近年来,苹果生产成本不断攀升已成为影响果园效益和果农收入的重要因素,很大程度上制约了苹果产业高质量发展。延安是我国优势苹果产业带核心区域,苹果产业作为延安农业的支柱产业,对当地经济社会高质量发展,特别是广大农民脱贫致富贡献巨大。开展了果园投入对果园效益和果农收入影响的调查,通过分析延安苹果生产投入现状、投入与产出矛盾,提出有效对策措施,为助力延安苹果高质量发展先行区建设提供参考。

The regulation of apple(Malus domestica)fruit texture during ripening is complex and a fundamental determinant of its commercial *** climacteric fruit,ripening-related processes are regulated by ethylene(ET),and jasmonate(JA)is also involved in the ethylene biosynthesis pathway,mainly through the transcription factor ***,the molecular genetic mechanism for fruit ripening processes between the JA and ET signaling pathways still needs to be *** order to explore how JA regulates apple fruit ripening through ERF4,we used’Gala’and’Ralls Janet’fruit at different developmental stages as experimental materials to determine the fruit firmness and related gene expression ***,we carried out different hormone treatments on’Gala’fruit at ripening ***,we show that ERF4 is a core JA signaling hub protein JASMONATE ZIM-DOMAIN(JAZ)interactor that affects ethylene signaling *** fruit development,ERF4 represses the expression of ACS1 and ACO1 by interacting with JAZ,as well as with the JA-activated transcription factor *** is promoted in JAZ-suppressed ***,ERF4 acts as a molecular link between ethylene and JA hormone signals,and the natural variation of the ERF4Ethylene-responsive binding factor-associated amphiphilic repression(EAR)motif decreases repression of ethylene biosynthesis genes.

Niche differentiation shapes the bacterial diversity and composition of apple

Microorganisms play an important role in plant growth,health and ecological *** is necessary to explore the microbial community because of the significant ecological and economic value of apples(Malus x domestica).However,the related bacterial communities in different ecological habitats are not clear in the unique scion/rootstock combinations of *** this study,the Illumina Miseq platform was used to analyze the bacterial communities in different niches(bulk soil,rhizosphere soil and roots)of two scion/rootstock combinations[Fuji/Malus xiaojinensis Cheng et Jiang(Mx)and Fuji/Malus baccata(L.)Borkh.(Mb)].The relationship between the microbial community,soil properties,and plant characters was *** results showed that the diversity of the bacterial community decreased from bulk soil to rhizosphere soil and then to *** addition,at the phylum level,Proteobacteria,Actinobacteria and Acitinobacteria were dominant and had notable divergence in different ecological *** niches had remarkable effects on the diversity and composition of the bacterial community according to *** rootstock genotype had an effect on the bacterial composition but no effect on the ***,soil organic matter(SOM),alkaline hydrolyzable nitrogen(AN)were the main factor affecting the structure of bacterial community in rhizosphere soil,as shown by Pearson *** taxa in the roots were affected by starch,fructose,sucrose and *** study showed the microbial community structure in different ecological niches of different scion/rootstock combinations and provided some evidence for understanding the microbial-plant interaction of apple rootstocks.

Cytokinins are members of a group of phytohormones involved in various growth and developmental processes in *** transferase(IPT)is the rate-limiting enzyme in catalyzing the biosynthesis of *** this study,to understand the role of IPT family in cold resistance,78 IPT candidates were identified and characterized in nine Rosaceae *** expansion of IPT families in the Rosaceae primarily occurred through segmental duplication rather than tandem *** general,purifying selection controlled the evolution of IPT families in the Rosaceae,with IPT3 and IPT5 homologs as the primary drivers of ***-elements,which are involved in the responses to many environmental stresses or phytohormone signals,were identified in the promoters of MdIPT *** was consistent with the trends of expression of the MdIPT genes in apple(Malus domestica)***5b was also found to exhibit multiple responses to phytohormones and stress *** ectopic expression of MdIPT5b resulted in an increase in cold resistance in transformed apple calli and tomato(Solanum lycopersicum)*** redox balance was partially stabilized through the accumulation of proline under cold ***,the ascorbate–glutathione cycle cannot be stabilized in the *** physiological and biochemical assays are preformed in *** results showed that regulating the expression of IPT genes for moderate cytokinin improvement could enhance the accumulation of proline to stabilize the osmotic and redox balances to improve resistance to cold stress.

ALKBH5 is a master regulator of N6-methyladenosine(m6A)modification,which plays a crucial role in many biological ***,we show that ALKBH5 is required for breast tumor ***,PRMT6 directly methylates ALKBH5 at R283,which subsequently promotes breast tumor ***,arginine methylation of ALKBH5 by PRMT6 increases LDHA RNA stability via m6A demethylation,leading to increased aerobic ***,PRMT6-mediated ALKBH5 arginine methylation is confirmed in PRMT6-knockout ***,these findings identify a PRMT6-ALKBH5-LDHA signaling axis as a novel target for the treatment of breast cancer.

This study focuses on the suburbanization and subcentering of population and examines the nature of spatial restructuring in terms of the population distribution in the Beijing metropolitan *** of the classic density function approach,we employ the nonparametric analysis to characterize the spatial pattern of population densities in the Beijing metropolitan area and identify the suburban *** findings suggest that the population has spread with rapid urban growth in the Beijing metropolitan area,and the compact urban form has been replaced by a more dispersed polycentric spatial ***,compared with the decentralization of western cities,the spatial extent of the decentralization of population in the Beijing metropolitan area is quite *** rapid growth of population in the near suburbs has expedited the sprawl of the central city,with a larger central agglomeration of population dominating the metropolitan *** this sense,the spatial pattern of the Beijing metropolitan area is still characterized by the continuous ***,our findings do provide the evidence that the city has been turning to a polycentric *** find significant population subcenters have emerged in the suburbs of Beijing since the *** the polycentricity emerged in the Beijing metropolitan area is very different by nature from that observed in Western *** subcenters emerged are adherent to the development scheme planned for the city,so it can be referred to as the so called 'planned polycentricity'.

