Association between VDR Apal Polymorphism and Hip Bone Mineral Density Can Be Modified by Body Mass Index:A Study on Postmenopausal Chinese Women

Osteoporosis is a major public health problem for old *** factors are considered tobe major contributors to the pathogenesis of postmenopausal *** vitamin D receptor(VDR)gene is a prominent candidate gene for the regulation of postmenopausal bone mass;however,despite exten-sive studies,controversy remains regarding its association with postmenopausal body mineral density(BMD)*** this study,a total of 260 healthy postmenopausal Chinese women were genotyped at the VDRApaI locus using polymerase chain reaction(PCR)-restriction fragment length polymorphism (RFLP).Rawhip BMD was significantly associated with VDR ApaI polymorphism with and without adjusting for age(P=0.015 and 0.040,respectively).This genetic effect can explain 3.32% of hip BMD ***,the significant association vanished after correcting for both age and body mass index(BMI)(P=0.169).Inaddition,we observed a significant association between VDR ApaI polymorphism with unadjusted BMI(P=0.042)or BMI adjusted for age(P=0.049).The raw hip BMD was also found to be significantly corre-lated with BMI(r=0.517,P=0.0001),with BMI explaining 26.35% of the variation of hip *** of thesefacts prompt us to conclude that the significant association between the VDR ApaI genotype and hip BMDmay be modified by BMI in postmenopausal Chinese *** findings may partially explain the earlierinconsistent association results concerning the VDR gene and BMD,and highlight the importance of incorpo-rating covariates such as BMI into osteoporosis association studies.

Aim:To evaluate whether estrogen receptor α(ER-α)and vitamin D receptor(VDR)genes are associated with the age at menarche in Chinese ***:A total of 390 pre-menopausal Chinese women were genotyped at the ER-α PvuII,XbaI,and VDR Apal loci using polymerase chain reaction(PCR)-restriction frag-ment length polymorphism(RFLP).Results:Neither the ER-α gene nor the VDRgene individually had significant effects on the age at menarche in our subjects(P>0.10).However,evidence of interaction effects between the two genes wereobserved:with the aa genotype at the VDR ApaI locus,subjects with haplotypePX at the ER-α gene had,on average,6 months later onset of menarche than thenon-carriers(P=0.01).Conclusion:We found that neither the ER-α gene or theVDR gene had a significant association with the age at menarche ***,potential interaction effects between the two genes were observed inChinese women.

Studies on monoclonal antibody AU14-1 and immunology of uterine cervical cancer

The article begins with a brief description of the preparation of monoclonal antibodies against uterine cervical cancer, turning then to the long-term culture of relevant hybridoma cells and transfectant cells in serum-free media. In this article, a hypothesis of "evolutionary antigen" is advanced as supported by the presence of a common antigen of human and mouse uterine cervical cancer. In addition, application of the monoclonal antibodies to clinical diagnosis is suggested while the use of these antibodies in the treatment of experimental cancer would seem feasible.

Isoprenaline enhances local Ca2+ release in cardiac myocytes

Aim:Contraction of cardiac myocytes is controlled by the generation and ampli-fication of intracellular Ca2+*** key step of this process is the couplingbetween sarcolemma L-type Ca2+channels（LCCs）and ryanodine receptors（RyRs）in the sarcoplasmic reticulum（SR）.β-Adrenergic stimulation is an important regu-latory mechanism for this coupling *** the details underlied the globallevel,which require local Ca2+release study are still *** present study isto explore the effects of β-adrenergic stimulation on local Ca2+***:Using confocal microscopy combined with loose-seal patch-clamp approaches,effects of isoprenaline(1 μmol·L-1),a β-adrenergic agonist,on local SR Ca2+re-lease triggered by Ca2+influx through LCCs in intact rat cardiac myocytes ***:Isoprenaline increased the intensity of ensemble averagedlocal Ca2+transients,the peak of which displayed a typical bell-shaped voltage-dependence over the membrane voltages ranging from～-40mV to～+35m ***-ther analysis showed that this enhancement could be explained by the increasedcoupling fidelity（which refers the increased probability of RyRs activation upondepolarization）,and the increased amplitude of evoked Ca2+sparks(due to moreCa2+releases through local RyRs).In addition,isoprenaline decreased the firstlatency,which displayed a typical"U"-shaped voltage-dependence,showing theavailable acceleration and synchronization of β-adrenergic stimulation on intrac-ellular calcium ***:Isoprenaline enhances local Ca2+release incardiac *** results underscore the importance of regulation of β-adrenergic stimulation on local intermolecular signals between LCCs and RyRs inheart cells.

