Initial Approach to the Cellular Irradiation Injury of Human Pancreatic Carcinoma Cell line MIA PaCa-2 by High Dose per Fraction

OBJECTIVE To investigate an initial approach of radiotherapy, which produces cellular radiation injury by high dose in one fraction. METHODS Human pancreatic carcinoma cell line MIA PaCa-2, was cultivated and divided into 5 groups： 0, 2, 5, 10, 17 Gy. Cultivated cells were irradiated by 6MV-X ray in one fraction. Analysis were done as follows： comet assay, which assessed the level of DNA damage in the treated cells right after the cell was irradiated, flow cytometry, which was performed at 0, 6, 12, 24, 36 h after the cell line treated to ladder, which quantitatively asses changes of its cell cycle, DNA assessed the degree of DNA injury after 6 and 12 h, and histological examination, which analyzed cellular morphology after 24 h. RESULTS （1） After X-ray irradiated, the morphological change of human pancreatic carcinoma cell line （MIA PaCa-2） was mainly swelling. （2） When the dose of radiation was lower than 10 Gy, increasing the dose could greatly improve cell necrosis, apoptosis and blockage of cell cycle in GJM phase, which was consistent with the theory of radiation biology. （3） When radiation dose was more than the 10 Gy, the peak of apoptotic necrosis appeared strong and early. （4） The degree of DNA injury was also related to the dose of radiation therapy and most obvious in the 10 Gy group and not so obvious in the 17 Gy group. （5） When dose was less than 10 Gy, DNA ladder was a single electrophoretic band; in the 10 Gy group, the electrophoresis showed a multiple ladder band; when dose was more than 10 Gy, a vague and irregular band appeared on the electrophoresis. CONCLUSION Oncotic necrosis may be the main cell death style when dose per fraction is high, which differs from conventional dose fraction radiation therapy.

In this article, a novel strong interaction by forming complex between bovine serum albumin （BSA） and copper ion was utilized for the preparation of molecular imprinted bydrogel in aqueous solution. Results show that the inclusion of copper ion in preparation can bridge the template BSA and functional monomers together and improve the imprinting effect compared to the polymer made without copper ion added. High selectivity factor and large adsorption capacity are also observed for the obtained BSA-imprinted hydrogel.

Isothermal Crystallization of iPP in Environment-friendly Diluents:Effect of Binary Diluents and Crystallization Temperature on Crystallization Kinetics

The growing demand for non-toxic solvents for membrane preparation has motivated the studies for green and sustainable alternatives of solvents. The effect of droplet isothermal growth within the liquid-liquid phase separation region on isothermal spherulitic growth rate of isotactic polypropylene(iPP) was investigated. The results showed that the droplets grew up at a rate of 0.0172 μm·s^(-1). The larger droplets slowed down the isothermal spherulitic growth rate of iPP. Higher mass ratio of carnauba wax(Cwax)/soybean oil(SO)resulted in faster droplet growth due to weak interaction with polymers. The isothermal crystallization behaviors of iPP in environmentfriendly binary diluents consisting of Cwax and SO mixture were further investigated experimentally using polarized optical *** was demonstrated that the isothermal spherulitic growth rate of iPP in diluents decreased nonlinearly with the increasing crystallization temperature. Compared with virgin iPP, isothermal spherulitic growth rate of iPP in SO diluent was significantly slowed down. The spherulitic growth was further retarded after the addition of Cwax in mixed diluents, resulting in a lower crystallization rate than that in SO. Moreover, the crystal form of iPP membranes was found to be α type through the characterization of small angle X-ray scattering and wide angle X-ray diffraction.