Apple rootstock breeding has achieved great progress *** this review,we first summarize the rootstock breeding targets and utilization in main apple-producing ***,we discuss the focus and important research areas of apple rootstock breeding through five aspects:parent selection and setting of crosses,target genes and marker-assisted breeding,root configuration-guided breeding,apomictic resource utilization,and the application of genetic ***,we propose an apple rootstock division plan for China,which has a large potential to provide guidance for apple rootstock breeding and utilization in different apple-producing areas of China.

目的:分析慢性心力衰竭病人血清微小核糖核酸-487b(miR-487b)表达水平及其与心肌重构的关系。方法:选取2019年10月—2021年10月西安高新医院收治的96例慢性心力衰竭病人作为研究组,按照纽约心脏病协会心功能分级将病人分为Ⅰ级或Ⅱ级22例,Ⅲ级41例,Ⅳ级33例;另选取同期在同一所医院体检的健康者102名作为对照组。采用实时荧光定量聚合酶链式反应(qRT-PCR)法检测所有研究对象血清miR-487b表达水平。比较研究组与对照组血清miR-487b表达水平及研究组不同心功能分级病人血清miR-487b表达水平。采用Pearson法分析慢性心力衰竭病人血清miR-487b表达水平与心肌重构指标[包括左室重构指数(LVRI)、左室舒张末期内径(LVEDD)、左心房内径(LAD)、左室质量指数(LVMI)、左室后壁厚度(LVPW)]的关系。结果:研究组血清miR-487b表达水平低于对照组(P<0.001);研究组LVEDD、LAD、LVMI、LVPW均高于对照组(P<0.001),LVRI低于对照组(P<0.001)。研究组心功能Ⅳ级病人LVEDD、LAD、LVMI、LVPW均高于Ⅰ级或Ⅱ级、Ⅲ级病人(P<0.05),miR-487b表达水平、LVRI均低于Ⅰ级或Ⅱ级、Ⅲ级病人(P<0.05);Ⅲ级病人LVEDD、LAD、LVMI、LVPW均高于Ⅰ级或Ⅱ级病人(P<0.05),miR-487b表达水平、LVRI均低于Ⅰ级或Ⅱ级病人(P<0.05)。Pearson相关分析结果显示,慢性心力衰竭病人血清miR-487b与LAD、LVMI、LVPW均呈负相关(r值分别为-0.578、-0.502、-0.514,P<0.001),与LVRI呈正相关(r=0.464,P<0.001)。结论:慢性心力衰竭病人血清miR-487b表达水平降低,且与心肌重构密切相关。

[目的]探讨银杏素调节核因子E2相关因子2(Nrf2)/溶质载体家族7成员11(SLC7A11)/谷胱甘肽过氧化物酶4(GPX4)信号通路对氧化型低密度脂蛋白(ox-LDL)诱导的血管内皮细胞铁死亡的影响。[方法]将人脐静脉内皮细胞***926分为正常对照组(正常培养)、ox-LDL组(50 mg/L ox-LDL)、银杏素低剂量组(50 mg/L ox-LDL+10μmol/L银杏素)、银杏素中剂量组(50 mg/L ox-LDL+20μmol/L银杏素)、银杏素高剂量组(50 mg/L ox-LDL+40μmol/L银杏素)、ML385组(50 mg/L ox-LDL+40μmol/L银杏素+1μmol/L的Nrf2抑制剂ML385)、Erastin组(50 mg/L ox-LDL+40μmol/L银杏素+5μmol/L的SLC7A11抑制剂Erastin)、RSL3组(50 mg/L ox-LDL+40μmol/L银杏素+0.5μmol/L的GPX4抑制剂RSL3);MTT法检测细胞存活率;试剂盒检测细胞超氧化物歧化酶(SOD)、丙二醛(MDA)、还原型谷胱甘肽(GSH)水平;特异性荧光探针法检测细胞内铁含量;DCFH-DA荧光探针法和氟硼二吡咯(BODIPYTM)法检测细胞内活性氧(ROS)和脂质ROS水平;Western blot检测细胞Nrf2、SLC7A11、GPX4、4-羟基壬烯酸(4-HNE)、环氧合酶2(COX2)、p53蛋白表达。[结果]与正常对照组相比,ox-LDL组细胞存活率、SOD含量、GSH含量以及Nrf2、SLC7A11、GPX4表达明显降低(P<0.05),MDA含量、细胞内Fe2+含量、细胞内ROS、脂质ROS以及4-HNE、COX2、p53表达显著升高(P<0.05)。与ox-LDL组相比,银杏素低、中、高剂量组细胞存活率、SOD含量、GSH含量及Nrf2、SLC7A11、GPX4表达明显升高(P<0.05),MDA含量、Fe2+含量、细胞内ROS、脂质ROS及4-HNE、COX2、p53表达显著降低(P<0.05)。与银杏素高剂量组相比,ML385组、Erastin组、RSL3组均减弱了银杏素对ox-LDL诱导的血管内皮细胞铁死亡的抑制作用。[结论]银杏素通过激活Nrf2/SLC7A11/GPX4通路抑制ox-LDL诱导的血管内皮细胞铁死亡。
