Human fibrinogen-related protein-1/liver fibrinogen-related protein-1 (HFREP-l/LFIRE-1), a liver-specific protein, is a member of fibrinogen superfamily that exerts various biological activities. However, the function of HFREP-l/LFIRE-1 in liver remains unknown. Here we isolated its mouse ortholog gene-mouse fibrinogen-related protein-1 (mfrep-1), which encoded 314.amino acids, exhibiting 80.4. similarity to HFREP-l/LFIRE-1. Northern blot analysis revealed that 1.2-kb mfrep-1 mRNA was detected selectively in mouse liver. To explore the function of MFREP-1, we examined the levels of mfrep-1 mRNA during regeneration after 70% partial hepatectomy (PHx) in mice, mfrep-1 mRNA increased in the regenerating liver and reached the first shoulder peak at 2-4.h after PHx. Cycloheximide pretreatment could suppress the induction of mfrep-1, indicating the up-regulation of this gene need de novo protein synthesis. Hemodynamic factors play important roles in the formation, progression and rupture of cerebral aneurysms, and the Wall Shear Stress （WSS） and Oscillatory Shear Index （OSI） on the aneurysms are considered to be correlated with their growth and rupture. In this article, two computational models based on patient-specific cerebral aneurysms with daughter saccule are constructed from 3D-RA image data, one is lateral aneurysm located in middle cerebral artery （CA1） and the other is terminal aneurysm located in anterior communicating artery （CA2）, The corresponding models of the two aneurysms by removing daughter saccule are established in order to investigate the initiation and growth of the daughter saccule. The flow patterns and the distributions of hemodynamic factors in the two aneurysms before and after daughter saccule is removed are obtained by solving the governing equations with the commercial CFD software Ansys CFX11.0 under the non-Newtonian fluid assumption. By analyzing the flow patterns, it is concluded that the aneurysms with daughter saccules have more complex and unstable flow patterns and hence are prone to rupture. By comparing the distribution of OSI, a hypothesis that a high OSI causes the growth of the daughter saccule is presented.

Aim:To have a better understanding of the expression and regulation of versicanisoforms in neural precursor cells (NPC) and ***:By immunocytochemistry,RT-PCR,and real-time PCR,we examined the tem-poral expression of versican in NPC isolated from embryonic d 16 rats as wellas in oligodendrocyte (OL) lineage cells induced to differentiate from NPC,which mimicked the oligodendrogliogenesis in ***:We found thatversican was constitutively expressed in NPC and their lineage cells,includingneurons,astrocytes,and *** addition,2 versican isoforms,V1/V0 and V2,were found to express at low levels in NPC,but at significantly higher levels inOL lineage *** peak expression of versican V2 was found at the oligo-dendrocyte precursor cell ***,the treatment of 2 pro-inflam-matory cytokines,TNF-α and IFN-γ,enhanced the transcription of versican V2in NPC in a dose-dependent manner,but showed no effect on V1/V0 ***:Taken together,our results demonstrate that versican,particularlythe inhibitory V2 isoform,is increasingly expressed in OL lineage cells inducedto differentiate from *** increase in versican V2 expression after cytokinestimulation implies the interplay between the injury-induced upregulation ofinflammatory cytokines and chondroitin sulfate proteoglycan-mediated inhibi-tion of axonal regeneration after central nervous system injury.

采用Hilbert-Huang变换(HHT)方法对人体颈动脉、腕动脉脉搏压力波进行同步信号分析,对健康志愿者以及冠心病患者术前、术后的颈、腕同步脉搏波分别进行模态分解,同时引入互相关方法,从而得到健康志愿者以及冠心病患者术前、术后的颈、腕同步脉搏压力波各阶相应模态之间的互相关特征.通过互相关特征曲线发现,健康人体颈、腕动脉第2阶模态的波形周期对应于其脉搏周期的1/3,第3阶模态的波形周期对应于其脉搏周期的1/2,第4.模态与脉搏波周期相近.健康人体颈动脉、腕动脉第1阶模态均呈现较为标准的X波模态.冠心病患者颈、腕动脉第1阶模态互相关特征明显变异,术后相应模态特征得到显著改善.

