Objective At present,a number of very severe aplastic anemia(VSAA)patients cannot receive hematopoietic stem cell transplantation(HSCT)or standard immunosuppressive therapy(IST)due to the high cost of therapy,shortage of sibling donors,and lack of resources to support the *** addition,some VSAA patients with autoantibodies have no life-threatening infections or bleeding at the time of initial *** the disease condition,economics and other factors,the present study designed a new and relatively mild treatment strategy:cyclosporine A plus pulsed high-dose prednisone(CsA+HDP).Methods The present study retrospectively analyzed 11 VSAA patients,who were treated with CsA+HDP in our hospital from August 2017 to August *** The median follow-up time for these patients was 24.9 *** overall response rate was 54.5%(6/11)at six months after the initiation of IST and 81.8%(9/11)at *** patients achieved complete remission and four patients met the criteria for partial response at the last *** median time to response for responders was 110 *** patients underwent HSCT due to the poor effect of CsA+HDP or to find a suitable transplant *** and clonal evolution were not found in any of these *** estimated 3-year overall survival rate and 3-year failure-free survival rate were 100.0%and 72.7%,*** addition,the results revealed that the cyclosporine-prednisone-associated toxicity was mild and well-tolerated by most *** The novel CsA+HDP regimen has good therapeutic effect and safety for VSAA patients with autoantibodies,who have no serious life-threatening infections or bleeding at the time of initial diagnosis.

Semicontinuous equal-channel angular extrusion（ SC-ECAE） is a novel severe plastic deformation technique that has been developed to produce ultrafine-grain steels. Instead of external forces being exerted on specimens in the conventional ECAE,driving forces are applied to dies in SC-EACE. The deformation of interstitial-free（ IF） steel w as performed at room temperature,and individual specimens w ere repeatedly processed at various passes. An overall grain size of 0. 55 μm w as achieved after 10 passes. During SC-ECAE,the main textures of IF steel included { 111} ,{ 110} ,{ 112} ,{ 110} ,and { 110} At an early stage,increasing dislocations induce new textures and increase intensity. When the deformation continues,low-angle boundaries are formed betw een dislocation cell bands,w hich cause some dislocation cell bands to change their orientation,and therefore,the intensity of the textures begins to decrease. After more passes,the intensity of textures continues to decrease w ith high-angle boundaries,and the sub-grains in dislocation cell bands continuously increase. The present study reports the evolution of textures during deformation; these w ere examined and characterized using high-resolution electron backscattered diffraction（ EBSD） in a field emission scanning electron microscope. The mechanisms of texture evolution are discussed.

Purpose-This study aims to research the large cross-section tunnel stability evaluation method corrected after considering the thickness-span ***/methodology/approach-First,taking the Liuyuan Tunnel of Huanggang-Huangmei High-Speed Railway as an example and taking deflection of the third principal stress of the surrounding rock at a vault after tunnel excavation as the criterion,the critical buried depth of the large section tunnel was ***,the strength reduction method was employed to calculate the tunnel safety factor under different rock classes and thickness-span ratios,and mathematical statistics was conducted to identify the relationships of the tunnel safety factor with the thickness-span ratio and the basic quality(BQ)index of the rock for different rock ***,the influences of thickness-span ratio,groundwater,initial stress of rock and structural attitude factors were considered to obtain the corrected BQ,based on which the stability of a large cross-section tunnel with a depth of more than 100 m during mechanized operation was *** evaluation method was then applied to Liuyuan Tunnel and Cimushan No.2 Tunnel of Chongqing Urban Expressway for ***-This study shows that under different rock classes,the tunnel safety factor is a strict power function of the thickness-span ratio,while a linear function of the BQ to some *** is more suitable to use the corrected BQ as a quantitative index to evaluate tunnel stability according to the actual conditions of the ***/value-The existing industry standards do not consider the influence of buried depth and span in the evaluation of tunnel *** stability evaluation method of large section tunnel considering the correction of overburden span ratio proposed in this paper achieves higher accuracy for the stability evaluation of surrounding rock in a full or large-section mechanized excavation of double line high-speed railway tunnels.

Multi-drug loaded vitamin E-TPGS nanoparticles for synergistic drug delivery to overcome drug resistance in tumor treatment

A facile self-assembly strategy to labricate vitamin E-tocopheryl polyethylene glycol succinate （TPGS） based nanoparticles with a high drug loading level for co-delivery of multiple drugs was developed. To overcome multi-drug resistance, a chemotherapeutic agent （paclitaxel, PTX） and a drug resistance inhibitor （tariq- uidar, TQR） were co-loaded in the vitamin E-TPGS based nanoparticle to obtain TPGS/PTX/TQR nanoparticles. The sizes of mono-drug loaded TPGS/PTX nanoparticles and dual-drug loaded TPGS/PTX/TQR nanoparticles were below 150 nm with relatively narrow size distributions. The efficiencies of the drug delivery systems on tumor cells inhibition and reversal of drug resistance were evaluated in drug resistant cancer cells （MCF-7/ADR） as compared with nonresistant cells （HeLa）. Due to the effective inhibition of the P-gp efflux by TQR, the dual-drug loaded nanoparticles exhibited significantly improved tumor cell inhibitory efficiency. To study the immunostimulatory effect, the effect of the drug delivery system on the production of IL- 10, a cytokine related to tumor development, in the drug resistant cells was determined. After the treatment by the drug delivery systems, the concentration of IL-10 in the cell supernatant decreased. The multi-drug loaded vitamin E-TPGS nanoparticles for synergistic drug delivery have promising applications in reversal of drug resistance in tumor treatments.

Objective To provide a set of useful analysis tools for the researchers to explore the microRNA data. Methods The R language was used for generating the Graphical Users Interface and implementing most functions. Some Practical Extraction and Report Language (Perl) scripts were used for parsing source files. Results We developed a graphical R package named miRE, which was designated for the analysis of microRNA functions, genomic organization, etc. This package provided effective and convenient tools for molecular biologists to deal with routine analyses in microRNA-related research. With its help, the users would be able to build a desktop- centered microRNA research environment quite easily and effectively. miRE is freely available at http://www. ***/~kanghu/WorkPresentation/miRE/***. A detailed user manual and tutorials with example code and image are also available. Conclusion miRE is a tool providing an open-source, user-friendly, integrated interface for microRNA-related analysis. With its help, researchers can perform microRNA-related analysis more efficiently.
