STAT1 Antisense Oligonucleotides Attenuate the Proinflammatory Cytokine Release of Alveolar Macrophages in Bleomycin-Induced Fibrosis

To investigate the effect of signal transducers and activators of transcription 1 （STAT1） antisense oligonucleotides （ASON） on concentrations of TNF-α IL-8, NO secreted by alveolar macrophages （AMs） in bleomycin-induced rat pulmonary fibrosis, five adult female Wistar rats were intratracheally instilled with bleomycin. After 7 days, the rats were killed by right ventricle of heart exsanguinations under ketamine anaesthesia and bronchoalveolar lavage （BAL） was performed to obtain AMs. AMs were divided into four groups, treated with STAT1 ASON, STAT1 sense oligonucleotides （SON）, dexamethasone （DEX） and medium alone （control）, respectively. AMs and media were collected after culture for 3. h. The mRNA and protein expressions of STAT1 and ICAM-1 in AMs were detected by RT-PCR and ELISA, respectively. The concentrations of TNF-α IL-8, NO in cultured medium were detected. The STAT1 mRNA expression by AMs in the STAT1 ASON group was lower than those of AMs in the STAT1 SON group, the DEX group and the control group （p 〈 0.05）. Moreover, the STAT1 mRNA expression by AMs in the DEX group was also lower than those of AMs in the STAT1 SON group and the control group （p 〈 0.05）, but the STAT1 mRNA expression by AMs in the STAT1 SON group was not different from that of the control group （p 〉 0.05）. The protein expressions of STAT1 and ICAM-1 and the mRNA expression of ICAM-1 showed similar changes to the STAT1 mRNA expression by AMs. The concentrations of TNF-α IL-8, NO in cultured medium from STAT1 ASON group were lower than those from STAT1 SON, DEX and the control groups （p 〈 0.05）. Moreover, the concentrations of TNF-α IL-8, NO in cultured medium from DEX group were also lower than those from the control and STAT1 SON group （p 〈 0.05）, but no difference between STAT1 SON group and the control （p 〉 0.05）. The results suggest that STAT1 ASON could inhibit the secretion of TNF-α IL-8, NO in AMs, and STAT1 could become a target of treating pulmonary fibrosis. Cellular ＆ Molecul

Association between VDR Apal Polymorphism and Hip Bone Mineral Density Can Be Modified by Body Mass Index:A Study on Postmenopausal Chinese Women

Osteoporosis is a major public health problem for old *** factors are considered tobe major contributors to the pathogenesis of postmenopausal *** vitamin D receptor(VDR)gene is a prominent candidate gene for the regulation of postmenopausal bone mass;however,despite exten-sive studies,controversy remains regarding its association with postmenopausal body mineral density(BMD)*** this study,a total of 260 healthy postmenopausal Chinese women were genotyped at the VDRApaI locus using polymerase chain reaction(PCR)-restriction fragment length polymorphism (RFLP).Rawhip BMD was significantly associated with VDR ApaI polymorphism with and without adjusting for age(P=0.015 and 0.040,respectively).This genetic effect can explain 3.3.% of hip BMD ***,the significant association vanished after correcting for both age and body mass index(BMI)(P=0.169).Inaddition,we observed a significant association between VDR ApaI polymorphism with unadjusted BMI(P=0.042)or BMI adjusted for age(P=0.049).The raw hip BMD was also found to be significantly corre-lated with BMI(r=0.517,P=0.0001),with BMI explaining 26.3.% of the variation of hip *** of thesefacts prompt us to conclude that the significant association between the VDR ApaI genotype and hip BMDmay be modified by BMI in postmenopausal Chinese *** findings may partially explain the earlierinconsistent association results concerning the VDR gene and BMD,and highlight the importance of incorpo-rating covariates such as BMI into osteoporosis association studies.

用5条制备有 Thomas 胰瘘和胃瘘的狗作慢性麻醉实验,观察刺激迷走神经和酸化十二指肠对胰液分泌的相互影响,结果如下:1.在酸化肠的情况下,刺激迷走神经所引起的胰蛋白质和碳酸氢盐的排出量显著增多,其效应超过单独刺激迷走神经和酸化肠所产生效应之和。2.在酸化肠引起胰分泌停止后的短时间内,再刺激迷走神经,胰液分泌的潜伏期缩短,蛋白质和碳酸氢盐排出量增多。3.阻断迷走冲动或注射阿托品后,酸化肠所引起胰液的分泌明显减少。4.用利多卡因麻痹肠粘膜后,酸化肠所引起胰液的分泌也明显降低。这些结果提示,在酸化十二指肠引起胰液分泌的机制中,有迷走神经和局部神经参与,迷走冲动和促胰液素及促胰酶素共同作用靶器官时,有相互加强作用,一旦迷走冲动被阻断,这两种激素的作用即明显降低。

Effects of esculentoside A on production of interleukin-1, 2, and prostaglandin E2

AIM: To investigate the influence of esculentoside A (EsA) on immunological function and its mechanism of anti- inflammation. METHODS: Interleukin-1 production was measured by thymocyte co-stimulating assay; the radio- activity of [3.]arachidonic acid (AA) was used to evaluate the release of AA; prostaglandin E2 production was measured with radioimmunoassay (RIA); IL-2 and IFN-γ were detected by ELISA method. RESULTS: EsA (3.12 μmol/L)could potently inhibit the production of IL-1 and PGE2 from both silent and LPS induced macrophages. EsA had no significant effect on the release of AA from murine macrophages. EsA could inhibit the production of IL-2 from murine lymphocytes induced by ConA, but not affect the production from silent lymphocytes. EsA showed no effect on the production of IFN-γ from both silent and ConA induced lymphocytes. CONCLUSION: EsA could affect the immunological function through inhibiting the production of IL-2 from activated splenocytes and the inhibition of production of IL-1 and PGE2 might be one of the anti-inflammation mechanisms of EsA.

