Experimental investigation on shear strength deterioration at the interface between different rock types under cyclic loading

The shear strength deterioration of bedding planes between different rock types induced by cyclic loading is vital to reasonably evaluate the stability of soft and hard interbedded bedding rock slopes under earthquake;however,rare work has been devoted to this subject due to lack of *** this study,experimental investigations on shear strength weakening of discontinuities with different joint wall material(DDJM)under cyclic loading were conducted by taking the interface between siltstone and mudstone in the Shaba slope of Yunnan Province,China as research objects.A total of 99 pairs of similar material samples of DDJM(81 pairs)and discontinuities with identical joint wall material(DIJM)(18 pairs)were fabricated by inserting plates,engraved with typical surface morphology obtained by performing three-dimensional laser scanning on natural DDJMs sampled from field,into mold *** shear tests were conducted on these samples to study their shear strength changes with the cyclic number considering the effects of normal stress,joint surface morphology,shear displacement amplitude and shear *** results indicate that the shear stress *** displacement curves under each shear cycle and the peak shear strength *** number curves of the studied DDJMs are between those of DIJMs with siltstone and mudstone,while closer to those of DIJMs with *** peak shear strengths of DDJMs exhibit an initial rapid decline followed by a gradual decrease with the cyclic number and the decrease rate varies from 6%to 55.9%for samples with varied surface morphology under different testing *** normal stress,joint surface morphology,shear displacement amplitude and shear rate collectively influence the shear strength deterioration of DDJM under cyclic shear loading,with the degree of influence being greater for larger normal stress,rougher surface morphology,larger shear displacement amplitude and faster shear rate.

Fibrosis is a necessary process in the progression of chronic disease to cirrhosis or even cancer,which is a serious disease threatening human *** studies have shown that the early treatment of fibrosis is turning point and particularly ***,how to reverse fibrosis has become the focus and research hotspot in recent *** far,the considerable progress has been made in the development of effective anti-fibrosis drugs and targeted drug ***,the existing research results will lay the foundation for more breakthrough delivery systems to achieve better anti-fibrosis ***,this review summaries anti-fibrosis delivery systems focused on three major organ fibrotic diseases such as liver,pulmonary,and renal fibrosis accompanied by the elaboration of relevant pathological mechanisms,which will provide inspiration and guidance for the design of fibrosis drugs and therapeutic systems in the future.

Myocardial ischemia is a serious threat to human health,and vascular dysfunction is its main *** Tongyu(BXTY)Granule is an effective traditional Chinese medicine(TCM)for treating myocardial ***,the underlying mechanism of BXTY is still *** this study,we demonstrate that BXTY ameliorates myocardial ischemia by activating the soluble guanylate cyclase(sGC)-30,50-cyclic guanosine monophosphate(cGMP)-protein kinase G(PKG)signaling pathway in vascular smooth muscle cells(VSMCs)to dilate the *** was given by gavage for ten consecutive days before establishing an animal model of acute myocardial ischemia in mice via the intraperitoneal injection of *** results showed that BXTY alleviated the symptoms of myocardial ischemia induced by pituitrin in mice,including electrocardiogram abnormalities and changes in plasma *** addition,BXTY dilated pre-constricted blood vessels and inhibited the vasoconstriction of the superior mesenteric artery in a dose-dependent but endothelial-independent *** effects were eliminated by preincubating vascular rings with the sGC inhibitors NS 2028 or ODQ,or with the PKG inhibitor KT ***,BXTY increased the protein expression of sGC-b1 and the intracellular second messenger cGMP level in mouse aortic vascular smooth muscle cells(MOVAs).NS 2028 or ODQ reversed these effects of *** expression level of the cGMP downstream effector protein PKG-1 increased after treating MOVAs with *** 2028,ODQ,or KT 5823 also reversed this effect of *** conclusion,BXTY can improve the symptoms of acute myocardial ischemia in mice,and activating the sGC-cGMP-PKG pathway in VSMCs to induce vasodilation is its key pharmacodynamic mechanism.

