A comparative molecular field analysis of cytotoxic beta-carboline analogs

AIM: To derive a model that could be used in drug design. METHODS: Beta-carbolines are reported to have antitumor activities on cultured cancer cell lines. A comparative molecular field analysis (CoMFA) was undertaken to elucidate the correlation of cytotoxities and structural parameters of 16 beta-carboline analogs (1-16). The compound 12 was finally used as a template for the other compounds in the dataset because of its highest biological activity. RESULTS: The CoMFA applied to the final alignment resulted in a qcv of 0.656 and it showed that the 2 steric fields contributed 43.3 % of the model information while the electrostatic fields represented the other 56.7 %. CONCLUSION: Three designed compounds, which were predicted to have high, moderate and low activities respectively, were synthesized. The IC50 values of these compounds indicated the significance of the analysis in this study. The model derived from the current study could be further used in design for more active compounds.

G-banding and karyotype analyses of cells in seventeen passages of SO-Rb_(50) during a long period of culture for about four years were performed. Three chromosome markers 13q14^-, 1p36^+ and 12p13^+ were found. Cells possessed 13q14^- reduced to zero after the 200th passage while 1p^+ and 12p^+ cells increased to 100% after 30 and 200 passages respectively. BACKGROUND:Liver enriched natural killer (NK) cells are of high immune ***,the function of donor liver NK cells in allogeneic liver transplantation (LTx) remains ***:Ten Gy of whole body gamma-irradiation (WBI) from a 60 Co source at 0.6 Gy/min was used for depleting donorderived leukocytes,and transfusion of purified liver NK cells isolated from the same type rat as donor (donor type liver NK cells,dtl NKs) through portal vein was performed immediately after grafting the irradiated ***-transplant survival observation on recipients and histopathological detection of liver grafts were adoptive to evaluate the biological impact of donor liver NK cells on recipients’ survival in rat ***:Transfusion of dtl NKs did not shorten the survival time among the recipients of spontaneous tolerance model (BN to LEW rat) after rat LTx,but prolonged the liver graft survival among the recipients depleted of donor-derived leukocytes in the acute rejection model (LEW to BN rat).Compared to the recipients in the groups which received the graft depleted of donor-derived leukocytes,better survival and less damage in the allografts were also found among the recipients in the two different strain combinations of liver allograft due to transfusion of dtl ***:Donor liver NK cells alone do not exacerbate liver allograft acute ***,they can alleviate it,and improve the recipients’ survival.

Background and Objective: Little is known about the incidence of hepatitis B virus (HBV) infection in Hodgkin's lymphoma patients. This study was to evaluate the impact of HBV infection on the survival of Hodgkin's lymphoma patient. Methods: Clinical data of 120 Hodgkin's lymphoma patients treated at the sun yat-sen university Cancer Center between January 2004 and October 2007 were collected. The impact of prognostic factors including HBV infection on survival was examined by univariate and multivariate analyses. A log-rank test was used for univariate analysis and the Cox proportional hazards regression model was used for multivariate analysis. Results: Of the 120 patients, 18 (15.0%) were hepatitis B virus surface antigen (HBsAg)-positive. The HBsAg-positive patients had lower 5-year survival rate than did the HBsAg-negative ones (66.9% vs. 91.3%, P = 0.006). When the patients were divided into early-stage (Ⅰ+Ⅱ) and advanced-stage (Ⅲ+Ⅳ) groups, the 5-year survival rate was significantly different between the HBsAg-positive and -negative patients in early-stage group (64.8% vs. 96.0%, P < 0.001), while not significantly different in advanced-stage group (75.0% vs. 84.8%, P=0.667). Both univariate and multivariate analyses showed that radiotherapy and HBV infection were independent prognosis factors for the patients with early-stage Hodgkin's lymphoma (P=0.006 and 0.014, respectively). Conclusions: The incidence of HBV infection is similar between Hodgkin's lymphoma patients and normal population. HBV infection is an independent prognosis factor for survival in the patients with early-stage Hodgkin's lymphoma.

PURPOSE: To investigate the expression status of p53, Fas and bcl-2 in the development of retinal degeneration in C3H, rd and rds mice. METHODS: Expression of p53, Fas and bcl-2 in the retina of rd, rds and C3H mice as well as in normal C3B mice in different periods was examined by immunohistochemical technique. RESULTS: The expression of Fas and p53 was not detected in normal C3B mice. Fas and p53 were expressed in the ganglionic layer in the early stage and then in the inner nuclear layer, while the scale and intense increased in the ganglionic layer. There is no difference of bcl-2 expression between the normal mice and mice with retinal degeneration. CONCLUSION: Fas and p53 may be involved in the retinal cell death in rd, rds, and C3B, but bcl-2 may not. It is unknown why p53 and Fas appeared first in ganglion layer but not in the outer nuclear layer where retinal cells death was noticed in early stage.

Aim: Hippocampal long-term potentiation (LTP), an activity-dependent plasticity in synaptic transmission, is believed as a fundamental mechanism of learning and memory. This form of plasticity also exists in spinal dorsal horn. Our previous work showed that electrical as well as natural noxious stimulation of afferent C-fibers or acute injury of the sciatic nerve produces LTP of C-fiber evoked field potentials in spinal dorsal horn.

Aim:To test the hypothesis that Cl~- channel blockers affect T cell proliferationthrough Ca2+-release-activated Ca2+(CRAC)signaling and examine the effects ofthe combination of a CRAC channel blocker and a Cl~- channel blocker on con-canavalin A(ConA;5 mg/mL)-induced Ca2+signaling,gene expression and cellu-lar proliferation in human peripheral T ***:[~3H]Thymidineincorporation,Fura-2 fluorescent probe,RNase protection assay,and reversetranscription-polymerase chain reaction were ***:The Cl~- channelblocker 4,4’-diisothiocyanostilbene-2,2’-disulfonic acid(DIDS)inhibited ConA-induced Ca2+influx,interleukin-2 mRNA expression and T lymphocyte prolifera-tion in a concentration-dependent manner,and also enhanced the inhibitory ef-fects of 1-{beta-[3-(4-methoxyphenyl)propoxyl]-4-methoxyphenethyl}-1H-imida-zole(SK&F96365)on the above key events during T cell activation.A combina-tion of DIDS(1 μmol/L)and SK&F96365(1 μmol/L)significantly diminished ConA-induced ClC-3 mRNA expression by 64%,whereas DIDS(1 μmol/L)or SK&F96365(1 μmol/L)alone decreased ConA-induced ClC-3 mRNA expression by only 16%and 9%,***:These results suggest that there is an interac-tion between CRAC-mediated Ca2+signaling and DIDS-sensitive Cl~- channelsduring ConA-induced T cell activation and ***,the DIDS-sensitive Cl~- channels may be related to the C1C-3 Cl~- channels.

The contrast sensitivity of 51 low vision patients (95 eyes) showed a substantial decrease in all spatial frequencies. The peak contrast sensitivity was shifted to 1 C/D from the normal 3-4 C/D. The high-frequency cut off is correlated to the visual acuity positively. Of the 33 patients, the preferred eye was the eye with better peak sensitivity in 28 patients (84%), while the preferred eye was the eye with better visual acuity in 22 patients (66%). Q-switched Nd:YAG laser was used for the peripheral iridotomy in 68 cases(80 eyes) of primary angle closed glaucoma. The average number of laser shots is 20.5(1～127 shots), and the average shot energy is 2.5 mJ(0.9～5.6mJ). The successful rate of iris penetration is about 97.6%.
