AIM： TO investigate the multicellular resistance of human hepatocellular carcinoma HepG2 cells in three-dimensional culture to delisheng, 5-fluorouracil and adriamycin, and the possible molecular mechanisms of delisheng. METHODS： Human hepatocellular carcinoma HepG2 cells were cultured with a liquid overlay technique. After the formation of multicellular spheroids, morphology was analyzed by phase contrast microscopy, scanning electron microscopy and transmission electron microscopy. Sensitivity of HepG2 cells to delisheng, 5-fluorouracil and adriamycin was investigated by Ml-I- assay in multicelluar spheroids and monolayers. Vascular endothelial growth factor （VEGF） and endostatin expression were analyzed in multicellular spheroids treated with delisheng, 5-fluorouracil, adriamycin and negative control PBS, with immunohistochemical staining. RESULTS： Multicellular spheroids exhibited structural characteristics somewhat different to those in monolayers. The cells in three-dimensional cell culture turned out to be less sensitive to delisheng, 5-fluorouracil and adriamycin than the cells cultured in monolayer. This showed that delisheng had a satisfactory cells inhibition ratio compared to 5-fluorouracil and adriamycin. Immunohistochemical staining showed that VEGF and endostatin expression was positive during growth as multicellular spheroids, and endostatin expression in spheroids with treatment of delisheng was higher than that with 5-fluorouracil, adriamycin and PBS （139.35 ± 7.83, 159.23 ± 10.34, 162.83 ± 3.47 and 148.48 ± 11.06, P 〈 0.05）.CONCLUSION： Chinese medicine compound delisheng has satisfactory anti-tumor activity in HepG2 cells in three-dimensional culture, and the effects are associated with up-regulation of endostatin.

Urban expansion is a phenomenon of urban space increase,and an important measuring index of the process of *** Shanghai as an example,the changes of urban average height and built-up area were studied to represent city's vertical and horizontal increases respectively,and statistical methods were used to analyze the driving forces of urban *** research drew following conclusions:1) The urban expansion process of Shanghai from 1985 to 2006 had a clear periodic feature,and could be divided into three stages:vertical expansion in dominance,coordinated vertical and horizontal expansion,and horizontal expansion in dominance.2) The average height and quantity of buildings in core city were significantly bigger than those in suburbs,but the changing speed of the latter was *** 3) urbanization process was the major driving force for the city's horizontal expansion,while industrial structure improvement was the key driving factor for the vertical *** two driving forces were simultaneously affected by city's political factors.

X-ray observations play a crucial role in time-domain *** Einstein Probe(EP),a recently launched X-ray astronomical satellite,emerges as a forefront player in the field of time-domain astronomy and high-energy *** a focus on systematic surveys in the soft X-ray band,EP aims to discover high-energy transients and monitor variable sources in the *** achieve these objectives,a quick and reliable classification of observed sources is *** this study,we developed a machine learning classifier for autonomous source classification using data from the EP-WXT Pathfinder—Lobster Eye Imager for Astronomy(LEIA)and EP-WXT *** proposed Random Forest classifier,built on selected features derived from light curves,energy spectra,and location information,achieves an accuracy of approximately 95%on EP simulation data and 98%on LEIA observational *** classifier is integrated into the LEIA data processing pipeline,serving as a tool for manual validation and rapid classification during *** paper presents an efficient method for the classification of X-ray sources based on single observations,along with implications of most effective features for the *** work facilitates rapid source classification for the EP mission and also provides valuable insights into feature selection and classification techniques for enhancing the efficiency and accuracy of X-ray source classification that can be adapted to other X-ray telescope data.

