Enhancing growth, liver health, and bile acid metabolism of tilapia(Oreochromis niloticus) through combined cholesterol and bile acid supplementation in plant-based diets

The present study aimed to compare the nutritional effects of cholesterol, bile acids, and combination of cholesterol with bile acids in plant-based diets on juvenile genetically improved farmed tilapia(GIFT;Oreochromis niloticus). The isonitrogenous(321 g/kg crude protein) and isolipidic(76 g/kg crude fat) diets(Con diet) were based on plant protein sources, which included corn gluten meal, soybean meal, cottonseed meal and rapeseed meal. The Con diet was supplemented with 12 g/kg cholesterol(CHO diet), 0.2 g/kg bile acids(BAs diet), a combination of 12 g/kg cholesterol and 0.2 g/kg bile acids(CHO-BAs diet),respectively. Each diet was fed to three tanks in an indoor recirculating aquaculture system for 9 *** showed that compared to the Con group, fish had a higher weight gain rate, hepatosomatic index,and a lower feed conversion ratio in the CHO-BAs group. The highest levels of whole-fish fat and ash were found in the Con group. Serum parameters, including activities of alanine aminotransferase(ALT) and aspartate transaminase(AST), along with levels of glucose(GLU) and triglyceride(TG) except for total cholesterol(TCHO), were lower in the CHO, BAs, and CHO-BAs groups than those in the Con group(P <0.001). Histological examination revealed that fish in the Con group exhibited severe hepatocyte vacuolization and diminished hepatocyte proliferation. Gene expression analysis indicated that the transcriptional levels of bile acid metabolism-related genes(including fxr, fgf19, bsep) were up-regulated in the CHO-BAs group(P < 0.05), whereas cholesterol metabolism-related genes(acly and hmgcr) were downregulated in both CHO and CHO-BAs groups(P < 0.001). Moreover, UPLC-MS/MS analysis revealed that the higher taurine-conjugated bile acids(T-BAs), followed by free bile acids(Free-BAs) and glycine(G-BAs)were determined in tilapia bile. Inhibition of Aloperine on Dextran Sulphate Sodium-induced Chronic Colitis in C57BL/6 Mice

Objective To investigate the effects of aloperine (ALO) on a model of dextran sulphate sodium (DSS)-induced chronic colitis in C57BL/6 mice. Methods Repeated colitis was induced by administration of four cycles of 4% DSS. The severity of colitis was assessed on the basis of clinical signs, ratio of colon weight and colon length, and histological grading scores. Moreover, secretory immunoglobulin A (S-IgA) and plasma haptoglobin (HP) were analyzed by enzyme-linked immunosorbent assay, and the changes of mRNA expression of ICAM-1 and MIF gene in colorectal tissue were detected by quantitative reverse transcriptase real-time polymerase chain reaction using SYBR Green I. Results ALO administration significantly attenuated the colon damage, caused substantial reductions of the rise in HP, and maintained the level of cecum S-IgA. ALO inhibited the ICAM-1 mRNA expression and had no effect on MIF mRNA expression. Conclusion The effect of ALO on DSS-induced chronic colitis in mice is investigated for the first time, which suggests that ALO could be an attractive therapeutic candidate in the treatment of inflammatory bowel disease.

Discontinuity development patterns and the challenges for 3D discrete fracture network modeling on complicated exposed rock surfaces

Natural slopes usually display complicated exposed rock surfaces that are characterized by complex and substantial terrain undulation and ubiquitous undesirable phenomena such as vegetation cover and *** study presents a systematic outcrop research of fracture pattern variations in a complicated rock slope,and the qualitative and quantitative study of the complex phenomena impact on threedimensional(3D)discrete fracture network(DFN)*** the studies of the outcrop fracture pattern have been so far focused on local variations,thus,we put forward a statistical analysis of global *** entire outcrop is partitioned into several subzones,and the subzone-scale variability of fracture geometric properties is analyzed(including the orientation,the density,and the trace length).The results reveal significant variations in fracture characteristics(such as the concentrative degree,the average orientation,the density,and the trace length)among different ***,the density of fracture sets,which is approximately parallel to the slope surface,exhibits a notably higher value compared to other fracture sets across all *** improve the accuracy of the DFN modeling,the effects of three common phenomena resulting from vegetation and rockfalls are qualitatively analyzed and the corresponding quantitative data processing solutions are ***,the 3D fracture geometric parameters are determined for different areas of the high-steep rock slope in terms of the subzone *** results show significant variations in the same set of 3D fracture parameters across different regions with density differing by up to tenfold and mean trace length exhibiting differences of 3e4 *** study results present precise geological structural information,improve modeling accuracy,and provide practical solutions for addressing complex outcrop issues.

