Aim:The aim of the present study was to determine the effect of 5-aminoimidazole-4-carboxamide-ribonucleoside （AICAR） on proliferation,cell cycle,and apoptosisin the human epithelial cervical cancer cell line CaSki ***:Cell countand 3.（4,5-dimethylthiazol-2-yl）-2,5-diphenyltetrazolium bromide assay were usedto determine cell proliferation and *** 3.258 staining was con-ducted to distinguish the apoptotic *** cycle and Annexin-V/propidiumiodide staining were analyzed by fluorescence-activated cell sorting （FACS）.AWestern blot assay was used to evaluate the expression of AKT （also known asprotein kinase B）,mammalian target of rapamycin （mTOR）,p53.and extracellularsignal-regulated kinase （ERK）.Results:AICAR （500 μmol/L） significantly inhibi-ted the proliferation of CaSki cells treated for 24,48,and 72 h as determined by *** cells at the G1and G2phases were dramatically decreased while cells atthe S phase were increased in response to AICAR treatment for 24,48,and 72 *** MTT assay showed less viable cells and Hoechst fluorescent staining showedmore apoptotic cells upon AICAR *** results of the Annexin-Vstaining demonstrated a time-dependent increase of apoptosis in cells treatedwith AICAR for 24,3.,and 48 ***,AICAR activated caspase-3.in atime-dependent *** was also found that AICAR inhibited the phosphory-lation of AKT and mTOR,which are important kinases regulating cell growth *** stimulation obviously increased the expression of the tumorsuppressor p53.and the phosphorylation of ***:AICAR inhibitedproliferation and induced S phase arrest and promoted apoptosis in CaSki cells,which might be mediated by the downregulation of the AKT/mTOR pathway andthe upregulation of the p53.ERK pathway.

Expression of PDCD5 , a novel apoptosis related protein, in human osteoarthritic cartilage

AIM: To investigate the expression features of PDCD5 (programmed cell death 5 protein) in osteoarthritic andnormal human cartilage, and speculate on its potential functions in the pathogenesis of osteoarthritis (OA).METHODS: Articular cartilage specimens were obtained from 3. patients with OA and 16 healthy patients at thetime of arthroplasty. Expression of PDCD5 was detected by flow cytometry, immunofluorescence, RT-PCR andimmunohistochemical analysis. RESULTS: Enhanced expression and nuclear accumulation of PDCD5 in OAchondrocytes were found. PDCD5-positive chondrocytes were mainly distributed in the superficial and deep zonesof OA tissue sections, as opposed to, in the superficial and middle regions of normal healthy tissue ***: Since apoptotic chondrocyte death occurs more frequently in OA cartilage than in normal healthycartilage and PDCD5 is an apoptosis-related protein, the different expression patterns of PDCD5 in OA cartilagefrom that in normal healthy cartilage indicate that PDCD5 is involved in the pathogenesis of OA.

Adeno-associated virus-mediated bone morphogenetic protein-7 gene transfer induces C2C12 cell differentiation into osteoblast lineage cells

Aim: To investigate the effects of bone morphogenetic protein-7 (BMP7)-expressing recombinant adeno-associated virus (AAV) vector on the differentiation of C2C12 cells. Methods: AAV-BMP7 was packaged by infecting the stable cell clone BHK-21 (integrated with recombinant AAV vector plasmid pSNAV-BMP7) with recombinant herpes simplex virus type 1, which expresses AAV-2 Rep and Cap and possesses AAV packaging functions. Following infection with AAV-BMP7 at multiplicities of infection of lx 10～5 vector genomes per cell and subse-quent culture, C2C12 cells were assessed qualitatively for BMP7 production, alka-line phosphatase activity, osteocalcin production and Cbfal and MyoD expression. Results: C2C12 cells transduced with AAV-BMP7 could produce BMP7 protein until d 28. Alkaline phosphatase in the cultured C2C12 cell lysate was elevated. Secreted osteocalcin in the culture medium was detectable at d 12 and Cbfal mRNA expression level was upregulated, coinciding with downregulation of MyoD in a temporal manner. Conclusion: The present in vitro study demonstrated that AAV-BMP7 could infect and efficiently convert C2C12 cells from myoblasts into osteoblast lineage cells.

