Genistein stimulates osteoblastic differentiation via p3. MAPK-Cbfa1 pathway in bone marrow culture

Aim:To test the hypothesis that genistein stimulates the osteoblastic differentia-tion through the p3. mitogen activated protein kinase (MAPK)-core-binding fac-tor 1 (Cbfa1) ***:The activation of p3. MAPK was detected byWestern *** phosphatase (ALP) activity and calcium depositionwere assessed for osteoblastic differentiation of bone marrow-derived mesenchy-mal stem cell (BMSC) *** expression of Cbfa1 was analyzed at both themRNA and protein *** activity of Cbfa1 was detected by electrophoreticmobility shift *** sialoprotein (BSP),ALP,osteocalcin (OC),andosteopontin (OPN) gene transcription were also evaluated by either RT-PCR orNorthern ***:Genistein (0.01-1 μmol/L) dose dependently led tothe rapid and sustained activation of the p3. MAPK pathway in mouse *** with genistein (1 μmol/L) resulted in increased ALP activityand calcium deposition of BMSC cultures as a function of *** alsoenhanced Cbfa1 DNA binding activity and promoted the expressions of Cbfa1itself as well as several Cbfa1-regulated genes,including ALP,BSP,OC,and *** treatment with p3. MAPK inhibitor (SB203.80) diminished thegenistein-induced osteoblastic maturation and p3. MAPK-Cbfa1 activation inmouse BMSC ***:These results indicated that genistein couldstimulate the osteoblastic differentiation of BMSC cultures through the p3.MAPK-Cbfa1 pathway.

In this paper,it is pointed out that the descriptions of alloy phase structures are dependent on structural unit *** the systematic science of alloys(SSA),the alloy phase structures are described by means of the symmetry element sequence combining with characteristic atom *** is named the characteristic atom arranging structure,which can display the characteristic atoms at the lattice sites and the micro-inhomogeneity,besides the *** characteristic atom has its own characters:neighboring configuration,potential energy,volume and electronic *** micro-inhomogeneity of alloy phases can be described by concentrations and short-range ordered parameters of characteristic *** differences between the electronic structures of alloy phases and electronic structures of characteristic atoms in the alloy phases are also discussed.

Phosphorylation/dephosphorylation is probably the most common and important reversible post-translational modification of proteins. Analyzing the functional effects of phosphorylation is helpful for understanding the biological functions of proteins. Identification of the phosphorylation sites of phosphorylated protein is a prerequisite for research on phosphorylation. In this work, an effective and simple method of identification of protein phosphorylation sites has been developed. Phosphopeptides were selectively enriched with immobilized metal affinity chromatography (IMAC) and subsequently chemically modified by 4-sulfophenyI isothiocyanate, and then the chemically modified phosphopeptides were sequenced with post-source decay (PSD) matrix-assisted laser desorption/ionization (MALDI) time-of-flight mass spectrometry for detecting phosphorylation sites. The charge of derivatization by 4-sulfophenyl isothiocyanate introduces a negative sulfonic acid group at the N-terminus of a peptide, and enables the selective detection of only a single series of C-terminal y-type ions. This chemically assisted method greatly simplifies the extremely complex pattern of PSD fragment ions and makes the PSD spectra more easier to be interpreted. The phosphorylation sites of a synthesized model phosphopeptide and human c-myc protein have been successfully identified by this method.

In this paper, a new risk model is studied in which the rate of premium income is regarded as a random variable, the arrival of insurance policies is a Poisson process and the process of claim occurring is p-thinning process. The integral representations of the survival probability are gotten. The explicit formula of the survival probability on the infinite interval is obtained in the special casc cxponential *** Lundberg inequality and the common formula of the ruin probability are gotten in terms of some techniques from martingale theory.

Upgrading graphite by flotation at Bogala Mines in Sri Lanka

Graphite is naturally floatable due to its hydrophobic pro pe rty and also soft and smears on other gangue particles, rendering the gangue mor e or less floatable too. Due to this reason it is important to concentrate on ar eas such as suitable flotation reagents, depression agents, pH modifiers, and pa rticle size to be fed during the process. The paper surveys and analyses the sui table particle size to be fed to achieve high-grade concentrate. According to t h e work carried out the author suggested the ideal cost effective flotation f low sheet for improved results at Bogala Mines in Sri Lanka.