The Influence of Different Single Radiation Dose on Delayed Growth of Transplanted Tumor in Athymic Mouse

OBJECTIVE To reveal the biological effects and effective dosage in radiotherapy model which applies high single-dose irradiation by animal experiment. METHODS We inoculated subcutaneouly human pancreatic carcinoma cell line （MIA PaCa-2） in the lateral of the right lower extremity of the athymic mouse to grow transplantation tumor. While the median diameter of transplantation tumor reached 10 mm approximately, the animals were randomly divided into 7 groups （6 animals per group） and irradiation by different dose in one fixed with consciousness for fraction （0, 2, 5, 10, 17, 25, 35 Gy）. All were kept on to be bred for observation of the change in gross tumor volume, calculation of delayed growth time and delayed growth curve. RESULTS With increased dose per fraction, cutaneous reaction on the neoplasma surface of the animal, which was mainly moist yellow effusion was more and more severe. When dosage is less than 10 Gy, all animals showed similar effects, that＇s the delayed tumor growth was not obvious. Tumors receiving more than 10 Gy in one fraction showed very good biological effect and the delayed tumor growth was obviously related to dosage. The difference in delayed tumor growth between the 2 groups was statistically significant. The delayed tumor growth time in 10, 17, 25 Gy group was respectively 3 weeks, 6 weeks and more. CONCLUSION The biological effect of the model which applies high single-dose irradiation （more than 10 Gy in one fraction） was very good. The effect of delayed tumor growth was obviously related to the dosage after transplantation tumor was radiated. Because of its higher dose per fraction and biological effects, the model of high single-dose irradiation can get better clinical effects.

Topography,structure,and formation kinetic mechanism of carbon deposited onto nickel in the temperature range from 400 to 850°C

The carbon deposition behavior on nickel particles was observed within the temperature range from 400 to 800°C in a pure methane atmosphere. The topography, properties, and molecular structure of the deposited carbon were investigated using field-emission scanning electron microscopy (FESEM), temperature-programmed oxidation (TPO) technology, X-ray diffraction (XRD), and Raman spectroscopy. The deposited carbon is present in the form of a film at 400-450°C, as fibers at 500-600°C, and as particles at 650-800°C. In addition, the structure of the deposited carbon becomes more ordered at higher temperatures because both the TPO peak temperature of deposited carbon and the Raman shift of the G band increase with the increase in experimental temperature, whereas the intensity ratio between the D bands and the G band decreases. An interesting observation is that the carbon deposition rate is suppressed in the medium-temperature range (M-T range) and the corresponding kinetic mechanism changes. Correspondingly, the FWHM of the G and D1 bands in the Raman spectrum reaches a maximum and the intensities of the D2, D3, and D4 bands decrease to low limits in the M-T range. These results indicate that carbon structure parameters exhibit two different tendencies with respect to varying temperature. Both of the two group parameters change dramatically as a peak function with increasing reaction temperature within the M-T range.

Expression of interleukin-22/STAT3 signaling pathway in ulcerative colitis and related carcinogenesis

AIM:To investigate the expression of interleukin (IL)-22 and its related proteins in biopsy specimens from patients with ulcerative colitis (UC) and UC-related carcinogenesis. METHODS:Biopsy specimens were obtained from patients with inactive (n = 10), mild-to-moderately active (n = 30), severely active (n = 34), initial (n = 30), and chronic UC (n = 44), as well as UC patients with dysplasia (n = 10). Specimens from patients without colonic abnormalities (n = 20) served as controls. Chronic colitis in experimental mice was induced by 2.5% dextran sodium sulfate. The expression levels of IL-22, IL-23, IL-22R1 and phosphorylated STAT3 (p- STAT3) were determined by immunohistochemistry. Bcl-2, cyclin D1 and survivin expression was detected by Western blotting. RESULTS:Patients with active UC had significantly more IL-22, IL-23, IL-22R1 and p-STAT3-positive cells than the patients with inactive UC and normal controls. Furthermore, IL-22 and related proteins were closely related to the severity of the colitis. The expression of IL-22 and IL-22R1 in the tissue of initial UC was stronger than in that of chronic UC, whereas the expression of p-STAT3 was significantly increased in chronic UC tissues. In dysplasia tissues, the expression level of IL-22 and related proteins was higher compared with controls. Mouse colitis model showed that expression of IL-22, IL-22R1 and IL-23 was increased with time, p-STAT3 and the downstream gene were also remarkably ***:IL-22/STAT3 signaling pathway may be related to UC and UC-induced carcinogenesis and IL-22 can be used as a biomarker in judging the severity of UC.