The effects of tourmaline on nitrogen removal performance and biofilm structures were comparatively investigated in two identical laboratory-scale sequencing batch biofilm reactors（SBBRs）（denoted SBBR1 and SBBR2） at different nitrogen loading rates（NLRs） varying from（0.24 ± 0.01） to（1.26 ± 0.02） g N/（L·day）. SBBR1 was operated in parallel with SBBR2, but SBBR1 was filled with polyurethane foam loaded tourmaline（TPU） carriers and another（SBBR2） filled with polyurethane foam（PU） carriers. Results obtained from this study showed that the excellent and stable performance of SBBR1 was obtained. Ammonia nitrogen removal and total nitrogen removal were higher in SBBR1 than that in SBBR2 with increase of NLR. At an NLR of（0.24 ± 0.01） g N/（L·day）, the majority of the spherical and elliptical bacteria were surrounded by the extracellular polymeric substance（EPS） and bacillus or filamentous bacteria in two SBBRs biofilms. When NLR increased to（1.26 ± 0.02） g N/（L·day）, the clusters were more obvious in the SBBR1 biofilm than that in the SBBR2 biofilm. Bacteria in SBBR1 were inclined to synthesis more EPS, and the formed EPS could protect the bacteria from free ammonia（FA） under extreme condition NLR（1.26 ± 0.02） g N/（L·day）. The results of polymerase chain reaction-denaturing gradient gel electrophoresis analysis showed that the microbial community similarity in SBBR2 decreased more obviously than that in SBBR1 with the increase of NLR, which the microbial community in SBBR1 was relatively stable.

Xuefu Zhuyu decoction has been used for treating traumatic brain injury and improving post-traumatic dysfunction, but its mechanism of action needs further investigation. This study established rat models of traumatic brain injury by controlled cortical impact. Rat models were intragastrically administered 9 and 18 g/kg Xuefu Zhuyu decoction once a day for 14 or 21 days. Changes in neurological function were assessed by modified neurological severity scores and the Morris water maze. Immunohistochemistry, western blot assay, and re- verse-transcription polymerase chain reaction were used to analyze synapsin protein and mRNA expression at the injury site of rats. Our results showed that Xuefu Zhuyu decoction visibly improved neurological function of rats with traumatic brain injury. These changes were accompanied by increased expression of synaptophysin, synapsin I, and postsynaptic density protein-95 protein and mRNA in a dose-de- pendent manner. These findings indicate that Xuefu Zhuyu decoction increases synapsin expression and improves neurological deficits alder traumatic brain injury.

CFD simulation on the generation of turbidites in deepwater areas: a case study of turbidity current processes in Qiongdongnan Basin, northern South China Sea

Turbidity currents represent a major agent for sediment transport in lakes, seas and oceans. In particu-lar, they formulate the most significant clastic accumulations in the deep sea, which become many of the world＇s most important hydrocarbon reservoirs. Several boreholes in the Qiongdongnan Basin, the north-western South China Sea, have recently revealed turbidity current deposits as significant hydrocarbon res-ervoirs. However, there are some arguments for the potential provenances. To solve this problem, it is es-sential to delineate their sedimentary processes as well as to evaluate their qualities as reservoir. Numerical simulations have been developed rapidly over the last several years, offering insights into turbidity current behaviors, as geologically significant turbidity currents are difficult to directly investigate due to their large scale and often destructive nature. Combined with the interpretation of the turbidity system based on high-resolution 3D seismic data, the paleotophography is acquired via a back-stripping seismic profile integrated with a borehole, i.e., Well A, in the western Qiongdongnan Basin; then a numerical model is built on the basis of this back-stripped profile. After defining the various turbidity current initial boundary conditions, includ-ing grain size, velocity and sediment concentration, the structures and behaviors of turbidity currents are investigated via numerical simulation software ANSYS FLUENT. Finally, the simulated turbidity deposits are compared with the interpreted sedimentary bodies based on 3D seismic data and the potential provenances of the revealed turbidites by Well A are discussed in details. The simulation results indicate that a sedimen-tary body develops far away from its source with an average grain size of 0.1 mm, i.e., sand-size sediment.
