p38 Signaling Mediates Naringin-Induced Osteogenic Differentiation of Porcine Metanephric Mesenchymal Cells

Objective:To explore the potential of metanephric mesenchymal cells(MMCs)for osteogenesis and naringin’s ability to enhance this process and its molecular ***:Porcine MMCs at 70 days of gestation were used as tool cells,cultured in osteogenic induction medium,identified by immunocytochemistry *** potential of porcine MMCs and naringin’s ability to enhance this process was tested by detecting changes in cell viability,alkaline phosphatase(ALP)activity,the expression of runt-related transcription factor 2(Runx2),osteopontin(OPN)and osteocalcin(OCN),and the formation of mineralized nodules,and the application of the p38 signaling pathway inhibitor SB203580 vitiated the osteogenesis-promoting effect of ***:Immunocytochemical staining showed that the cells were Vimentin and Six2(+),E-cadherin and CK-18(−).Naringin can activate the p38 signaling pathway to enhance the osteogenesis of porcine MMCs by increasing cell viability,ALP activity,the expressions of Runx2,OPN and OCN,and the formation of mineralized nodules(P<0.05).The application of p38 signaling pathway inhibitor SB203580 vitiated the osteogenesis-promoting effect of naringin,manifested by decreased ALP activity,the expressions of Runx2,OPN and OCN,and the formation of mineralized nodules(P<0.05).Conclusion:Naringin,the active ingredient of Chinese herbal medicine Rhizoma Drynariae for nourishing Shen(Kidney)and strengthening bone,enhances the osteogenic differentiation of renal MMCs through the p38 signaling pathway.

Power quality improvements help guide and solve the problems of inefficient energy production and unstable power output in wind power *** purpose of this paper is mainly to explore the influence of different energy storage batteries on various power quality indicators by adding different energy storage devices to the simulated wind power system,and to explore the correlation between systementropy generation and various indicators,so as to provide a theoretical basis for directly improving power quality by reducing loss.A steady-state experiment was performed by replacing the wind wheel with an electric motor,and the output power qualities of the wind power systemwith andwithout energy storagewere compared and ***,the improvement effect of different energy storage devices on various indicatorswas ***,based on the entropy theory,the loss of the internal components of the wind power system generator is simulated and explored by Ansys *** the analysis of power quality evaluation indicators,such as current harmonic distortion rate,frequency deviation rate,and voltage fluctuation,the correlation between entropy production and each evaluation indicator was explored to investigate effective methods to improve power quality by reducing entropy *** results showed that the current harmonic distortion rate,voltage fluctuation,voltage deviation,and system entropy production are positively correlated in the tests and that the power factor is negatively correlated with system entropy *** the frequency range,the frequency deviationwas not significantly correlated with the systementropy production.

Deep oil and gas reservoirs are under high-temperature conditions, but traditional coring methods do not consider temperature-preserved measures and ignore the influence of temperature on rock porosity and permeability, resulting in distorted resource assessments. The development of in situ temperaturepreserved coring(ITP-Coring) technology for deep reservoir rock is urgent, and thermal insulation materials are key. Therefore, hollow glass microsphere/epoxy resin thermal insulation materials(HGM/EP materials) were proposed as thermal insulation materials. The materials properties under coupled hightemperature and high-pressure(HTHP) conditions were tested. The results indicated that high pressures led to HGM destruction and that the materials water absorption significantly increased; additionally,increasing temperature accelerated the process. High temperatures directly caused the thermal conductivity of the materials to increase; additionally, the thermal conduction and convection of water caused by high pressures led to an exponential increase in the thermal conductivity. High temperatures weakened the matrix, and high pressures destroyed the HGM, which resulted in a decrease in the tensile mechanical properties of the materials. The materials entered the high elastic state at 150.C, and the mechanical properties were weakened more obviously, while the pressure led to a significant effect when the water absorption was above 10%. Meanwhile, the tensile strength/strain were 13.62 MPa/1.3%and 6.09 MPa/0.86% at 100.C and 100 MPa, respectively, which meet the application requirements of the self-designed coring device. Finally, K46-f40 and K46-f50 HGM/EP materials were proven to be suitable for ITP-Coring under coupled conditions below 100.C and 100 MPa. Launching and recovering human-occupied vehicles(HOVs)has always been a challenging *** current recovery process requires staff to manually complete the tethering task,which is inefficient and endangers the lives of *** paper suggests moving the recovery position from the surface to underwater at approximately half the wavelength of the water depth(30−50 m underwater),where the HOV experiences less environmental *** ROV equipped with a ultra-short baseline beacon(USBL)and a manipulator was used to complete the tethering ***,a shackle customized to the shape of the manipulator’s gripper is fitted to the end of the cable to simplify the tethering *** investigate the dynamic response of recovering the HOV using this suggested method,a comprehensive numerical model is developed in this *** effects of wind,surface waves,ocean currents,and nonlinear interaction between the installation vessel and the HOV are quantitatively *** results show that the proposed recovery method can reduce the motion amplitude of the HOV and that the wave has the greatest influence on the dynamic response of the HOV during the recovery *** model provides better insight into the proposed HOV recovery method and confirms the effectiveness of the heave compensation *** proposed approach aims to enhance safety and operational efficiency by reducing direct human involvement in the recovery process and mitigating potential *** finding holds particular significance,especially in environmentally sensitive areas,where reducing the impact on the surrounding ecosystem is crucial.

The gut microbiome interacts with the host to maintain body homeostasis,with gut microbial dysbiosis implicated in many ***,the underlying mechanisms of gut microbe regulation of host behavior and brain functions remain *** study aimed to elucidate the influence of gut microbiota on brain functions via post-translational modification mechanisms in the presence or absence of bacteria without any *** conducted succinylome analysis of hippocampal proteins in germ-free(GF)and specific pathogen-free(SPF)mice and metagenomic analysis of feces from SPF *** results were integrated with previously reported hippocampal acetylome and phosphorylome data from the same batch of *** bioinformatics analyses revealed 584 succinylation sites on 455 proteins,including 54 up-regulated succinylation sites on 91 proteins and 99 down-regulated sites on 51 proteins in the GF mice compared to the SPF *** constructed a panoramic map of gut microbiota-regulated succinylation,acetylation,and phosphorylation,and identified cross-talk and relative independence between the different types of post-translational modifications in modulating complicated intracellular *** correlation analysis indicated that 13 taxa,predominantly belonging to the Bacteroidetes phylum,were correlated with the biological functions of post-translational *** correlations between these taxa and succinylation and negative correlations between these taxa and acetylation were identified in the modulation of intracellular *** study highlights the hippocampal physiological changes induced by the absence of gut microbiota,and proteomic quantification of succinylation,phosphorylation,and acetylation,contributing to our understanding of the role of the gut microbiome in brain function and behavioral phenotypes.
