Self-Protection against Triptolide-Induced Toxicity in Human Hepatic Cells via Nrf2-ARE-NQO1 Pathway

Objective: To find the signaling pathway of triptolide(TP)-induced liver injury and to reveal whether NF-E2-related factor 2(Nrf2) plays an important role in cellular self-protection. Methods: The L-02 and HepG2 cells were cultured and treated with various concentrations of TP. The cell viability was observed, and the cell medium was collected for detecting the aspartate aminotransferase(ALT), alanine aminotransferase(AST), lactate dehydrogenase(LDH), superoxide dismutase(SOD) and L-glutathione production(GSH) levels. Nrf2 and its downstream target NAD(P)H: quinine oxidoreductase 1(NQO1) and heme oxygenase-1(HO-1) expression, the nuclear translocation of Nrf2, and the binding ability of Nrf2 and antioxidant response element(ARE) were also identified. Meanwhile,shRNA was used to silence Nrf2 in L-02 cells to find out whether Nrf2 plays a protective role. Results: The viability of the L-02 and HepG2 cells treated with TP decreased in a doseand time-dependent manner, and TP(20–80 μg/mL) markedly induced the release of ALT, AST and LDH(P<0.05 or P<0.01), reduced the levels of SOD and GSH(P<0.01), and increased the intracellular reactive oxygen species. Meanwhile, TP augmented the Nrf2 expression in L-02 and HepG2 cells(P<0.05 or P<0.01), induced Nrf2 nuclear translocation, increased the Nrf2 ARE binding activity, and increased HO-1 and NQO1 expressions. Nrf2 knockdown revealed a more severe toxic effect of TP(P<0.05 or P<0.01). Conclusions: Human hepatic cells treated with TP induced oxidative stress, and led to cytotoxicity. Self-protection against TP-induced toxicity in human hepatic cells might be via Nrf2-ARE-NQO1 transcriptional pathway.

Inference for High-Dimensional Streamed Longitudinal Data

With the advent of modern devices, such as smartphones and wearable devices, highdimensional data are collected on many participants for a period of time or even in perpetuity. For this type of data, dependencies between and within data batches exist because data are collected from the same individual over time. Under the framework of streamed data, individual historical data are not available due to the storage and computation burden. It is urgent to develop computationally efficient methods with statistical guarantees to analyze high-dimensional streamed data and make reliable inferences in practice. In addition, the homogeneity assumption on the model parameters may not be valid in practice over time. To address the above issues, in this paper, we develop a new renewable debiased-lasso inference method for high-dimensional streamed data allowing dependences between and within data batches to exist and model parameters to gradually change. We establish the large sample properties of the proposed estimators, including consistency and asymptotic *** numerical results, including simulations and real data analysis, show the superior performance of the proposed method.

Tumor suppressor p53-mediated cell cycle arrest and DNA damage repair may exert cytoprotective effects against cancer therapies,including WEE1 *** that p53 activation can also lead to multiple types of cell death,the role of this tumor suppressor in WEE1 inhibitor-based therapies remains *** this study,we reported that nucleolar stress-mediated p53 activation enhanced the WEE1 inhibitor AZD1775-induced ferroptosis to suppress lung cancer *** findings showed that AZD1775 promoted ferroptosis by blocking cystine uptake,an action similar to that of ***,inhibition of WEE1 by the WEE1 inhibitors or siRNAs induced compensatory upregulation of SLC7A11,which conferred resistance to ***,AZD1775 prevented the enrichment of H3K9me3,a histone marker of transcriptional repression,on the SLC7A11 promoter by repressing the expression of the histone methyltransferase SETDB1,thereby enhancing NRF2-mediated SLC7A11 *** finding was also validated using the H3K9me3 inhibitor ***,we found that the nucleolar stress-inducing agent Actinomycin D(Act.D)inhibited SLC7A11 expression by activating p53,thus augmenting AZD1775-induced ***,the combination of AZD1775 and Act.D synergistically suppressed wild-type p53-harboring lung cancer cell growth both in vitro and in ***,our study demonstrates that AZD1775 promotes ferroptosis by targeting cystine uptake but also mediates the adaptive activation of SLC7A11 through the WEE1-SETDB1 cascade and NRF2-induced transcription,and inhibition of SLC7A11 by Act.D boosts the anti-tumor efficacy of AZD1775 by enhancing ferroptosis in cancers with wild-type p53.