Our previous work showed that the cartilage proteoglycan aggrecan could induce an erosive polyarthritis and spondylitis in BALB/c mice and the GI globular domain of the aggrecan (GI) contained the arthritogenic region. To elucidate whether autoreactive T cells to G1 are expressed in rheumatoid arthritis patients, we analyzed the frequency of human G1-specific T cells in the peripheral blood of five rheumatoid arthritis patients and tried to establish G1-reactive T cell lines from these rheumatoid arthritis patients. The results showed that the G1-specific T cells in PBL were detectable at the range of 4.97 ±0.5 ×10-6 in peripheral blood lymphocytes. We have also generated 15 G1-specific T lymphocyte lines from these pateints with a standard split-well method. All these cells expressed fine specificity to human recombinant G1, but not to unrelated antigen. All the 15 lines expressed a panT cell marker and 13 of them selectively used the αβ T cell receptor. Two of them used rye T cell receptor. The 13 of these T cell lines was CD4.positive. One line expressed CD8. One line expressed both CD4.and CD8. Moreover, 14.out of 15 lines expressed the Th-1 cytokine profile, characterized by interferon-γpositivity and IL-4. negativity. No Th-2 type cell line was generated. These data provide strong evidence in favor of the presence of autoreactive T cells in the rheumatoid arthritis pateints. What is the mechanism(s) that these autoreactive T cells attack self-target and whether these G1-specific, Th-1 type T cell lines can induce arthritis in immune deficiency mice are currently under investigation.

The aim of this study is to assess the effects of DNA methylation and historic acetylation, alone or in combination, on the expression of several tumor-associated genes and cell cycle progression in two established human colon cancer cell lines: Colo-320 and SW1116. Treatments with 5-aza-2’-deoxycytidine (5-aza-dC) and trichostatin A, alone or in combination, were applied respectively. The methylation status of the CDKN2A promoter was determined by methyla-tion-specific PCR, and the acetylated status of the histones associated with the p21WAF1 and CDKN2A genes was examined by chromatin immunoprecipitation. The expression of the CDKN2A, p21WAF1, p53, p73, APC, c-myc, c-Ki-ras and survivin genes was detected by real-time RT-PCR and RT-PCR. The cell cycle profile was established by flow cytometry. We found that along with the demethylation of the CDKN2A gene promoter in both cell lines induced by 5-aza-dC alone or in combination with TSA, the expression of both CDKN2A and APC genes increased. Aim:The present work was designed to investigate the relationship betweenhemodynamic parameters and the susceptibility of ventricular arrhythmia toaconitine in conscious Lyon hypertensive rats(LH).Methods:Male LH and Lyonlow blood pressure rats(LL)were *** the determination of baroreflexsensitivity(BRS),ventricular arrhythmia was induced by aconitine infusion inconscious *** pressure(BP)was recorded during the period of ***:Compared with the LL rats,the LH rats possessed significantly higherBP,blood pressure variability and lower *** threshold of aconitine requiredfor ventricular fibrillation and cardiac arrest in the LH rats were significantly lowerthan those in the LL *** was found that all the hemodynamic parametersstudied were not correlated with the threshold of aconitine required for arrhythmia,with the exception of BRS,which was positively related to the threshold ofaconitine required for ventricular premature ***:The LH rats pos-sessed greater susceptibility to aconitine-induced ventricular arrhythmias whencompared to the LL *** greater susceptibility could not be attributed to anyone of the hemodynamic parameters alone studied in the LH *** is proposedthat various hypertension-associated abnormalities,including the abnormalhemodynamics,may co-contribute to this vulnerability to ventricular arrhythmias.