Static recrystallization(SRX)characteristics of a powder metallurgy superalloy were investigated by isothermal compression at 1080–1170℃ under strain rates of 0.01–0.1 s^(−1),strains of 0.1,0.22,or 0.5,and holding time of 0–300 *** impacts of temperature,strain rate,holding time,and strain on the SRXed grain size,volume fraction,and microtexture were explored by electron backscatter diffraction *** was found that temperature played a key role in these *** SRX progressed,thefiber parallel to the axis compression direction gradually weakened and was replaced by thefiber becausewas the preferred recrystallization orientation and grain growth direction for the Ni-based ***,high temperatures and low strain rates promoted the formation of the*** nucleation mechanisms during SRX process were found:grain boundary bulging,primary twin assistance,and subgrain *** boundary bulging occurred under all process conditions;however,at low temperatures and high strain rates,the latter two mechanisms could provide additional nucleation *** addition,SRX size and volume fraction models were established.

Vanadium disulfide(VS_(2)) as a typical two-dimensional transition metal chalcogenide has excellent competitiveness for sodium-ion storage due to its wide layer spacing(0.575 nm),high theoretical capacity of 932mAh·g^(-1) originating from multi-electron electrochemical ***,continuous sodiation process accompanied by crystal structural evolution and collapse cause rapid capacity ***,novel few-layer VS_(2)nanosheets with open(001) crystal planes are in-situ constructed on reduced graphene oxide to solve these issues mentioned *** indicates that few-layer VS_(2)provides more Na^(+) storage activity due to the low Na^(+)surface migration energy barrier on exposed crystal(001)*** flexible and high electronic conductivity of carbon matrix also effectively builds multi-level buffer structure and electron transport kinetics to boost the Na^(+)insertion/conversion reactive activity on VS_(2) as well as Na^(+) pseudocapacitance storage kinetics on edges and defects of nano *** coupling effects result in high rate capability and long cycling stability as a battery/capacitor *** delivers conspicuous high energy density of 81 and 40 Wh·kg^(-1) at power density of 118 and 10,286W·kg^(-1),as well as 80% energy retention rate after 5000cycles,confirming its great application potential in sodiumbased storage devices.

Effects of specific surface area of metallic nickel particles on carbon deposition kinetics

Carbon deposition on nickel powders in methane involves three stages in different reaction temperature ranges. Temperature programing oxidation test and Raman spectrum results indicated the formation of complex and ordered carbon structures at high deposition temperatures. The values of I（D）/I（G） of the deposited carbon reached 1.86, 1.30, and 1.22 in the first, second, and third stages, respectively. The structure of carbon in the second stage was similar to that in the third stage. Carbon deposited in the first stage rarely contained homogeneous pyrolytic deposit layers. A kinetic model was developed to analyze the carbon deposition behavior in the first stage. The rate-determining step of the first stage is supposed to be interfacial reaction. Based on the investigation of carbon deposition kinetics on nickel powders from different resources, carbon deposition rate is suggested to have a linear relation with the square of specific surface area of nickel particles.

Operating principles and detection characteristics of the Visible and Near-Infrared Imaging Spectrometer in the Chang'e-3

The Visible and Near-Infrared Imaging Spectrometer （VNIS）, using two acousto-optic tunable filters as dispersive components, consists of a VIS/NIR imag- ing spectrometer （0.45-0.95 μm）, a shortwave IR spectrometer （0.9-2.4 p.m） and a calibration unit with dust-proofing functionality. The VNIS was utilized to detect the spectrum of the lunar surface and achieve in-orbit calibration, which satisfied the re- quirements for scientific detection. Mounted at the front of the yutu rover, lunar ob- jects that are detected with the VNIS with a 45° visual angle to obtain spectra and ge- ometrical data in order to analyze the mineral composition of the lunar surface. After landing successfully on the Moon, the VNIS performed several explorations and cal- ibrations, and obtained several spectral images and spectral reflectance curves of the lunar soil in the region of Mare Imbrium. This paper describes the working principle and detection characteristics of the VNIS and provides a reference for data processing and scientific applications.
