Aim:To investigate the effects of tanshinone ⅡA（Tan ⅡA）on the regulation ofthe production of endothelin（ET）-1（including large ET-1）,mRNA levels of ET-1,endothelin-converting enzyme-1（ECE-1）,endothelin-A receptor（ETA）andendothelin-B receptor（ETB）induced by TNF-α in rat brain microvascular endo-thelial cells（BMVEC）.Methods:The ET-1 release（including large ET-1）into theculture medium was determined by enzyme *** levels of ET-1,ECE- 1,ETA,and ETBmRNA were measured by *** receptor bind-ing was also ***:The induction of ET-1 release by TNF-α from cul-tured BMVEC was dose-dependently reduced by Tan IIA,but large ET-1 levelsprogressively increased in response to Tan ⅡA;the mRNA expression of ET-1 *** ⅡA also caused a decrease in ETAreceptor mRNA and ECE-1expression in a dose-dependent *** receptor binding was unal-tered in BMVEC stimulated with TNF-α alone or a combination of TNF-α and TanⅡ***:These findings suggest that Tan ⅡA may inhibit ET-1 produc-tion in TNF-α-induced BMVEC through the suppression of ECE-1 synthesis.

Transcriptional regulation of the neuronal nitric oxide synthase gene(nNOS)is particularlycomplex as 12 distinct transcripts derived from different first exons are expressed in a tissue- and cell-specific *** exon 1f mRNA is relatively highly expressed in nervous system and relies upon exon1f-specific promoter *** conventional and real-time reverse transcription-polymerase chain reaction,we found exon 1f mRNA was the major transcript of the nNOS gene in human neuroblastoma *** analyzed a 1090 bp fragment of 1f promoter by TRANSFAC-TESS and Match softwares andluciferase assay,and found an important positive transcriptional regulation region that contained a putativenuclear factor(NF)-κB binding ***,using electrophoresis mobility shift and chromatinimmunoprecipitation assays,we identified this site to be the NF-κB responsive element,a crucial positiveregulator in the activation of the nNOS 1f *** together,our study identified an NF-κB responsiveelement within nNOS 1f promoter and showed that it plays an important role in the transactivation of nNOS1f mRNA,the major transcript of nNOS in SK-N-SH cells.

AIM: To investigate the association of the cyclooxygenases-2 (COX-2) polymorphisms and susceptibility to gastric cancer (GC) by means of meta-analysis. METHODS: Publications addressing the association between polymorphisms of COX-2 and susceptibility to GC were selected from the MEDLINE, EMBASE and CBMdisc databases. Data was extracted from the studies by 2 independent reviewers. The meta-analyses were performed by RevMan 5.0.23. From these data, odds ratio (OR) with 95% confidence interval (CI) was ***: Ten studies were retrieved reporting a total of 11 COX-2 polymorphisms. Carriers of -765C, -1195A, -1290G, *2430T alleles and *429TT genotype revealed increased risk for GC (OR = 1.71, 95% CI: 1.01-2.90, P = 0.05; OR = 1.58, 95% CI: 1.05-2.38, P = 0.03; OR = 1.55, 95% CI: 1.01-2.39, P = 0.05; OR = 2.62, 95% CI: 1.20-5.73, P = 0.02 and OR = 0.74, 95% CI: 0.59-0.95, P = 0.02, respectively). CONCLUSION: The -765C, -1195A, -1290G, *2430T alleles and *429TT genotype of COX-2 polymorphisms were determined a significant association with susceptibility to GC.

We conducted a study concerning the suppressive mechanism of KAI1/CD82 on hepatoma cell *** growth factor(HGF)induces the migration of hepatoma cells through activation of cellular sphingosine kinase 1(SphK1).Adenovirus-mediated gene transfer of KAI1(Ad-KAI1)downregulates the SphK1 expression and suppresses the HGF-induced migration of SMMC-7721 human hepatocellcular carcinoma *** of KAI1/CD82 significantly elevates Sprouty2 at the protein *** of Sprouty2 with RNA interference can block the KAI1/CD82-induced suppression of hepatoma cell migration and downregulation of SphK1 *** is demonstrated that KAI1/CD82 suppresses HGF-induced migration of hepatoma cells via upregulation of Sprouty2.

