The crosstalk between clathrin-mediated endocytosis(CME)and the autophagy pathway has been reported in mammals;however,the interconnection of CME with autophagy has not been established in ***,we report that the Arabidopsis CLATHRIN LIGHT CHAIN(CLC)subunit 2 and 3 double mutant,clc2-1 clc3-1,phenocopies Arabidopsis AUTOPHAGY-RELATED GENE(ATG)mutants in both autoimmunity and nutrient ***,the autophagy pathway is significantly compromised in the clc2-1 clc3-1 ***,multiple assays demonstrate that CLC2 directly interacts with ATG8h/ATG8i in a domain-specific *** expected,both GFP-ATG8h/GFP-ATG8i and CLC2-GFP are subjected to autophagic degradation,and degradation of GFP-ATG8h is significantly reduced in the clc2-1 clc3-1 ***,simultaneous knockout of ATG8h and ATG8i by CRISPR-Cas9 results in enhanced resistance against Golovinomyces cichoracearum,supporting the functional relevance of the CLC2-ATG8h/8i *** conclusion,our results reveal a link between the function of CLCs and the autophagy pathway in Arabidopsis.

Thermal crown of roll is an important factor, which affects strip profile. It is necessary to analyze the temperature field and thermal crown of roll for hot strip mill. A new simplified finite element method （FEM） was used to analyze the temperature field and thermal crown of roll, and corresponding models were built according to the practical boundary conditions. Transient roll temperature field and thermal crown were simulated by ANSYS FEM software with considering transient thermal contact and complex boundary condition. Temperature and thermal crown variations on roll surface nodes were obtained. The thermal crown results of roll obtained by FEM simulation were in good agreement with the measured data, indicating that simplified FEM models and results were correct.

Effects of stress ratio on the temperature-dependent high-cycle fatigue properties of alloy steels

This paper addresses the effects of stress ratio on the temperature-dependent high-cycle fatigue （HCF） properties of alloy steels 2CrMo and 9CrCo, which suffer from substantial vibrational loading at small stress amplitude, high stress ratio, and high frequency in the high-temperature environments in which they fimcfion as blade and rotor spindle materials in advanced gas or steam turbine engines. Fatigue tests were performed on alloy steels 2CrMo and 9CrCo subjected to constant-amplitude loading at four stress ratios and at four and three temperatures, respectively, to determine their temperature-dependent HCF properties. The interaction mechanisms between high temperature and stress ratio were deduced and compared with each other on the basis of the results of fractographic analysis. A phenomenological model was developed to evaluate the effects of stress ratio on the temperature-dependent HCF properties of alloy steels 2CrMo and 9CrCo. Good correlation was achieved between the predictions and actual experiments, demonstrating the practical and effective use of the proposed method.

The wood sorrel family,Oxalidaceae,is mainly composed of annual or perennial herbs,a few shrubs,and trees distributed from temperate to tropical *** of Oxalidaceae are of high medicinal,ornamental,and economic *** the rich diversity and value of Oxalidaceae,few molecular markers or plastomes are available for phylogenetic analysis of the ***,we reported four new whole plastomes of Oxalidaceae and compared them with plastomes of three species in the family,as well as the plastome of Rourea microphylla in the closely related family *** eight plastomes ranged in length from 150,673 bp(Biophytum sensitivum)to 156,609 bp(***).Genome annotations revealed a total of 129e131 genes,including 83e84 protein-coding genes,eight rRNA genes,37 tRNA genes,and two to three *** analyses showed that the plastomes of these species have minor variations at the gene *** smaller plastomes of herbs *** and three Oxalis species are associated with variations in IR region sizes,intergenic region variation,and gene or intron *** identified sequences with high variation that may serve as molecular markers in taxonomic studies of *** phylogenetic trees of selected superrosid representatives based on 76 protein-coding genes corroborated the Oxalidaceae position in Oxalidales and supported it as a sister to *** research also supported the monophyly of the COM(Celastrales,Oxalidales,and Malpighiales)clade.

α-Pinene is an important monoterpene,which is widely used as a flavoring agent and in fragrances,pharmaceuticals and *** an evolved strain Escherichia coli YZFP,which had higher tolerance to pinene and titer,has been successfully used to produce high levels of pinene,the pinene titer is much lower than that of hemiterpene(isoprene)and sesquiterpenes(farnesene)to ***,the overall cellular physiological and metabolic changes caused by higher tolerance to pinene and overproduction of pinene remains *** reveal the mechanism of Escherichia coli YZFP with the higher tolerance to pinene and titer,a comparative genomics and transcriptional level analyses combining with CRISPR activation(CRISPRa)and interference(CRISPRi)were carried *** results show that the tolerance to pinene and the overproduction of pinene in *** may be associated with:1)the mutations of the DXP pathway genes,the rpoA and some membrane protein genes,and their upregulations of transcription levels;and 2)the mutations of some genes and their downregulation of transcriptional *** comparative omics analyses provided some genetic modification strategies to further improve pinene *** of the mutated cbpA,tabA,pitA,rpoA,sufBCDS,mutS,ispH,oppF,dusB,dnaK,dxs,dxr and flgFGH genes further improved pinene *** study also demonstrated that combining comparative omics analysis with CRISPRa and CRISPRi is an efficient technology to quickly find a new metabolic engineering strategy.
