Physiological and molecular responses to cold stress in rapeseed(Brassica napus L.)

Low temperature is one of the most important abiotic factors inhibiting growth, productivity, and distribution of rapeseed(Brassica napus L.). Therefore, it is important to identify and cultivate cold-tolerant germplasm. The objective of this study was to figure out the mechanism of chilling(4 and 2°C) and freezing(–2 and –4°C) stresses along with a control(22°C) in B. napus cultivars(1801 and C20) under controlled environment(growth chamber). The experiment was arranged in a complete randomized design with three replications. Our results exhibited that under chilling and freezing stresses, the increment of proline accumulation, soluble sugar and protein contents, and antioxidant enzyme activity were enhanced more in 1801 cultivar compared with C20 cultivar. At –2 and –4°C, the seedlings of C20 cultivar died completely compared with 1801 cultivar. Hydrogen peroxide(H2 O2) and malondialdehyde contents(MDA) increased in both cultivars, but when the temperature was decreased up to –2 and –4°C, the MDA and H2 O2 contents continuously dropped in 1801 cultivar. Moreover, we found that leaf abscisic acid(ABA) was enhanced in 1801 cultivar under chilling and freezing stresses. Additionally, the transcriptional regulations of cold-tolerant genes(COLD1, CBF4, COR6.6, COR15, and COR25) were also determined using real-time quantitative PCR(RT-q PCR). RT-q PCR showed that higher expression of these genes were found in 1801 as compared to C20 under cold stress(chilling and freezing stresses). Therefore, it is concluded from this experiment that 1801 cultivar has a higher ability to respond to cold stress(chilling and freezing stresses) by maintaining hormonal, antioxidative, and osmotic activity along with gene transcription process than C20. The result of this study will provide a solid foundation for understanding physiological and molecular mechanisms of cold stress signaling in rapeseed(B. napus).

Changes of Antioxidative Enzymes and Lipid Peroxidation in Leaves and Roots of Waterlogging-Tolerant and Waterlogging-Sensitive Maize Genotypes at Seedling Stage

To better understand the physiological and biochemical mechanisms of waterlogging tolerance, waterlogging effects on lipid peroxidation and the activity of antioxidative enzymes were investigated in leaves and roots of two maize genotypes, HZ32 （waterlogging-tolerant） and K12 （waterlogging-sensitive）. Potted maize plants were waterlogged at the second leaf stage under glasshouse conditions. Leaves and roots were harvested 1 d before and 2, 4, 6, 8 and 10 d after the start of waterlogging treatment. Through comparing the activities of superoxide dismutase （SOD）, ascorbate peroxidase （APX）, glutathione reductase （GR）, catalase （CAT） and guaiacol peroxidase （POD） between waterlogging-tolerant and waterloggingsensitive genotype, we deduced that CAT was the most important H2O2 scavenging enzyme in leaves, while APX seemed to play a key role in roots. POD, APX, GR and CAT activities in conjunction with SOD seem to play an essential protective role in the O2^- and H2O2 scavenging process. Lipid peroxidation was enhanced significantly only in K12 （P 〈 0.001） and there was no difference （P 〉 0.05） in HZ32 up to 6 d after waterlogging stress. These results indicated that oxidative stress may play an important role in waterlogging-stressed maize plants and that the greater protection of HZ32 leaves and roots from waterlogging-induced oxidative damage results, at least in part, through the maintenance of increased antioxidant enzyme activity.

Comparison of transcriptomes undergoing waterlogging at the seedling stage between tolerant and sensitive varieties of Brassica napus L.