On quasi-convex mappings of order α in the unit ball of a complex Banach space

In this paper, a class of biholomorphic mappings named quasi-convex mapping of order a in the unit ball of a complex Banach space is introduced. When the Banach space is confined to Cn, we obtain the relation between this class of mappings and the convex mappings. Furthermore, the growth and covering theorems of this class of mappings are given on the unit ball of a complex Banach space X. Finally, we get the second order terms coefficient estimations of the homogeneous expansion of quasi-convex mapping of order a defined on the polydisc in Cn and on the unit ball in a complex Banach space, respectively.

Phytoestrogen genistein decreases contractile response of aortic artery in vitro and arterial blood pressure in vivo

AIM: To determine the mechanisms of effects of phytoestrogen genistein on the contracted rabbit aortic arteries in vitro, and observe the effect of genistein and 17-β estradiol on mean arterial pressure (MAP) in ovariectomized (OVX) rats. METHODS: (1) Strips of rabbit aortic smooth muscle were suspended in organ baths containing Kreb’s solution, and then isometric tension was measured. (2) Female mature Wistar rats underwent a bilateral ovariectomy (OVX). Sham-operated rats (SHAM) were used as controls. After administration of genistein (0.4 mg·kg-1·d-1, sc), 17-β estradiol (1 mg·kg-1·d-1, sc) or theirvehicle sesame oil for 21 d,MAP was measured. RESULTS: (1) Similar to 17-β estradiol, genistein could dose-dependently relax 40 mmol/L KCl-precontracted arterial strips. Incubation with Nω-L-nitro-arginine (L-NNA), methylene blue (MB), indomethacin, propranolol or endothelium removal did not affect relaxation induced by genistein. In calcium-free solution containing 0.01mmol/L egtazic acid (EGTA), genistein inhibited not only the first phase contraction induced by noradrenaline (NA), but also the second contraction induced by CaCl2. In addition, genistein could reduce the contractile responses of NA, KCl and CaCl2, and shift their cumulative concentration-response curves rightward. (2) MAP in OVX rats was significantlyhigher compared with that of SHAM rats. However,after chronically treatment with genistein or 17-β estradiol for 21 d the baseline MAP in OVX rats was reduced significantly. CONCLUSIONS: (1) The vasodilator effect of genistein in vitro is endothelium independent and not related to the nitric oxide, its mechanisms being probably due to inhibition of Ca2+ influx through calcium channels in a noncompetitive manner and Ca2+ release from intracellular store induced by NA. (2) Administration of genistein or 17-β estradiol can chronically decrease MAP in OVX rats.

Changes in Autoantibodies against β1-Adrenoceptor and M2-Muscarinic Receptor during Development of Renovascular Hypertension in Rats

In an experimental rat’s renovascular hypertension model, we studied the genesis of anti- cardiac β1-adrenoceptor and M2-muscarinic receptor autoantibodies in relation to the changes in immunological function during the development of renal hypertension. The biological activities of these autoantibodies were also examined. It was shown that after two weeks of operation both the frequency of occurrence and the titre of autoantibodies to cardiac β1-adrenoceptor and M2-muscarinic receptor were significantly increased as compared with the control of pre-treatment. The increased autoantibodies lasted for several weeks and then automatically decreased gradually to the pre-clipping level at 10 weeks. Meanwhile the ratio of CD4 /CD8 + + was also undergone an initial increase followed by gradual recovery and correlated well with the changes in antibody titre. The biological effects of these autoantibodies displayed an “agonistic-like” activities on the beating frequency of cultured neonatal cardiomyocyte. It is suggested that autoimmune mechanisms are involved in the pathogenesis of renal hypertension and the cardiac receptor autoantibodies might be one of the mechanisms leading to cardiac dysfunction.

The mode of yearly biological cycle of malignant growth of hepatocellular carcinoma on computed tomography

OBJECTIVE: To detect the mode of yearly biological cycle of malignant biological behavior of hepatocellular carcinoma (HCC). METHODS: Twenty-one kinds of CT signs reflecting various degrees of malignant biological behavior were determined. A total of 360 patients were collected by random sampling of 30 patients each month. CT signs of each patient were fitted in corresponding group of yearly cyclic data respectively by cosine curves and analysed in terms of sequence characteristics (cosinor). RESULTS: With a 95% confidence, 10 CT signs showed biological rhythm (P20° from March to April. For low-invasive growth, however, the CT features were relatively low and concentrated between -180° and -270° from July to September. No acrophase was shown between -120° and -180° from May to June, and between -270° and -330° from October to November. CONCLUSION: Between CT signs of the highly-invasive and those of low-invasive growth of HCC, a sequential difference in biological cycles can be observed.