日本血吸虫病是在我国广为流传的传染病和寄生虫病,对人体的健康造成了极其严重的危害,关于日本血吸虫病的传播动力学模型引起了广泛的讨论。在吴建宏等建立的双宿主日本血吸虫病的自治动力学模型的基础上,考虑到湖泊型地区钉螺数量的季节性变化因素,本文考察了相应的非自治的传播动力学模型,研究了其周期解的稳定性,并在此基础上进行了数值模拟。研究表明,在一定的参数条件下,无论开始时疾病传播情况如何,疾病终将趋于消亡;否则,在一定的初始条件下,疾病传播形成周期性的地方病。数值模拟发现,在一定的参数条件下,钉螺数量的季节性变化振幅充分大时,可使疾病趋于消亡;此外,同时对患病的人与牛进行治疗,也有利于使疾病消亡。本文中还研究了Barbour双宿主模型的非自治动力学模型,不仅对其周期解的稳定性进行了讨论,还得到该系统周期解的存在性条件。

Spatial differentiation of carbon emissions from energy consumption based on machine learning algorithm:A case study during 2015–2020 in Shaanxi,China

Carbon emissions resulting from energy consumption have become a pressing issue for governments *** estimation of carbon emissions using satellite remote sensing data has become a crucial research *** studies relied on statistical regression models that failed to capture the complex nonlinear relationships between carbon emissions and characteristic *** this study,we propose a machine learning algorithm for carbon emissions,a Bayesian optimized XGboost regression model,using multi-year energy carbon emission data and nighttime lights(NTL)remote sensing data from Shaanxi Province,*** results demonstrate that the XGboost algorithm outperforms linear regression and four other machine learning models,with an R^(2)of 0.906 and RMSE of *** observe an annual increase in carbon emissions,with high-emission counties primarily concentrated in northern and central Shaanxi Province,displaying a shift from discrete,sporadic points to contiguous,extended spatial *** autocorrelation clustering reveals predominantly high-high and low-low clustering patterns,with economically developed counties showing high-emission clustering and economically relatively backward counties displaying low-emission *** findings show that the use of NTL data and the XGboost algorithm can estimate and predict carbon emissionsmore accurately and provide a complementary reference for satellite remote sensing image data to serve carbon emission monitoring and *** research provides an important theoretical basis for formulating practical carbon emission reduction policies and contributes to the development of techniques for accurate carbon emission estimation using remote sensing data.

High-dimensional large-scale mixed-type data imputation under missing at random

Missingness in mixed-type variables is commonly encountered in a variety of areas. The requirement of complete observations necessities data imputation when a moderate or large proportion of data is ***, inappropriate imputation would downgrade the performance of machine learning algorithms, leading to bad predictions and unreliable statistical inference. For high-dimensional large-scale mixed-type missing data, we develop a computationally efficient imputation method, missing value imputation via generalized factor models（MIG）, under missing at random. The proposed MIG method allows missing variables to be of different types,including continuous, binary, and count variables, and are scalable to both data size n and variable dimension p while existing imputation methods rely on restrictive assumptions such as the same type of missing variables,the low dimensionality of variables, and a limited sample size. We explicitly show that the imputation error of the proposed MIG method diminishes to zero with the rate Op(max{n-1/2, p-1/2}) as both n and p tend to infinity. Five real datasets demonstrate the superior empirical performance of the proposed MIG method over existing methods that the average normalized absolute imputation error is reduced by 5.3%–34.1%.