RNA sequencing of the sensitive GH01 variety of Brassica napus L. seedling roots under 12 h of waterlogging was compared with previously published data of the ZS9 tolerant variety to unravel genetic mechanisms of waterlogging tolerance beyond natural variation. A total of 2 977 genes with similar expression patterns and 17 genes with opposite expression patterns were identiifed in the transcription proifles of ZS9 and GH01. An additional 1 438 genes in ZS9 and 1 861 genes in GH01 showed strain speciifc regulation. Analysis of the overlapped genes between ZS9 and GH01 revealed that waterlogging tolerance is determined by ability to regulate genes with similar expression patterns. Moreover, differences in both gene expression proifles and abscisic acid (ABA) contents between the two varieties suggest that ABA may play some role in waterlogging tolerance. This study identiifes a subset of candidate genes for further functional analysis.

Exogenous application of a low concentration of melatonin enhances salt tolerance in rapeseed(Brassica napus L.) seedlings

Melatonin is a naturally occurring compound in plants. Here, we tested the effect of exogenous melatonin on rapeseed（Brassica napus L.） grown under salt stress. Application of 30 μmol L^-1 melatonin alleviated salt-induced growth inhibition, and the shoot fresh weight, the shoot dry weight, the root fresh weight, and the root dry weight of seedlings treated with exogenous melatonin increased by 128.2, 142.9, 122.2, and 124.2%, respectively, compared to those under salt stress. In addition, several physiological parameters were evaluated. The activities of antioxidant enzymes including peroxidase（POD）, catalase（CAT） and ascorbate peroxidase（APX） were enhanced by 16.5, 19.3, and 14.2% compared to their activities in plants without exogenous melatonin application under salt stress, while the H2O2 content was decreased by 11.2% by exogenous melatonin. Furthermore, melatonin treatment promoted solute accumulation by increasing the contents of proline（26.8%）, soluble sugars（15.1%） and proteins（58.8%）. The results also suggested that higher concentrations（〉50 μmol L^-1） of melatonin could attenuate or even prevent the beneficial effects on seedling development. In conclusion, application of a low concentration of exogenous melatonin to rapeseed plants under salt stress can improve the H2O2-scavenging capacity by enhancing the activities of antioxidant enzymes such as POD, CAT and APX, and can also alleviate osmotic stress by promoting the accumulation of osmoregulatory substances such as soluble proteins, proline, and water soluble glucan. Ultimately, exogenous melatonin facilitates root development and improves the biomass of rapeseed seedlings grown under salt stress, thereby effectively alleviating the damage of salt stress in rapeseed seedlings.

Gradient microstructure and superior strength-ductility synergy of AZ61 magnesium alloy bars processed by radial forging with different deformation temperatures

Gradient microstructure modification is a cost-efficient strategy for high strength without compromising ductility,which is urgently needed in the fundamental science of engineering *** this study,heterogeneous structures of AZ61 alloy bars with anisotropic gradients(with different grain size distributions from the surface to the center)were observed to exhibit strong strength-ductility synergies under different deformation tem *** results reveal that the grain refinement process under mediumlow temperature deformation conditions(≤350℃)consists of four transition stages along the radial direction,i.e.,twin activations and deformation band formations,dislocation cells and pile-ups,ultrafine sub-grains,and randomly orientated quasi-micron *** deformation temperatures have a great influence on twin activations and deformation band formations,and the high temperature can easily provoke the initiation of non-basal *** deformation bands were determined as a primary nucleation site due to their highly unstable dislocation hindrance *** in combination with the Radial forging(RF)deformation process,the differences of dynamic precipitates can be attributed to microstructural difference and solubility limit of Al at different tem *** summarizing the tensile test results,the sample forged at 350℃exhibited the best strength-ductility synergy,exhibiting the highest elongation(EL)of 23.2%with a 251 MPa yield strength(YS)and 394 MPa ultimate tensile strength(UTS)in center region,and combined with the highest strength value of 256 MPa YS and 420 MPa UTS in the center region,while the EL was slightly degraded to 19.8%.