Study on the two and three-parameter working stability regions of 3.-TZP suspension

We determined the variation tendency of viscosity (η) at various solids load- ing (α) for 3.-TZP suspension as a function of dispersant concentration (Cw) using vis- cosity measurement, measured the diameter of particles as a function of Cw in very dilute suspension using light scattering method, and obtained the surface features of the sedi- ment of suspension with different Cw using SEM. We also discussed the influence of the microstructure of adsorbed polymer layers on particles and their interactions in the dis- persing medium on the stability of suspension. Then two different stable states and two different unstable states for 3.-TZP suspension were given. Accordingly, the (Cw, η), (Cw, α), and (α, η) two-parameter, and (Cw, α, η) three-parameter stability maps were con- structed. Based on the DLVO theory calculations, the dispersant concentration, particle distance (r), and interparticle potential energy (VT) maps with various solids loading for 3.-TZP aqueous suspension were also constructed. In (Cw, r) two-parameter coordinate system, the stable motion region map for particles was established. The Cw, α, and VT,max (the highest potential energy) surface of the potential barrier map was obtained through calculation and simulation on the basis of the surfaces of Cw, r, VT maps introduced above. The (Cw, α) two-parameter stability map was then obtained from the Cw, α, VT,max map. The results showed that this theoretical map can qualitatively prove the experimentally obtained results—the existence of different dispersed states of particles in suspension system and the variation tendency of suspension stability with changing each parame- ter—were reasonable.

Mechanism for calcite dissolution and its contribution to development of reservoir porosity and permeability in the Kela 2 gas field,Tarim Basin,china

This study is undertaken to understand how calcite precipitation and dissolution contributes to depth-related changes in porosity and permeability of gas-bearing sandstone reservoirs in the Kela 2 gas field of the Tarim Basin, Northwestern china. Sandstone samples and pore water samples are col-lected from well KL201 in the Tarim Basin. Vertical profiles of porosity, permeability, pore water chem-istry, and the relative volume abundance of calcite/dolomite are constructed from 3.00 to 4000 m below the ground surface within major oil and gas reservoir rocks. Porosity and permeability values are in-versely correlated with the calcite abundance, indicating that calcite dissolution and precipitation may be controlling porosity and permeability of the reservoir rocks. Pore water chemistry exhibits a sys-tematic variation from the Na2SO4 type at the shallow depth (3.00-3.3. m), to the NaHCO3.type at the intermediate depth (3.3.―3.95 m),and to the CaCl2 type at the greater depth (3.28―3.3. m). The geochemical factors that control the calcite solubility include pH, temperature, pressure, Ca2+ concen-tration, the total inorganic carbon concentration (ΣCO2), and the type of pore water. Thermodynamic phase equilibrium and mass conservation laws are applied to calculate the calcite saturation state as a function of a few key parameters. The model calculation illustrates that the calcite solubility is strongly dependent on the chemical composition of pore water, mainly the concentration difference between the total dissolved inorganic carbon and dissolved calcium concentration (i.e., [ΣCO2] -[Ca2+]). In the Na2SO4 water at the shallow depth, this index is close to 0, pore water is near the calcite solubility. Calcite does not dissolve or precipitate in significant quantities. In the NaHCO3.water at the intermedi-ate depth, this index is greater than 0, and pore water is supersaturated with respect to calcite. The Songshugou dunite body, which occupies an area of about 20 km2, is the largest ultramafic massif in the eastern Qinling orogenic belt, Central china. The major compo-nent of this body is dunitic rocks including mylonitic dunite and coarse-grained dunite; they oc-cupy about 95 vol% of the total body. Petrography, mineral composition, major and trace ele-ments and primitive melt inclusions have been investigated in this paper; all revealed that this dunite body is the product of melt-rock interaction by porous percolation flow. In comparison with dunite sills or veins in harzburgite from the basal part of Oman ophiolite, this dunite body is characterized by lower contents of Al2O3. CaO and HREE but higher content of highly incom-patible elements and Zr and Hf. The LREE enriched patterns and primitive mantle normalized spidergrams of trace elements are also different from the Oman dunite. Combining with melt in-clusions observed in olivines, we conclude that this dunite body is the product of large- scale porous percolation flow of high-MgO melts within depleted mantle peridotites. The high-MgO magma, in essence, was most probably produced at the hot head of an upwelling mantle plume. The occurrence of the Songshugou dunite body is closely associated with the activity of mantle super-plume in Neo-Proterozoic era in the Yangtze Craton.