Ultra-high nickel layered oxide cathode material with high energy density is the most promising material to improve the electrochemical performance of lithium-ion batteries(LIBs).However,the poor structural stability and severe surface/interface side reactions of the material lead to poor rate performance and cyclic stability,which limits its application in *** this paper,the dual-modification strategy of Co doping and La_(2)O_(3) coating is used to meet the above *** doping can effectively widen layer spacing and reduce Li^(+)/Ni^(2+) mixing,and La_(2)O_(3) coating can effectively eliminate the residual alkali on the surface of active material,inhibit the thickening of cathode electrolyte interphase(CEI)film and reduce surface/interface side ***,the modified material(NM90-CL)with excellent electrochemical properties is achieved through the synergistic enhancement of Co doping and La_(2)O_(3) *** capacity retention rate can reach 77.9%after 200 cycles at 1.0℃ and 75.7%after 200 cycles at 5.0℃.Its reversible capacity can up to 153.5 mAh·g^(–1) at 10.0℃.This dual-modification strategy will provide theoretical guidance and technical support for the synthesis of other high-performance electrode materials.

As the brightest gamma-ray burst ever observed,GRB 221009A provided a precious opportunity to explore spectral line *** this article,we performed a comprehensive spectroscopy analysis of GRB 221009A jointly with GECAM-C and Fermi/GBM data to search for emission and absorption *** the first time we investigated the line feature throughout this GRB including the most bright part where many instruments suffered problems,and identified prominent emission lines in multiple time *** central energy of the Gaussian emission line evolves from about 37 to 6 MeV,with a nearly constant ratio(about 10%)between the line width and central ***,we find that both the central energy and the energy flux of the emission line evolve with time as a power law decay with power law index of–1 and–2,*** suggest that the observed emission lines most likely origin from the blue-shifted electron positron pair annihilation 511 ke V *** find that a standard high latitude emission scenario cannot fully interpret the observation,thus we propose that the emission line comes from some dense clumps with electron positron pairs traveling together with the *** this scenario,we can use the emission line to directly,for the first time,measure the bulk Lorentz factor of the jet(Γ)and reveal its time evolution(i.e.,Γ~t^(-1))during the prompt ***,we find that the flux of the annihilation line in the co-moving frame keeps *** discoveries of the spectral line features shed new and important lights on the physics of GRB and relativistic jet.

Hemorrhagic activity and mechanism of FIIa,a fibrinolytic enzyme from Agkistrodon acutus venom

AIM: To study the local hemorrhagic activity of a fibrinolytic enzyme (FIIa) from Agkistrodon acutus venom and its mechanism. METHODS: The local hemorrhagic activity was determined by subcutaneous injection on the back of mouse. The effects of FIIa on factor X, prothrombin, gelatin, and collagen were visualized by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). Platelet aggregation assays were performed in rat platelet- rich plasma (PRP). Human umbilical vein endothelial cells (HUVEC) were cultured and passaged in complete M199 medium. Cell viability and nuclear morphology change were determined by fluorescein diacetate (FDA) staining and Hoechst 33258 staining, respectively. RESULTS: The minimum hemorrhagic dose (MHD) of FIIa was 89 μg. In vitro, FIIa (0.25 g/L) degraded factor X, prothrombin, collagen, and gelatin, and dose-dependently (0.25, 0.50, 0.75, and 1.00 g/L) inhibited the platelet aggregation induced by ADP in rat PRP. When HUVEC in culture treated with FIIa, HUVEC showed detachment in a dose-dependent manner, but no apoptosis sign was observed. CONCLUSION: FIIahad local hemorrhagic activity, and the mechanism was related to the degradation of factor X, prothrombin, gelatin, and collagen, the inhibition of ADP-induced platelet aggregation, and inducement of HUVEC detachment.2Now in Beijing Universityof Chinese Medicine,Beijing 100029,china.3Correspondence to Dr Xiu-xia LIANG.

Differential transcription-activating capability of NS1 proteins from different influenza virus subtypes expressed in yeast

Influenza A virus NS1 protein is an important regulatory factor with multiple functions and contributes greatly to viral *** the present study,transcription-activating potential of NS1 from different influenza A virus subtypes was examined in yeast two-hybrid *** bait vectors contain-ing different NS1 genes,along with an empty prey vector,were transformed into yeast AH109(for growth assay on QDO plate and α-galactosidase assay),and Y187(for β-galactosidase assay).AH109 transformants with NS1 gene from H1N1,H5N1,and H9N2 viruses grew vigorously on the QDO plate and secreted high level of α-***,Y187 bearing the above NS1 genes exhibited en-hanced β-galactosidase ***,H3N2-NS1-transformed AH109 and Y187 yeasts did not grow on QDO plate and secrete β-galactosidase,*** findings denote the remarkable variation in NS1 proteins from different influenza A virus subtypes on the transcription-stimulating capability in yeast.