Circulating tumor cells(CTCs)have tremendous potential to indicate disease progression and monitor therapeutic response using minimally invasive *** the limitations of affinity strategies based on their cost,effectiveness,and simplicity,size-based enrichment methods that involve low-cost,label-free,and relatively simple protocols have been further ***,the key challenges of these methods are clogging issues and cell aggregation,which reduce the recovery rates and *** by the natural phenomenon that the airflow around a windmill is disturbed,in this study,a windmill-like hole array on the SU-8 membrane was designed to perturb the fluid such that cells in a fluid would be able to self-mix and that the pressure acting on cells or the membrane would be dispersed to allow a greater *** addition,based on the advantages of fluid coatings,a lipid coating was used to modify the membrane surface to prevent cell aggregation and clogging of the *** the optimal conditions,recovery rates of 93%and 90%were found for A549 and HeLa cells in a clinical simulation test of our platform with a CTC concentration of 20-100 cells per milliliter of *** white blood cell(WBC)depletion rate was 98.7%(n=15),and the CTC detection limit was less than 10 cells per milliliter of blood(n=6).Moreover,compared with conventional membrane filtration,the advantages of the proposed device for the rapid(2 mL/min)and efficient enrichment of CTCs without clogging were shown both experimentally and *** to its advantages in the efficient,rapid,uniform,and clog-free enrichment of CTCs,our platform offers great potential for metastatic detection and therapy analyses.

1例58岁女性乳腺癌患者行单侧乳腺改良根治术后给予环磷酰胺+盐酸多柔比星脂质体注射液方案化疗2次,化疗前均给予地塞米松注射液5 mg预处理,每次化疗后都出现不同程度的黏膜溃疡,双手、双足红肿,症状呈加重趋势,但几天后均自行好转。第3次化疗后患者双手、双足红肿伴水疱渗液,体温升高,最高达40.0℃,背部出现大片红斑,腋下红斑伴糜烂渗出,口腔黏膜溃疡。考虑患者手足综合征的可能性大,予甲泼尼龙琥珀酸钠、头孢西丁钠、奥美拉唑等药物治疗,呋喃西林溶液湿敷患处及保持皮肤湿润等外部皮肤护理措施。治疗15 d后患者手足症状好转。后期停用盐酸多柔比星脂质体注射液,改用多西他赛治疗,手足症状未再出现。

Searching for novel ferromagnetic oxides with high Curie temperature(TC)has been one of the main goals for oxide *** well-known perovskite cobaltate LaCoO_(3) is a classical ferromagnet in its thin-film form;however,it suffers from a low TC(~85 K).Here we report a new type of ferromagnetic La-Co-O films with an ultrahigh TC of~820 *** are fabricated by pulsed laser deposition from a LaCoO_(3) target at low oxygen partial *** structural analysis indicates that they crystallize in terms of the Ruddlesden–Popper phase of La_(2)CoO_(4±x).In sharp contrast to the antiferromagnetism of bulk La_(2)CoO_(4),the strong ferromagnetism in the La_(2)CoO_(4±x) thin films is firmly demonstrated by magnetometry measurements,X-ray magnetic circular dichroism characterization,and magnetotransport *** importantly,density functional theory calculations indicate that the nonstoichiometric oxygen induces an antiferromagnetic-to-ferromagnetic phase transition,accompanied by the orbital reconstruction of Co 3d ***,our study provides an attractive strategy for designing or synthesizing exotic magnetic oxides with high ordering temperatures.

Effect of diacylglycerol acyltransferase 2overexpression in 3T3-L1 is associated to an increase in mono-unsaturated fatty acid accumulation

Background：Fatty acid（FA） composition is the most important parameter affecting the flavor and nutritional value of the *** final and the only committed step in the biosynthesis of triglycerides is catalyzed by diacylglycerol acyltransferase 2（DGAT2）.The role of DGAT2 in lipid accumulation has been demonstrated in adipocytes,However,little is known about the effect of DGAT2 on the FA composition of these ***：To investigate the role of DGAT2 in regulating lipid accumulation,FA composition and the expression of adipogenic genes,we cloned the open reading frame of the porcine DGAT2 gene and established 3T3-L1 cells that overexpressed *** were then cultured in differentiation medium（DM） without FA,with a mixture of FAs（FA-DM）,or containing a ^（13）C stable isotope-labeled FA mixture（IFA-DM）.The FA composition of adipocytes was analyzed by gas chromatography-mass spectrometry and gas chromatography-isotope ratio mass *** PCR and western blotting were employed to detect expression of adipogenic genes in 3T3-L1 adipocytes cultured with FA-DM for 12 ***：The triacylglyceride（TAG） content was significantly higher in 3T3-L1 adipocytes overexpressing DGAT2 than in control *** cultured in DM or FA-DM for 12 d,cells overexpressing DGAT2 showed a higher proportion of unsaturated FAs（C16：1 and C18：1）.However,when cells overexpressing DGAT2 were cultured with FA-DM for30 min,the FA composition was almost identical to that of ***,the proportion of stable isotope-labeled FAs were similar in 3T3-L1 adipocytes overexpressing DGAT2 and control cells cultured in IFA-DM for 12 *** results collectively indicate that the higher proportion of mono-unsaturated FAs,C16：1 and C18：1,may originate from de novo FA synthesis but not from the uptake of specific FAs from the *** hypothesis is further supported by evidence that both mRNA and protein expression of genes involved in FA synthesis（ACACA,FASN,SCD1,and A-FABP）w