Dehydration melting of solid amphibolite at 2.0 GPa: Effects of time and temperature

Two sets of dehydration-melting with a natural solid amphibolite, collected from North Himalayan structure zone, Tibet, have been carried out in multi-anvil apparatus at 2.0 GPa and 800―1000℃, for 12―200 h. One is keeping the pressure at 2.0 GPa and the annealing time of 12 h, changing the temperature (800―1000℃). The other is keeping the pressure at 2.0 GPa and temperature at 850℃, varying the annealing time (12―200 h). The products are inspected with microscope and electron probe. The results indicate that at 2.0 GPa, annealing time of 12 h, garnets, melts and clinopyroxenes occur in amphibolite gradually with increasing temperature and the chemical compositions of melt vary from tonalite to granodiorite, and then to tonalite. However, at 2.0 GPa and 850℃, with the annealing time increasing, the garnets, melts and cli-nopyroxenes also occur in amphibolite gradually and the chemical compositions of melt vary from tonalite to granodiorite. In both cases, melts interconnect with each other when the contents of melt are over the 5 vol.%. the viscosities of the melt produced in amphibolite at temperature higher than 850℃ are on a level with 104 Pa·s. The interconnected melt with such a viscosity may segregate from the source rock and form the magma over reasonable geological time. Therefore, it is believed that at the lower part of the overthickened crust, the tonlitic and grano-dioritic magma may be generated through the dehydration melting of amphibolite.

Distinguishing malignant from normal stomach tissues and its in vivo, in situ measurement in operating process using FTIR Fiber-Optic techniques

Gastric tissue samples were studied using mid-IR fiber-optic attenuated total reflectance (ATR) spectroscopy. FTIR spectra of 90 tissue samples from 48 patients, including 3. normal and 58 malignant tissue samples, were chosen as examples. Malignancy was usually characterized by the absence of CH and C=O bands, a weak amide II band near 1545 cm-1, a shift of the amide I band to lower wave number, a decrease in the ～1450 cm-1 peak to less than the～1400 cm-1 peak. Subtraction spectra indicate that the amide I and amide II bands of normal and malignant tissues have larger differences in peak positions and relative intensities. The statistical analysis results confirm this conclusion. The results indicate that FTIR fiber optic techniques provide important information about cancerous tissue of the stomach, which can be used to differentiate the malignant tissue from the normal tissue. Based on the above results we successfully realize the detection of the tumor tissues of digestive tract in vivo and in situ. And the results of detection cancer near operating room and in vivo and in situ in the operating room are consistent with the conclusions for the samples stored in liquid N2, which is the basis for the clinical application.

Some studies have shown that native Chinese speakers have different laterality in matched Stroop tasks from native English ***,many imaging data,which show left laterality of English-matched Stroop interference,have been *** a few functional imaging studies have been conducted to investigate the phenomenon of the Chinese version of Stroop *** this study,functional activity in the lateral prefrontal cortex of a group of normal Chinese boys with functional near-infrared imaging during a Stroop color-word task was measured to show different Stroop interferences in the prefrontal *** results show obvious fluctuation of the cerebral blood volume in the right prefrontal cortex in all boys,which agrees with the finding of previous studies,that is,Chinese native boys have right laterality in their brain when the Chinese version of Stroop color-word task is applied.