CYP2E1 mediated isoniazid-induced hepatotoxicity in rats

AIM: To investigate the role of CYP2E1 in isoniazid (INH)-induced hepatotoxicity and the influence of rifampicin(RFP) on INH-induced liver injury. METHODS: Rats were treated with INH alone (100 mg/kg, ip) or co-administeredwith RFP (100 mg/kg, ig) for 10 d and 21 d. Hepatotoxicity was assayed by plasma enzymes (sALT, sAST) andhistopathological examinations. Hepatic CYP2E1 activity was measured by aniline hydroxylase (ANH), and CYP2E1mRNA expression was determined by RT-PCR. Plasma hydrazine concentration was determined by ***: For a 10 d INH-treatment, hepatic CYP2E1 level was increased to 3.7-fold over the control; liverimpairment appeared after 21 d treatment, while CYP2E1 and plasma hydrazine were, respectively, increased to 4.6-fold and 1.7-fold. However, in INH-RFP group for 10 d, CYP2E1 and plasma hydrazine were, respectively,decreased by 13.% and 18 % over INH group; similarly, hepatic injury is equal to INH group appeared after 21 d,and CYP2E1 was further decreased by 26 %. Correlation analysis showed that sALT had a positive correlation withplasma hydrazine and with CYP2E1 activity; CYP2E1 activity was also markedly correlated with plasma *** compared with control, there is no difference in changes of CYP2E1 mRNA expression in INH and INH-RFPtreatment for 21 d. CONCLUSION: The metabolite of INH, hydrazine, plays an important role in INH-inducedhepatotoxicity in rats. The induction of CYP2E1 by hydrazine is involved in the hepatotoxicity of INH. RFP doesnot exacerbate INH-induced hepatotoxicity in short term, which relates to down-regulation of CYP2E1.

Carrier Screening and Prenatal Gene Diagnosis of β-thalassemia by PCR-RDB Technique

In order to identify the distribution of gene types of β-thalassemia and reduce the birthrates of β-thalassemia major in Guiyang area, 1054 pregnant women and their spouses from Affiliated Hospital, Guiyang medical college were screened. The positive samples were analyzed with polymerase chain reaction and reverse dot blot method (PCR-RDB). When both partners were heterozygous identified as carriers for β- thalassemia, the risk of having a fetus who was homozygous or compound heterozygous was 2.66 %; the ratio of male to female was 1/1.15. Seven types of mutation were identified. CD17 and CD41-42 were dominant among them. Among the 4 cases subject to prenatal gene diagnosis, one fetus was completely normal and 3.fetuses were diagnosed as having β-thalassemia major (1 homozygous and 2 compound heterozygous). The fetuses diagnosed as β-thalassemia major were selectively terminated within two weeks. It was concluded that the birthrate of β-thalassemia major in Guiyang area was reduced and the target of improving birth outcome and child development has been achieved.

Hemorrhagic activity and mechanism of FIIa,a fibrinolytic enzyme from Agkistrodon acutus venom

AIM: To study the local hemorrhagic activity of a fibrinolytic enzyme (FIIa) from Agkistrodon acutus venom and its mechanism. METHODS: The local hemorrhagic activity was determined by subcutaneous injection on the back of mouse. The effects of FIIa on factor X, prothrombin, gelatin, and collagen were visualized by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). Platelet aggregation assays were performed in rat platelet- rich plasma (PRP). Human umbilical vein endothelial cells (HUVEC) were cultured and passaged in complete M199 medium. Cell viability and nuclear morphology change were determined by fluorescein diacetate (FDA) staining and Hoechst 3.258 staining, respectively. RESULTS: The minimum hemorrhagic dose (MHD) of FIIa was 89 μg. In vitro, FIIa (0.25 g/L) degraded factor X, prothrombin, collagen, and gelatin, and dose-dependently (0.25, 0.50, 0.75, and 1.00 g/L) inhibited the platelet aggregation induced by ADP in rat PRP. When HUVEC in culture treated with FIIa, HUVEC showed detachment in a dose-dependent manner, but no apoptosis sign was observed. CONCLUSION: FIIahad local hemorrhagic activity, and the mechanism was related to the degradation of factor X, prothrombin, gelatin, and collagen, the inhibition of ADP-induced platelet aggregation, and inducement of HUVEC detachment.2Now in Beijing Universityof Chinese Medicine,Beijing 100029,China.3.orrespondence to Dr Xiu-xia LIANG.

Objective To test a modified otologic drill under different drilling conditions for its ability to identify drilling faults and stop *** Based on force analysis and previous works,an otologic drill was modified and equipped with three *** various conditions,the drill was used to simulate three drilling faults and normal drilling,and signals from the drill were analyzed to extract the characteristic signal.A multi-sensor information fusion system and a stop program were designed to recognize drilling faults and stop *** Signals from each sensor changed consistently in response to drilling condition changes,with high repeatability and *** average identification rate was 72.625%,68.575%,70.5% and 81.3. respectively for the three simulated drilling faults and normal *** stop program stopped drilling in 0.2～ 0.3.seconds when a drilling faults was *** This study shows that the forces acting on the drill bit change predictably in the three simulated drilling conditions;that using suitable BP neural networks,the drilling faults can be reliably identified,and that a stop program based upon characteristic signal recognition can stop drilling quickly upon detecting drilling *** lays a foundation for development of a system capable of predicting drilling faults and automatic drill *** studies are being undertaken for practical application of such a system.