MgH_(2) is a promising high-capacity solid-state hydrogen storage material,while its application is greatly hindered by the high desorption temperature and sluggish ***,intertwined 2D oxygen vacancy-rich V_(2)O_(5) nanosheets(H-V_(2)O_(5))are specifically designed and used as catalysts to improve the hydrogen storage properties of MgH_(2).The as-prepared MgH_(2)-H-V_(2)O_(5) composites exhibit low desorption temperatures(Tonset=185℃)with a hydrogen capacity of 6.54 wt%,fast kinetics(Ea=84.55±1.37 kJ mol^(-1) H_(2) for desorption),and long cycling ***,hydrogen absorption can be achieved at a temperature as low as 30℃ with a capacity of 2.38 wt%within 60 ***,the composites maintain a capacity retention rate of~99%after 100 cycles at 275℃.Experimental studies and theoretical calculations demonstrate that the in-situ formed VH_(2)/V catalysts,unique 2D structure of H-V_(2)O_(5) nanosheets,and abundant oxygen vacancies positively contribute to the improved hydrogen sorption ***,the existence of oxygen vacancies plays a double role,which could not only directly accelerate the hydrogen ab/de-sorption rate of MgH_(2),but also indirectly affect the activity of the catalytic phase VH_(2)/V,thereby further boosting the hydrogen storage performance of MgH_(2).This work highlights an oxygen vacancy excited“hydrogen pump”effect of VH_(2)/V on the hydrogen sorption of Mg/MgH_(2).The strategy developed here may pave a new way toward the development of oxygen vacancy-rich transition metal oxides catalyzed hydride systems.

Economical Quadrupedal Multi-Gait Locomotion via Gait-Heuristic Reinforcement Learning

In order to strike a balance between achieving desired velocities and minimizing energy consumption,legged animals have the ability to adopt the appropriate gait pattern and seamlessly transition to another if *** ability makes them more versatile and efficient when traversing natural terrains,and more suitable for long *** the same way,it is meaningful and important for quadruped robots to master this *** achieve this goal,we propose an effective gait-heuristic reinforcement learning framework in which multiple gait locomotion and smooth gait transitions automatically emerge to reach target velocities while minimizing energy *** incorporate a novel trajectory generator with explicit gait information as a memory mechanism into the deep reinforcement learning *** allows the quadruped robot to adopt reliable and distinct gait patterns while benefiting from a warm start provided by the trajectory ***,we investigate the key factors contributing to the emergence of multiple gait *** tested our framework on a closed-chain quadruped robot and demonstrated that the robot can change its gait patterns,such as standing,walking,and trotting,to adopt the most energy-efficient gait at a given ***,we deploy our learned controller to a quadruped robot and demonstrate the energy efficiency and robustness of our method.

Voluntary wheel running ameliorated the deleterious effects of high-fat diet on glucose metabolism,gut microbiota and microbial-associated metabolites

Exercise training is critical for the early prevention and treatment of obesity and diabetes ***,the mechanism with gut microbiota and fecal metabolites underlying the effects of voluntary wheel running on high-fat diet induced abnormal glucose metabolism has not been fully elaborated.C57BL/6 male mice were randomly assigned to 4 groups according to diets(fed with normal chow diet or high-fat diet)and running paradigm(housed in static cage or with voluntary running wheel).An integrative 16S rDNA sequencing and metabolites profiling was synchronously performed to characterize the effects of voluntary wheel running on gut microbiota and *** showed that voluntary wheel running prevented the detrimental effects of high-fat feeding on glucose metabolism 16S rDNA sequencing showed remarkable changes in Rikenella and Marvinbryantia *** profiling indicated multiple altered metabolites,which were enriched in secondary bile acid biosynthesis *** conclusion,our study indicated that voluntary wheel running significantly improved glucose metabolism and counteracted the deleterious effects of high-fat feeding on body weight and glucose *** further found that voluntary wheel running could integratively program gut microbiota composition and fecal metabolites changes,and may regulate muricholic acid metabolism and secondary bile acid biosynthesis in high-fat fed mice.