BACKGROUND: Alcoholic hepatitis (AH) is a necrotizing inflammatory process caused by alcoholic liver injury. It carries a significant short-term mortality. The management of AH is challenging. Although corticosteroids have been demonstrated to exert anti-inflammatory and antifibrotic effects, their efficacy for the treatment of AH remains debatable. DATA SOURCES: A literature search was performed of MEDLINE, ScienceDirect, SpringerLink and Wiley InterScience using the key words 'alcoholic hepatitis', 'alcoholic liver disease', and 'corticosteroids'. The available data reported in the relevant literature were analyzed. RESULTS: More than 17 controlled trials and at least 13 meta-analyses have reported the efficacy of corticosteroids in the treatment of AH in the past 40 years. Many were poorly designed and used different inclusion/exclusion criteria, making it difficult to reach a consensus. In this review, we summarized all the controversial data in the past decade and analyzed the potential causes for the varying therapeutic effects of corticosteroids in AH. The focus of the controversy has changed from 'whether steroids are beneficial or harmful for AH patients' to 'how to accurately identify responders to steroids early and rationalize corticosteroid treatment'. An early response to glucocorticoids, as determined by calculating the Lille score after 7 days of treatment, has been shown to be a clinically useful indicator. Moreover, down-regulation of steroid sensitivity, risk of infection, and a rational therapeutic strategy of corticosteroids in AH patients are all crucial for therapeutic ***: An early and accurate determination of steroid sensitivity is important. Besides, we need to overcome the down-regulation of steroid sensitivity, reduce the infection risk and rationalize the therapeutic strategy of corticosteroids. A fresh perspective is needed on the use of corticosteroids in AH patients.

BACKGROUND: Calcitonin gene-related peptide (CGRP) and nerve growth actor (NGF) cam improve spatial learning and memory abilities of rats with cerebral ischemia/reperfusion; however, the effect of combination of them on relieving learning and memory injury following cerebral ischemia/reperfusion should be further studied. OBJECTIVE: To study the effects of exogenous CGRP and NGF on learning and memory abilities of rats with focal cerebral ischemia/reperfusion. DESIGN: Randomized controlled animal study. SETTING: department of neurosurgery, the Second hospital of Xiamen; department of neurosurgery, the Second Affiliated hospital of china.Medical University; department of Neurobiology, Basic Medical College of china.Medical University. MATERIALS: A total of 30 healthy male SD rats, aged 8 weeks, of clean grade, weighing 250-300 g, were provided by Experimental Animal department of china.Medical University. All rats were randomly divided into sham-operation group, ischemia/reperfusion group and treatment group with 10 in each group. The main reagents were detailed as the follows: 100 g/L chloral hydrate, 0.5 mL CGRP (2 mg/L, Sigma Company, USA), and NGF (1× 106 U/L, 0.5 mL, Siweite Company, Dalian). METHODS: The experiment was carried out in the department of Neurobiology, Basic Medical College of china.Medical University from February to July 2005. Rat models of middle cerebral artery occlusion were established by method of occlusion, 2 hours after that rats were anesthetized and the thread was slightly drawn out for 10 mm under direct staring to perform reperfusion. Rats in the ischemia/reperfusion group received intraperitoneal injection of 1 mL saline via the abdomen at two hours later, while rats in the treatment group at 2 hours later received intraperitoneal injection of 2 mg/L CGRP (0.5 mL) and 1×106 U/L NGF (0.5 mL) once a day for 10 successive days. In mammals,the ribbon synapses of cochlear inner hair cells are a synaptic structure of the first sensory nerve in the pathway of acoustical signal transmission to the acoustic center,and it is directly involved in voice coding and neurotransmitter release. It is difficult to quantitatively analyze the ribbon synaptic number only using an electron microscope,because the ribbon synaptic number is relatively limited and their location is deep. In this study,the specific presynaptic structure-RIBEYE,and non-specific postsynaptic structure-GluR 2 & 3 in C57BL/6J mouse basilar membrane samples were treated by immunofluorescent labeling. Serial section was performed on the samples using a laser scanning confocal microscope,and then the serial sections were used to build three-dimensional models using 3DS MAX software. Each fluorescein color pair indicates one synapse,so the number of ribbon synapses of inner hair cells is obtained. The spatial distribution and the number of ribbon synapses of cochlear inner hair cells were clearly shown in this experiment,and the mean number of ribbon synapses per inner hair cell was 16.10±1.03. Our results have demonstrated the number of ribbon synapses is accurately calculated by double immunofluorescent labeling to presynaptic and postsynaptic structures,serial sections obtained using a laser scanning confocal microscope,and three-dimensional modeling obtained using 3DS MAX software. The method above is feasible and has important significance for further exploring the mechanism of sensorineural deafness.

