Viral vectors as a novel tool for clinical and neuropsychiatric research applications

Background A viral vector is a genetically modified vector produced by genetic engineering. As pathogenic genes in the virus are completely or largely eliminated, it is safe to be widely used in multidisciplinary research fields for expressing genes, such as neuroscience, metabolism, oncology and so on. Neuroscience and psychiatry are the most closely related disciplines in either basic research or clinical research, but the application of viral vectors in neuropsychiatry has not received much attention or not been widely *** This article will focus on the application of viral vectors in basic and clinical neuropsychiatric *** By using viral vectors, scientists can perform neurological labelling, gene expression regulation and physiological manipulation for investigating phenomenon from molecular mechanisms to behaviours. At the same time, to treat mental or neurological disorders, viral vectors can be designed for gene therapy, which alter gene expression levels or repair mutated genes in the brains of *** Viral vectors play an important role in basic research and clinical applications. To further understand brain function and prevent mental and neurological diseases, we hypothesize that viral vectors could be used along with various advanced technologies, such as sequencing and high-throughput expression analysis in the neuroscience research field.

Objective:To compare the effects of three antioxidants(sodium ascorbate,sodium ascorbate combined with a surfactant—Tween,catalase)on the microleakage of composite resin restorations after external tooth bleaching with 10%carbamide peroxide,to find effective ways to reduce microleakage. Methods:Sixty extracted human premolars,intact and health,were randomly divided into six groups:group 1,direct composite resin filling without bleaching;group 2 composite resin filling immediately after bleaching;group 3,immersed in artificial saliva three weeks after bleaching,then composite resin filling;group 4,cavity treated with sodium ascorbate after bleaching and then composite resin filling;group 5,cavity treated with sodium ascorbate combined with Tween after bleaching and then composite resin filling;group 6,cavity treated with catalase after bleaching and then composite resin *** 2000 thermal cycles,teeth were immersed in 2%methylene blue for 24 hours,then the microleakage of interface of restorations was observed under stereomicroscope. Results:Unbleached group displayed the least amount of microleakage,while the group of filling immediately after bleaching showed the greatest amount of microleakage,delay filling group and sodium ascorbate-treated group behaved similarly as the group of filling immediately after bleaching,having great amount of microleakage,showing no significant difference(P>0.05);the microleakage of sodium ascorbate combined with Tween-treated group and catalase-treated group decreased significantly(P< 0.05)compared to the group of filling immediately after bleaching. Conclusion:The microleakage increased significantly after external bleaching with 10%carbamide peroxide,then decreased treated with sodium ascorbate combined with Tween or catalase,and catalase displayed more efficiently,but delay filling and treated with sodium ascorbate perform badly.

We studied the quality of and internal changes in Huanghua pears (Pyrus pyrifolia Nakai,***) coated with shellac,SemperfreshTM (sucrose polyester base coating),and carboxymethyl chitosan (CMC) during cold storage (4℃). The changes in respiration rate,weight loss,cell membrane permeability and texture profile analysis (TPA) such as hardness, brittleness and chewiness were recorded periodically for up to 60 days after harvest to compare the effects of the applied coatings. Magnetic resonance imaging was used to inspect the internal characteristics of pears after *** soluble solid contents (SSCs),titratahle acidity,ascorbic acid concentration,and taste scores were also *** data suggested that shellac coating was more effective in reducing the respiration rate and weight loss and in maintaining the quality of pears than Semperfresh and CMC coatings.

＜正＞Objective Sacral neuromodulation（SNM） has been an effective method to recover and rebuild the bladder function after spinal cord injury（SCI）.The mechanism of SNM is still not well *** is generally believed that neuromodulation of reflex pathways at the spinal cord level as well as in the supraspinal pathways is involved in this *** present,it is still a problem how to choose suitable stimulation parameters for effective *** want to improve the effects of SNM by using different stimulation *** present,we are presenting a complete SCI rat model for spastic bladder to study SNM and standardize the surgical procedures and post-operative care. Methods The adult female Sprague-Dawley rats are completely transected in the T9-T10 vertebra after laminectomy in general *** gelfoam is inserted between the cut ends to fill the transection cavity of spinal cord to ensure complete *** prevent infection,every rat is introduced penicillin（150 mg/kg） for one week following *** SCI,the bladder of rats was manually expressed once a day（Crede’s maneuver）.The weight of expressed urine in SCI rats is used to reflect the residual volume and assess the recovery of bladder activity. Results The weight of urine expressed daily increases and reaches at a peak in 1-2 weeks post *** are all kinds of complications in SCI rats,some leading to death,but the complications can be decreased with proper postoperative *** several pilot experiments,the surgical procedures and post-operative care are standardized. We consider that the rat model behaves a flaccid bladder in the first 2 weeks after *** 2 weeks,along the recovery of spinal cord the rat behaves a spastic *** urine retention decreased dramatically,then urges urinary incontinence happens and could be used to study SNM.

Objective We sought to investigate the spatial torsion of the ipsilateral superior and inferior pulmonary veins(PVs),which was disregarded by many physicians. Methods A series of 48 consecutive atrial fibrillation patients,with 4 separate PVs,were e *** ostium plane of each PV was labeled by 3 landmarks on the reconstructed CT imag e in the Carto system ***,the left atrium(LA) axis was computed as a line con necting the 2 geometric centers of each side ***,a common plane was computed using the 2 geometric centers of the ipsilateral PVs and the center of the LA ***,the dih edral angle between the common plane and the ostium plane was *** torsion angle of the ipsilateral PVs was calculated as the absolute value of difference of the two dihedral angles. Results The torsion angle>15°was found in 16 left PVs(16/48) and in 9 right PVs(9/48, P=0.104).Moreover,in 2 cases(2.1%),the torsion angle exceeded 30°(1 left PVs and 1 right PVs).The torsion angle of the left PVs was significant greater than that of the right PVs(13.65°±5.90°vs.10.61°±5.96°,P=0.014). Conclusions Although with typical 4 seperate PVs,there was a significant torsion variance between the ipsilateral *** actual torsion variant of PVs should be taking into account whe n physicians plan their ablation to avoid a linear one plane encircle ablation.

＜正＞Parkinson’s disease（PD） probably represents a syndrome of different disorders and etiologies converging into a relatively uniform neurodegenerative *** loss of dopaminergic（DAergic） neurons in the substantia nigra（SN） is the pathological hallmark of PD but the mechanisms underlying the loss of DAergic neurons in PD are still poorly understood.A complex interaction of genetic and environmental factors has been thought to play a critical *** on our recent findings in the disease cause and pathogenesis,we postulate that the pathogenic mechanisms may begin much earlier,possibly even in the prenatal *** some patients with PD may be born with fewer than normal number of dopaminergic（and non-dopaminergic） neurons as a result of genetic or other abnormalities sustained during the prenatal or perinatal period,and as a result of the normal age-related neuronal attrition eventually reach the critical threshold（60% or more） of neuronal loss needed for onset of PD to become clinically *** this talk we provide emerging evidence that genetic disruption of normal development,coupled with subsequent environmental factors（the so called "multiple-hit hypothesis"）,plays an important role in the etiopathogenesis of PD.
