Identification of molecular biomarkers for the diagnosis of gastric cancer and lymph-node metastasis

Background and objective:Biomarkers are important tools for prompt diagnosis of *** study aimed to identify reliable biomarkers for clinical applications in the diagnosis of gastric cancer and lymph-node(LN)***:Between 1 December 2014 and 31 December 2015,we prospectively collected samples of gastric-cancer tissues,corresponding matched-pair normal gastric mucosa,and their peri-gastric metastatic and non-metastatic LNs to identify quantitatively reliable genes using quantitative real-time polymerase chain *** quantity(RQ)was used to calculate the mRNA expression levels of our target *** were calculated using one-way analysis of variance(ANOVA)and Tukey’s multiple comparison *** graphs were plotted using GraphPad ***:Of nine assessed genes,the mRNA levels of inhibin beta A(INHBA)and secreted phosphoprotein 1(SPP1)were most consistently highly expressed in tumor tissues by 15.4-and 15.6-fold,respectively,as compared with normal tissues(P<0.001),with 91.3%sensitivity and 95.7%specificity(receiver operating characteristic[ROC]curve area=0.974)for the former and 82.6%sensitivity and 87.0%specificity(ROC curve area=0.924)for the *** analysis revealed no differentiating significance of SPP1 mRNA expression between metastatic and non-metastatic LNs(P=0.470).In contrast,the INHBA mRNA level was up-regulated 4.1-fold in metastatic LNs(P<0.001),with 80.0%sensitivity and 81.5%specificity(ROC curve area=0.857),and was also able to successfully differentiate between more severe disease conditions,T3 and T4(P=0.003),M0 and M1(P=0.043)and different histological variants(intestinal type vs diffuse type,P=0.019).Conclusions:Our results showed that INHBA was the most optimally reliable biomarker for diagnosing gastric cancer and LN metastasis.

Coefficients of earth pressure at rest in thick and deep soils

The effect of test methods and stress paths on the experimental value of the coefficient of earth pressure at rest, K0, was investigated under high pressures. The results indicate that the rigid pressure chamber and flexible lateral confining pressure medium method gives a stress ratio at the initial stage that is not the real K0. Moreover, K0 increases during the loading process becoming greater at high pressures. In the unloading process, however, K0 increases only at the initial stage but decreases thereafter. In addition, the incremental magnitude definition, K0=dσ3/dσ1, gives higher values than the total magnitude definition, K0=σ3/σ1, under loading. This is also true during initial stages of unloading. The experiment results also indicate that earth pressure at rest in deep and thick soils can be estimated by a power function of axial and confining pressures. It is necessary to choose the appropriate Kn to avoid some accidents.

Aiming to solve the bottleneck problem of electromagnetic scattering simulation in the scenes of extremely large-scale seas and ships,a high-frequency method by using graphics processing unit(GPU)parallel acceleration technique is *** the implementation of different electromagnetic methods of physical optics(PO),shooting and bouncing ray(SBR),and physical theory of diffraction(PTD),a parallel computing scheme based on the CPU-GPU parallel computing scheme is realized to balance computing ***,a multi-GPU framework is further proposed to solve the computational difficulty caused by the massive number of ray tubes in the ray tracing *** using the established simulation platform,signals of ships at different seas are simulated and their images are achieved as *** is shown that the higher sea states degrade the averaged peak signal-to-noise ratio(PSNR)of radar image.

There are complaints about current machine learning techniques such as the requirement of a huge amount of training data and proficient training skills,the difficulty of continual learning,the risk of catastrophic forgetting,and the leaking of data privacy/*** research efforts have been focusing on one of those concerned issues separately,paying less attention to the fact that most issues are entangled in *** prevailing big model paradigm,which has achieved impressive results in natural language processing and computer vision applications,has not yet addressed those issues,whereas becoming a serious source of carbon *** article offers an overview of the learnware paradigm,which attempts to enable users not to need to build machine learning models from scratch,with the hope of reusing small models to do things even beyond their original purposes,where the key ingredient is the specification which enables a trained model to be adequately identified to reuse according to the requirement of future users who know nothing about the model in advance.