BACKGROUND: It has shown that magnetic field can improve blood circulation, decrease blood viscosity, inhibit free radicals, affect Ca2+ flow in nerve cells, control inflammatory and immunological reaction, and accelerate nerve cell regeneration. In addition, protective effect of magnetic field, which acts as an iatrophysics, on ischemic brain tissues has been understood gradually. OBJECTIVE: To investigate the effects of rotating magnetic field (RMF) on volume of cerebral infarction, cerebral edema and metabolism of free radicals in rats after cerebral ischemia/reperfusion injury. DESIGN: Randomized controlled animal study. SETTING: Rehabilitation Center of disabled children, Liaoniang; department of Rehabilitation, the Second Affiliated hospital, china.Medical University; department of Rehabilitation Physiotherapy, the First Affiliated hospital, china.Medical University. MATERIALS: A total of 70 healthy Wistar rats aged 18-20 weeks of both genders were selected and randomly divided into 3 groups: sham operation group with 12 rats, control group with 20 rats and treatment group with 38 rats. The treatment group included 4 time points: immediate reperfusion with 6 ones, 6-hour reperfusion with 20 ones, 12-hour reperfusion with 6 ones and 18-hour reperfusion with 6 rats. Main instruments were detailed as follows: magnetic head of rotating magnetic device was 6 cm in diameter; magnetic induction intensity at the surface of magnetic head was 0.25 T in silence; the maximal magnetic induction intensity was 0.09 T at the phase of rotation; the average rotating speed was 2500 r per minute. METHODS: The experiment was carried out in the china.Medical University in March 2003. Focal cerebral ischemic animal models were established with modified Longa’s method. AIM: To investigate the effects of inhibiting factor of cellcycle regulation p57kip2, retinoblastinoma protein (Rb protein)and proliferating cell nuclear antigen (PCNA) in the genesisand progression of human pancreatic ***: The expression of p57kip2, Rb protein and PCNAin tumor tissues and adjacent tissues of 32 patients withpancreatic cancer was detected with SP ***: p57kip2 protein positive-expression rate in tumortissues of pancreatic cancer was 46.9 %, which was lowerthan that in adjacent pancreatic tissues (75.0 %) (x2=5.317,P＜0.05), p57kip2 protein positive-expression correlatedsignificantly with tumor cell differentiation (well-differentiationversus moderate or low-differentiation, P＜0.05) but did notcorrelate significantly with lymph node metastasis (lymph nodemetastasis versus non-lymph node metastasis, P＞0.05); Rbgene protein positive-expression rate in tumor tissues was50.0 %, which was also lower than that in adjacent pancreatictissues (78.1%) (x2=5.497, P＜0.05); PCNA positive-expression rate was 71.9 %, being higher than that inadjacent pancreatic tissues (43.8 %) (x2=5.189, P＜0.05),PCNA positive-expression also correlated significantly withtumor cell differentiation and lymph node metastasis (well-differentiation versus moderate or low- differentiation, lymphnode metastasis versus non-lymph node metastasis, P＜0.05).Rb protein positive-expression rate in the tumor tissues ofp57kip2 protein positive-expression group was 53.3 %; andRb protein positive-expression rate in the tumor tissues ofp57kip2 protein negative-expression group was 47.1%. Therewas no significant relationship between the two groups(r=0.16507, P＞0.05).CONCLUSION: The decreased expression of p57kip2, Rbprotein or over-expression of PCNA protein might contributeto the genesis or progression of pancreatic cancer, p57kip2,Rb protein and PCNA may play an important role in genesisand progression of pancreatic cancer.

As a benign liver disease, end-stage viral hepatitis has been acknowledged as a main indication for liver transplantation. However, the recurrence of viral hepatitis is the major factor affecting the survival of transplant recipients. The recurrence of hepatitis B is defined as a return of positive HBsAg, while in case of hepatitis C, it is the return of positive HCV-RNA in serum. Several factors are believed as the hazardous factors of hepatitis recurrence, including the types and course of hepatitis, ethnical background, usage of antiviral drugs and immunosuppressive protocol. High dose, long term and combinative therapy of HBIg and novel drugs such as lamivudine and ribavirin offers the promising result of preventing hepatitis recurrence.