Evidence suggests associations between COVID-19 patients or vaccines and glycometabolic dysfunction and an even higher risk of the occurrence of ***,we retrospectively analyzed pancreatic lesions in autopsy tissues from 67 SARS-CoV-2 infected non-human primates(NHPs)models and 121 vaccinated and infected NHPs from 2020 to 2023 and COVID-19 ***-label immunofluorescence revealed direct infection of both exocrine and endocrine pancreatic cells by the virus in NHPs and *** and limited phenotypic and histopathological changes were observed in adult *** proteomics and metabolomics results indicated metabolic disorders,mainly enriched in insulin resistance pathways,in infected adult NHPs,along with elevated fasting C-peptide and C-peptide/glucose ratio ***,in elder COVID-19 NHPs,SARS-CoV-2 infection causes loss of beta(β)cells and lower expressed-insulin in situ characterized by islet amyloidosis and necrosis,activation ofα-SMA and aggravated fibrosis consisting of lower collagen in serum,an increase of pancreatic inflammation and stress markers,ICAM-1 and G3BP1,along with more severe glycometabolic *** contrast,vaccination maintained glucose homeostasis by activating insulin receptorαand insulin receptorβ.Overall,the cumulative risk of diabetes post-COVID-19 is closely tied to age,suggesting more attention should be paid to blood sugar management in elderly COVID-19 patients.

Somatic mutations of the chromatin remodeling gene ARID2 are observed in ～7% of human lung adenocarcinomas(LUADs). However, the role of ARID2 in the pathogenesis of LUADs remains largely unknown. Here we find that ARID2 expression is decreased during the malignant progression of both human and mice LUADs. Using two KrasG12 D-based genetically engineered murine models, we demonstrate that ARID2 knockout significantly promotes lung cancer malignant progression and shortens overall survival. Consistently, ARID2 knockdown significantly promotes cell proliferation in human and mice lung cancer cells. Through integrative analyses of Ch IP-Seq and RNA-Seq data, we find that Hspa1 a is up-regulated by Arid2 loss. Knockdown of Hspa1 a specifically inhibits malignant progression of Arid2-deficient but not Arid2-wt lung cancers in both cell lines as well as animal models. Treatment with an HSPA1 A inhibitor could significantly inhibit the malignant progression of lung cancer with ARID2 deficiency. Together, our findings establish ARID2 as an important tumor suppressor in LUADs with novel mechanistic insights, and further identify HSPA1 A as a potential therapeutic target in ARID2-deficient LUADs.

NYF型伟晶岩作为重要的战略性矿产资源,一直未受到广泛关注。相对于LCT型伟晶岩,NYF型伟晶岩的内部结构分带较差,极少存在区域性分带,但发育大量Nb、Y、F等矿物。铌铁矿族矿物、云母族矿物、电气石等矿物的地球化学特征能精细揭示NYF型伟晶岩的岩浆-热液演化过程,反映其演化程度。挥发分F、B、P和H_2O等不仅影响NYF型伟晶岩的结晶分异程度,对稀有稀土金属元素超常富集也起至关重要的作用。稀有稀土金属元素的富集、迁移与结晶贯穿整个NYF型伟晶岩的岩浆阶段、岩浆-热液过渡阶段以及热液阶段;高度演化的NYF型伟晶岩中,可以发生铌矿化、铍矿化、铷矿化、稀土(Y、Ce、Sc等)矿化以及锆-钍-铀矿化等。今后我国应将碱性岩-碱性花岗岩区域作为NYF型伟晶岩重点勘查区,加强NYF型伟晶岩的研究力度和指导找矿。
