated the levels of IκBγin ischemic striatum at 12 h after ischemia（P＜0.01）.GW9662 significantly decr

Objective The purpose of this study was to predict outcome at 6 months for mild Traumatic Brain Injury(MTBI) patients with negative head computed tomography(CT) scans by combining clinical and magnetic resonance imaging(MRI) *** and Methods This prospective study took place between June 2008 and June 2010 in a Neurosurgical Intensive Care Unit in Tai-zhou,*** criteria were MTBI patients with negative head CT scans within the first 24 hours after trauma and with MRI-positive within the first 72 hours after *** clinical neurological symptoms were assessed during the first administration:Nausea and/or vomiting,Feeling depressed,Forgetfulness and Poor *** and T2* acquisitions on MRI were used in order to localize brain *** linked regions(clusters) were *** was assessed at 6 months post-trauma using the Glasgow Outcome Scale- Extended(GOSE).Results 45 patients were included: 10 had Unfavorable outcome(GOSE 1-6) and 35 had favorable outcome(GOSE 7-8).Lesions in the clusters "The left lower frontal lobe","The left anterior temporal lobe" and the presence of Feeling depressed or Poor concentration were associated with poor outcome in multivariate *** combined clinical and MRI approach gives a much better prediction than MRI approach only(P<0.01),with an area under the ROC curve of 0.98(95 %CI,0.95-1.00).Conclusions These data suggest that MRI associated with clinical assessment improves outcome prediction in MTBI patients with negative head CT *** depressed or Poor concentration should be assessed acutely in MTBI patients with a negative head CT scan to determine whether to require further MRI scan of the brain or not,particularly the symptoms worsen.

目的　研究新型银杏叶提取物制剂GBE5 0对高脂血症大鼠脑缺血再灌注损伤的保护作用。方法　在高脂血症大鼠上 ,建立大脑中动脉栓塞 (MCAO)再灌注损伤模型 ,观察灌胃给予GBE5 0 (5 0、10 0、2 0 0mg/kg) 2周 ,对大鼠脑梗死体积、神经行为学、血液流变学等指标的影响。结果　高脂MCAO模型组大鼠脑梗死体积为(31± 5 ) % ,神经行为评分为 2 .7± 0 .8;给予GBE5 0小、中、大剂量三组的高脂MCAO大鼠的脑梗死体积分别为(13± 4 ) %、(14± 4 ) %和 (13.0± 2 .6 ) % ,与模型组比较差异有显著意义 (P <0 .0 1) ;神经行为学评分分别为 1.7± 0 .5、1.5± 0 .6和 1.3± 0 .5 ,大剂量组与模型组比较差别有显著意义 (P <0 .0 5 ) ,并使全血黏度和还原黏度有不同程度的改善。结论　GBE5 0可降低高脂MCAO大鼠的脑梗死体积 ,改善神经行为障碍和血液流变学。

So we first investigate the role of camosine in amygdaloid kindled seizures and PTZ-induce kindled seizure in rats. ＜正＞Caffeine,arguably the most widely consumed psychoactive drugs,has been recently shown to protection neuronal injury in animal models of stroke,Parkinson’s disease and multiple ***,the mechanism by which caffeine afford neuroprotection is not *** this study,we studied the effect of chronic treatment with caffeine on spinal cord extract-induced neuronal damage,an experimental autoimmune encephalomyelitis（EAE） model of multiple sclerosis in Wistar *** rats were treated with caffeine or water in drinking water at dose of 10 or 30 mg/kg per day,starting from 7 d prior to immunization,until 35 d *** day 7 following caffeine treatment,rats were immunized with guinea pig spinal cord homogenates emulsified in complete freund adjuvant（CFA）,or with CFA *** monitored their body weight,movement behavior,and neurological deficient scores daily for next 35 *** also examined demyelination of spinal cord by immunohistochemistry and inflammatory cell infiltration,expression of IFN-γ,TGF-β,IL-12P40 mRNA in cervical cord by reverse transcription- polymerase chain reaction（RT-PCR）.The results show that the incidence of EAE in rats treated with 10 or 30 mg/kg of caffeine were significantly lower（31.8%or 33.3%） than the rats treated with water（67.9%） following EAE induction（P＜0.05）,while there was no incidence of EAE in rats treated with CFA *** the EAE rats treated with water,massive inflammatory cells were observed in the cerebral cortex and sacral cord;however, inflammatory infiltration were significantly attenuated in the cerebrum and sacral cord of the EAE rats treated with caffeine. Furthermore,at the 35 d post-immunization,compared to the EAE rats treated with water,the IFN-γmRNA level in cervical cord was significantly reduced while the TGF-βlevel was significantly enhanced in the EAE rats treated with 30 mg/kg of caffeine（P＜0.05）.These findings suggest that chronic treatment with caffeine could relieve intlammatory spinal

Viral vectors as a novel tool for clinical and neuropsychiatric research applications

Background A viral vector is a genetically modified vector produced by genetic engineering. As pathogenic genes in the virus are completely or largely eliminated, it is safe to be widely used in multidisciplinary research fields for expressing genes, such as neuroscience, metabolism, oncology and so on. Neuroscience and psychiatry are the most closely related disciplines in either basic research or clinical research, but the application of viral vectors in neuropsychiatry has not received much attention or not been widely *** This article will focus on the application of viral vectors in basic and clinical neuropsychiatric *** By using viral vectors, scientists can perform neurological labelling, gene expression regulation and physiological manipulation for investigating phenomenon from molecular mechanisms to behaviours. At the same time, to treat mental or neurological disorders, viral vectors can be designed for gene therapy, which alter gene expression levels or repair mutated genes in the brains of *** Viral vectors play an important role in basic research and clinical applications. To further understand brain function and prevent mental and neurological diseases, we hypothesize that viral vectors could be used along with various advanced technologies, such as sequencing and high-throughput expression analysis in the neuroscience research field.