MicroRNAs(miRNAs) constitute a family of small non-coding RNA molecules 22-25 nucleotides in *** control the expression of target genes at the post-transcriptional level by inhibiting translation or by degrading target mRNA through binding to complementary sequences in the 3.-untranslated *** studies have demonstrated that miRNAs are intimately involved in processes leading to nasopharyngeal carcinoma,such as the Epstein-Barr virus-encoded latent membrane protein 1 activated signal transduction pathways,gene-regulatory networks,mitosis,tumor angiogenesis,invasion and *** the relationship between miRNAs and the development of nasopharyngeal carcinoma,will further the understanding of this cancer and provide new avenues for diagnosis and treatment.

Effect of simvastatin on endothelium-dependent vaso-relaxation and endogenous nitric oxide synthase inhibitor

AIM: To investigate the effect of simvastatin on endothelium-dependent vasorelaxation and endogenous nitric oxide synthesis inhibitor asymmetric dimethylarginine (ADMA) in rats and cultured ECV3.4 cells. METHODS: Endothe- lial injury was induced by a single injection of low density lipoprotein (LDL) (4 mg/kg, 48 h) in rats or incubation with LDL (3.0 mg/L) or oxidative-modified LDL (100 mg/L) in cultured ECV3.4 cells, and vasodilator responses to acetylcholine (ACh) in the aortic rings and the level of ADMA, nitrite/nitrate (NO) and tumor necrosis factor- alpha (TNF-α) in the serum or cultured medium were determined. And the adhesion of the monocytes to endothe- lial cells and the activity of dimethylarginine dimethylaminohydrolase (DDAH) in the cultured ECV3.4 cells were measured. RESULTS: A single injection of LDL decreased endothelium-dependent relaxation to ACh, markedly increased the serum level of endogenous ADMA and TNF-α, and reduced serum level of NO. Pretreatment with simvastatin (3. or 60 mg/kg) markedly attenuated inhibition of vasodilator responses to ACh, the increased level of TNF-α and the decreased level of NO by LDL, but no effect on serum concentration of endogenous ADMA. In cultured ECV3.4 cells, LDL or ox-LDL markedly increased the level of ADMA and TNF-α and potentiated the adhesion of monocytes to endothelial cells, concomitantly with a significantly decrease in the activity of DDAH and serum level of NO. Pretreatment with simvastatin (0.1, 0.5, or 2.5 μmol/L) markedly decreased the level of TNF- α and the adhesion of monocytes to endothelial cells, butdid not affectthe concentration of endogenous ADMA and the activity of DDAH. CONCLUSION: Simvastatin protect the vascular endothelium against the damages induced by LDL or ox-LDL in rats or cultured ECV3.4 cells, and the beneficial effects of simvastatin may be related to the reduction of inflammatory cytokine TNF-α level.

Ti/Al_2O_3.Functionally Gradient Material Prepared by the Explosive Compaction/SHS Process

Ti1Al2O3.Functionally Gradient Material (FGM) was prepared by an explosive compaction/SHS process. Ten sheets of the compounding powder were laminated and pressed to get a green body of FGM. It was then compacted explosively By burying the explosive compaction body into a stoichiometric Al/TiO2 mixture and igniting the combustion of the stoichiometric Al/TiO2 mixture, the SHS reaction of the explosive compaction body was initiated by the heat released from the combustion of the stoichiometric Al/TiO2 mixture. In this way, Ti/Al2O3.FGM was synthesized. The adiabatic temperatures of each gradient layer were calculated when the preheating temperatures were 298 K and 1173.K, respectively The microstructure, composition and properties of Ti/Al2O3.FGM and the reaction mechanism of each gradient layer were studied. It was found that Ti/Al2O3.FGM prepared by the explosive compaction/SHS process had a high density and a high microhardness. Its structure, composition and properties showed apparent gradient distribution. The structure of the standard stoichiometric ratio gradient layer of FGM was a network structure. Its reaction mode could be described as follows: Al powder melted first, then the molten Al penetrated into the TiO2 zone and reacted with TiO2, and big pores were left in the original positions of Al powder. The reaction of gradient layers with the addition of Al3.3.as diluents was similar to that of the standard stoichiometric ratio gradient layer, so were their structure and composition. However, the reaction of gradient layers with the addition of Ti as diluents was more complex and the composition deviated slightly from the designed one
