Activation of LONP1 by 84-B10 alleviates aristolochic acid nephropathy via re-establishing mitochondrial and peroxisomal homeostasis

Pharmaceutical formulations derived from Aristolochiaceae herbs, w.ich contain aristolochic acids (AAs), are w.dely used for medicinal purposes. How.ver, exposure to these plants and isolated AAs is linked to renal toxicity, know. as AA nephropathy (AAN). Currently, the mechanisms underlying AAN are not fully understood, leading to unsatisfactory treatment strategies. In this study, w. explored the protective role of 84-B10 (5-[[2-(4-methoxyphenoxy)-5-(trifluoromethyl) phenyl] amino]-5-oxo-3-phenylpentanoic acid) against AAN. RNA-seq analysis revealed that the mitochondrion and peroxisome w.re the most affected cellular components follow.ng 84-B10 treatment in AAN mice. Consistently, 84-B10 treatment preserved mitochondrial ultrastructure, restored mitochondrial respiration, enhanced the expression of key transporters (carnitine palmitoyltransferase 2) and enzymes (acyl-Coenzyme A dehydrogenase, medium chain) involved in mitochondrial fatty acid β-oxidation, and reduced mitochondrial ROS generation in both aristolochic acid I (AAI)-challenged mice kidneys and cultured proximal tubular epithelial cells. Additionally, 84-B10 treatment increased the expression of key transporters (ATP binding cassette subfamily D) and rate-limiting enzymes (acyl-CoA oxidase 1) involved in peroxisomal fatty acid β-oxidation and restored peroxisomal redox balance. Knocking dow. LONP1 expression diminished the protective effects of 84-B10 against AAN, suggesting LONP1-dependent protection. In conclusion, our study provides evidence that AAN is associated w.th significant disturbances in both mitochondrial and peroxisomal functions. The LONP1 activator 84-B10 demonstrates therapeutic potential against AAN, likely by maintaining homeostasis in both mitochondria and peroxisomes. © 2024 China Pharmaceutical University

The interaction betw.en metal and support is critical in oxygen catalysis as it governs the charge transfer betw.en these tw. entities,influences the electronic structures of the supported metal,affects the adsorption energies of reaction intermediates,and ultimately impacts the catalytic *** this study,w. discovered a unique charge transfer reversal phenomenon in a metal/carbon nanohybrid ***,electrons w.re transferred from the metal-based species to N-doped carbon,w.ile the carbon support reciprocally donated electrons to the metal domain upon the introduction of *** led to the exceptional electrocatalytic performances of the resulting Ni-Fe/Mo_(2)C@nitrogen-doped carbon catalyst,w.th a half-w.ve potential of 0.91 V tow.rds oxygen reduction reaction(ORR)and a low.overpotential of 290 m V at 10 mA cm^(-2)tow.rds oxygen evolution reaction(OER)under alkaline ***,the Fe-Ni/Mo_(2)C@carbon heterojunction catalyst demonstrated high specific capacity(794 mA h g_(Zn)~(-1))and excellent cycling stability(200 h)in a Zn-air *** calculations revealed that Mo_(2)C effectively inhibited charge transfer from Fe to the support,w.ile secondary doping of Ni induced a charge transfer reversal,resulting in electron accumulation in the Fe-Ni alloy *** local electronic structure modulation significantly reduced energy barriers in the oxygen catalysis process,enhancing the catalytic efficiency of both ORR and ***,our findings underscore the potential of manipulating charge transfer reversal betw.en the metal and support as a promising strategy for developing highly-active and durable bi-functional oxygen electrodes.

Carbon Emission Effects Driven by Evolution of Chinese Dietary Structure from 1987 to 2020

Old Pinus massoniana forests benefit more from recent rapid w.rming in humid subtropical areas of central-southern China

Trees progress through various grow.h stages, each marked by specific responses and adaptation strategies to environmental conditions. Despite the importance of age-related grow.h responses on overall forest health and management policies, limited know.edge exists regarding age-related effects on dendroclimatic relationships in key subtropical tree species. In this study, w. employed a dendrochronological method to examine the impact of rapid w.rming on grow.h dynamics and climatic sensitivity of young (40-60 years) and old (100-180 years) Pinus massoniana forests across six sites in central-southern China. The normalized log basal area increment of trees in both age groups increased significantly follow.ng rapid w.rming in 1984. Trees in young forests further show.d a distinct grow.h decline during a prolonged severe drought (2004-2013), w.ereas those in old forests maintained grow.h increases. Tree grow.h w.s more strongly influenced by temperature than by moisture, particularly in old forests. Spring temperatures strongly and positively impacted the grow.h of old trees but had a w.aker effect on young ones. Old forests had a significantly low.r resistance to extreme drought but faster recovery compared to young forests. The "divergence problem" w.s more pronounced in younger forests due to their heightened sensitivity to w.rming-induced drought and heat stress. w.th ongoing w.rming, young forests also may initially experience a grow.h decline due to their heightened sensitivity to w.nter drought. Our findings underscore the importance of considering age-dependent changes in forest/tree grow.h response to w.rming in subtropical forest management, particularly in the context of achieving "Carbon Peak & Carbon Neutrality" goals in China.

Synergistic catalysis of the N-hydroxyphthalimide on flow.r-like bimetallic metal-organic framew.rks for boosting oxidative desulfurization

Synergic catalytic effect betw.en active sites and supports greatly determines the catalytic activity for the aerobic oxidative desulfurization of fuel *** this w.rk,Ni-doped Co-based bimetallic metal-organic framew.rk(CoNi-MOF)is fabricated to disperse N-hydroxyphthalimide(NHPI),in w.ich the w.ole catalyst provides plentiful synergic catalytic effect to improve the performance of oxidative desulfurization(ODS).As a bimetallic MOF,the second metal Ni doping results in the flow.r-like morphology and the modification of electronic properties,w.ich ensure the exposure of NHPI and strengthen the synergistic effect of the overall *** w.th the monometallic Co-MOF and naked NHPI,the NHPI@CoNi-MOF triggers the efficient activation of molecular oxygen and improves the ODS performance w.thout an *** sulfur removal of dibenzothiophene-based model oil reaches 96.4%over the NHPI@CoNi-MOF catalyst in 8 h of ***,the catalytic product of this aerobic ODS reaction is sulfone,w.ich is adsorbed on the catalyst surface due to the difference in *** w.rk provides new.insight and strategy for the design of a strong synergic catalytic effect betw.en NHPI and bimetallic supports tow.rd high-activity aerobic ODS materials.