Prognostic values of chromosome 18q microsatellite alterations in stage Ⅱ colonic carcinoma

AIM: To investigate the prognostic value of chromosome 18q microsatellite alterations (MA) in stage Ⅱ colon cancer. METHODS: One hundred and six patients with sporadic stage Ⅱ colon cancer were enrolled in this study. DNA was extracted from formalin-fixed, paraffin-embedded tumor and adjacent normal mucosal tissue samples. MA, including loss of heterozygosity (LOH) and microsatellite instability (MSI), was analyzed by polymerase chain reaction, polyacrylamide gel-electrophoresis and DNA sequencing at 5 microsatellite loci on chromosome 18q (D18S474, D18S55, D18S58, D18S61 and D18S64).RESULTS: Among the 102 patients eligible for MA information, the overall frequencies of LOH, high and low frequency MSI/microsatellite stable were 49.0%, 17.6% and 82.4%, respectively. The high frequency of 18q-LOH was signif icantly associated with the poor 5-year overall survival (OS) (P=0.008) and disease free survival (P=0.006). High levels of MSI were significantly associated with a longer 5-year OS (P=0.045) while the higher frequency of 18q-LOH at the loci of D18S474 and D18S61 was significantly associated with a poorer 5-year OS (P=0.010 and 0.005, respectively). But multivariate analysis showed that only the frequency of 18q-LOH was significantly associated with the prognosis of the disease. CONCLUSION: High frequency of 18q-LOH is an independent prognostic factor indicating poor prognosis of the patients with stage Ⅱ colon cancer.

Comparison of transthoracic and transabdominal surgical approaches for the treatment of adenocarcinoma of the cardia

Background and Objective: Transthoracic and transabdominal approaches are commonly used for the surgical treatment of adenocarcinoma of the cardia. Which approach is better has been controversial for quite a long time. Our study aimed to compare the surgical trauma, range of lymph node dissection, and the prognosis of the transthoracic and transabdominal approaches for the treatment of adenocarcinoma of the cardia. Methods: The medical records of 331 patients with adenocarcinoma of the cardia treated in our hospital between 1994 and 2003 were analyzed. Of the 331 patients, 284 underwent operation via transthoracic approach and 47 via transabdominal approach. Surgery-related status, postoperative complications, range of removed lymph nodes and prognosis of the two groups were compared. Results: There was no significant difference in surgery-related status and postoperative complications between the two groups (P >0.05). The mean number of removed lymph nodes from the thoracic cavity was much higher in transthoracic group than in transabdominal group (P < 0.001), while that from the abdominal cavity was similar in both groups (P = 0.404). The thoracic lymph node metastasis rate was 18.8% in transthoracic group and 13.3% in transabdominal group. The median survival time was 29 months in transthoracic group and 28 months in transabdominal group, and the 5-year survival rates were 34.9% and 40.1% (P= 0.599). Conclusions: For the surgical treatment of adenocarcinoma of the cardia, the surgical trauma of the transthoracic approach is similar with that of transabdominal approach. The transthoracic approach has the advantage in thoracic lymph node dissection. The two approaches have no obvious effect on the prognosis.

A function-based behavioral modeling method for air combat simulation

Today’s air combat has reached a high level of uncertainty where continuous or discrete variables with crisp values cannot be properly represented using fuzzy sets. With a set of membership functions, fuzzy logic is well-suited to tackle such complex states and actions. However, it is not necessary to fuzzify the variables that have definite discrete ***, the aim of this study is to improve the level of model abstraction by proposing multiple levels of cascaded hierarchical structures from the perspective of function, namely, the functional decision tree. This method is developed to represent behavioral modeling of air combat systems, and its metamodel,execution mechanism, and code generation can provide a sound basis for function-based behavioral modeling. As a proof of concept, an air combat simulation is developed to validate this method and the results show that the fighter Alpha built using the proposed framework provides better performance than that using default scripts.
