Platelet-derived microvesicles deliver miR-30e and promote VSMC apoptosis after balloon injury

During vascular remodeling after intimal injury,circulating platelets are activated by exposed collagen and release platelet-derived microvesicles(PMVs),which may contribute to the injury-induced apoptosis of vascular smooth muscle cells(VSMCs).However,the mechanisms in this process are still *** a rat balloon injury model,platelet adhesion at the injured site was detected,and promoted medial VSMC apoptosis was revealed with dT-mediated dUTP nick-end labeling(TUNEL)in *** apoptosis was promoted by coincubation with PMVs in *** analysis indicated the abound expression of the microRNA-30(miR-30)family in platelets,and the expression of the miR-30 family in platelets and PMVs was confirmed with *** Pathway Analysis(IPA)software identified transcription factor 2(RUNX2)as a pivotal molecule linking miR-30 and cellular *** vitro,PMVs delivered miR-30e,an important member of the miR-30 family,into VSMCs and increased cell apoptosis.A dual-luciferase assay was adopted to verify the interaction of miR-30e with RUNX2,and fluorescence in situ hybridization(FISH)proved the cytoplasmic localization of miR-30e in *** apoptotic effect on VSMCs was further confirmed by using miR-30e mimics and RUNX2 ***,RUNX2 siRNA altered the expression of its downstream genes,Tnfrsf11b,Smad6 and Mmp13,which may participate in apoptosis,as suggested by *** results indicated that PMVs play important roles in the interactions between circulating elements and VSMCs by delivering miR-30e to VSMCs and then promoting *** and potential intercellular messages may be novel therapeutic targets for preventing pathological vascular remodeling after intimal injury.

Aims Long-term heavy grazing reduces plant diversity and ecosystem function by intensifying nitrogen(N)and water *** contrast,the absence of biomass removal can cause species loss by elevating light competition and weakening community stability,which is exacerbated by N and water ***,how to maintain species diversity and community stability is still a huge challenge for sustainable management of worldwide *** We conducted a 4-year manipulated experiment in six long-term grazing blocks to explore combination of resource additions and biomass removal(increased water,N and light availability)on species richness and community stability in semiarid grasslands of Inner Mongolia,*** Findings In all blocks treated with the combination of resource additions and biomass removal,primary productivity increased and species richness and community stability were maintained over 4 years of *** both species and plant functional group(PFG)levels,the aboveground biomass of treated plants remained temporally stable in treatments with the combination of N and/or water addition and biomass *** maintenance of species richness was primarily caused by the biomass removal,which could increase the amount of light exposure for grasses under resource *** species asynchrony and stability of PFGs contributed to the high temporal stability observed in these *** results indicate that management practices of combined resource enrichment with biomass removal,such as grazing or mowing,could not only enhance primary productivity but also maintain plant species diversity,species asynchrony and community ***,as overgrazing-induced degradation and resource enrichment-induced biodiversity loss continue to be major problems worldwide,our findings have important implications for adaptive management in semiarid grasslands and beyond.

Prognostic role of neutrophil-lymphocyte ratio in operable esophageal squamous cell carcinoma

AIM:To explore the relationship of clinicopathological features and the distribution of neutrophils in the t umor mic roenvironment wi t h t he prognosis of ***:Two hundred and fifty-four formalin-fixed and paraffin embedded tissue blocks were analyzed, including tissues from cholangiocarcinoma(n = 254), and tumor adjacent tissues(n = 238).Tissue sections were stained for CD15 using immunohistochemical ***15 expression was detected to identify the distribution of neutrophils in the local tumor *** neutrophil density of the tumor tissues and the adjacent tumor tissues was detected to reflect their inflammatory *** data and follow-up information of cholangiocarcinoma patients who underwent surgery from January 2004 to December 2010 were analyzed *** relationship between clinicopathological features and the distribution of neutrophils with prognosis of the patients were ***:The positive expression level of CD15 was only significantly related to the TNM ***15 expression was higher in tumor tissues than in adjacent tissues(73.6% vs 54.6%), with significant *** with high expression of CD15 had significantly shorter overall survival(OS) than those with low expression of CD15(median overall survival time 39.77 mo vs 16.87 mo, P = 0.008).Patients with high CD15 expression had significantly shorter disease free survival time(DFS) than those with low expression of CD15(median DFS 38.27 mo vs 16.83 mo, P = 0.029).COX multivariate analysis indicated that high CD15 expression in tumor tissues was an independent risk factor for predicting OS for patients with cholangiocarcinoma [P = 0.012, relative risk(RR) = 1.601], but it was not an independent risk factor for predicting DFS(P = 0.073, RR = 1.462).CONCLUSION:Patients with high CD15 expression in cancer tissues had shorter DFS and *** expression of CD15 is an independent risk factor for OS.

Femtosecond laser processing stainless steel foil and its Fourier spectrum detection

In this paper, we use femtosecond laser pulse to scribe 304 stainless steel foil, detect the Fourier transform infrared spectrum of the sample before and after processing, confirm the "cold processing" and "thermal processing" and their mutual conversion, and determine the "cold processing" parameter window. The ablation threshold and incubation coefficient of 304 stainless steel foil are calculated, and the effects of scanning speed and effective pulse number on the ablation threshold are analyzed. The ANSYS software is used to simulate the radial and axial temperature distributions of the surface on 304 stainless steel foil sample and the heat-affected zone with a femtosecond laser fluence of 10 J/cm2 and an effective number of pulses of 1 200 are obtained. In the aspect of spectral detection, the Fourier transform infrared spectra of the sample before and after processing are measured and two processing mechanisms of "cold processing" and "hot processing" are confirmed, which proves that we can achieve the conversion between "cold processing" and "hot processing" by changing the laser fluence and determine the "cold processing" laser fluence range.

Effect of FeS doping on large diamond synthesis in FeNi-C system

The large single-crystal diamond with FeS doping along the （111） face is synthesized from the FeNi-C system by the temperature gradient method （TGM） under high-pressure and high-temperature （HPHT）. the effects of different FeS additive content on the shape, color, and quality of diamond are investigated. It is found that the （111） face of diamond is dominated and the （100） face of diamond disappears gradually with the increase of the FeS content. At the same time, the color of the diamond crystal changes from light yellow to gray-green and even gray-yellow. The stripes and pits corrosion on the diamond surface are observed to turn worse. The effects of FeS doping on the shape and surface morphology of diamond crystal are explained by the number of hang bonds in different surfaces of diamond. It can be shown from the test results of the Fourier transform infrared （FTIR） spectrum that there exists an S element in the obtained diamond. The N element content values in different additive amounts of diamond are calculated. The XPS spectrum results demonstrate that our obtained diamond contains S elements that exist in S-C and S-C-O forms in a diamond lattice. This work contributes to the further understanding and research of FeS-doped large single-crystal diamond characterization.
